Relative Efficiencies of Live, Dry and Mixed Food Diets for Larval Rearing of Tigris Scraper (Capoeta umbla) and the Influence of Stocking Density on Fry Growth Characteristics

Gün, Abdulselam; Kızak, Volkan; Gündüz, Fatih

In this research, larval growth features by applying different diets and growth characteristics of fry at different stocking densities were studied under controlled conditions for the Tigris scraper (Capoeta umbla). In the first experiment, the growth and survival rates of Tigris scraper postlarvae were evaluated under three different diet regimes over 21 days. The highest mean total length was 24.04±0.27 mm in the group L1 fed solely Artemia nauplii, and the lowest mean total length was 17.89±0.16 mm in the group L2 fed only dry feed. The highest mean specific growth rate was in the L1, and the lowest mean specific growth rate was in the L2. In the second experiment in which the growth characteristics of fry were investigated at different stocking densities for 98 days, the highest mean live weight was found in S1 with 2.819±0.157 g and the lowest mean live weight was determined as 0.859±0.073 g in S5. The highest mean specific growth rate was in S1, while the lowest rate was in S5. High stocking densities were found to be disadvantageous for growth of C. umbla fry. The findings suggest that successful rearing of Tigris scraper postlarvae and fry is feasible under controlled conditions.

Evaluation of a new computer method (ANNs) and traditional methods (LWRS and VBGF) in the calculation of some growth parameters of two cyprinid species

    Ozcan, Ebru Ifakat; Serdar, Osman

Artificial Neural Networks (ANNs) are a very reliable computer program used in many areas such as growth parameters. The aim of this study to compare the growth estimates of tigris scraper (Capoeta umbla (Heckel, 1843)) and kura barbel (Barbus lacerta (Heckel, 1843) captured from Karasu River (East Anatolia, Turkey) between 2014 and 2016. The age groups were 0-9 for C. umbla and 0-8 for B. lacerta. The estimated parameters of von Bertalanffy growth equations were as L' =51.96 cm, W' 804.16 g, k=0.092 year-1, t0=-1.763 year for C. umbla and L' 32.23 FP, W' 435.87 J, k 0.127\HDU-1, t0-2.489 for B. lacerta. The growth performance index (Y) value was comput-ed as 2.395 for C. umbla and 2.120 for B. lacerta. The length and weight relationships for two fish species were as follows: W=0.0092L3.05 (R²=0.98) and W=0.0095L3.07 (R2=0.98). The growth of these two cyprinid species were isometric growth. A ANNs model is a interdepented system containing many neurons. There are three different layers in a neural network (input, hidden and output layers). Data enters (length/weight, age, sex) at the inputs and passes along the network, by layer, till it reach-es at the outputs (weight/length). MATLAB Ver R2016a (Neural Network Toolbox) was used for ANNs estimatings. MAPE (%) equation was used to compare ANNs and other methods. According to the comparison of the results obtained with MAPE (%), ANNs provide better results than the LWRs and VBGF. The results of this study could give useful insight that ANNs can be evaluated as an alternative for growth prediction.

Effect of L-Tryptophan on Sperm Quality of Tigris Scraper (Capoeta umbla) (Pisces: Cyprinidae) after Cryopreservation

    Kutluyer, F.; Aksu, Ö.; Kocabaş, M.

BACKGROUND: The aromatic amino acid L-tryptophan is a versatile molecule and needed for the biosynthesis of proteins. Due to metabolic functions, it has been widely used in research and clinical trials. OBJECTIVE: The present study aimed to test the usefulness of L-tryptophan as an extender of Tigris scraper (Capoeta umbla) sperm during cryopreservation. MATERIALS AND METHODS: Semen samples were diluted with a ratio of 1:3 (sperm: extender) and base extenders were supplemented with L-tryptophan (0 mM, 0.5 mM, 1 mM, 2 mM and 4 mM). Sperm motility and duration of post-thawed samples were assessed after the cryopreservation process. RESULTS: Our results revealed that motility rate and duration of sperm increased when the cryomedia was supplemented with L-tryptophan. On the other hand, an increase in the concentration of L-tryptophan in the extender caused a significant decrease (p<0.05) in the motility rate of Tigris scraper (C. umbla) sperm. The ideal results were obtained at 1 mM L-tryptophan; however, no motile sperm were shown in any samples when a concentration of 2 mM was used. CONCLUSION: The current study shows the importance of conducting further studies related to long-term storage and reproduction management.

C-REACTIVE PROTEIN AND ADRENOMEDULLIN LEVELS IN CAPOETA UMBLA (HECKEL, 1843) AS NOVEL BIOINDICATORS FOR BIOMONITORING THE MICROBIAL CONTAMINATION IN INLAND WATERS OF TUNCELI, TURKEY

Yildirim, Nuran Cikcikoglu; Danabas, Durali; Yildirim, Numan; Tanyol, Mehtap; Danabas, Seval

The present study focused on changes in the C-reactive protein (CRP) and adrenomedullin (ADM) levels in the liver and gill tissues of Capoeta umbla as biomarkers of environmental pollution and ecosystem health in inland waters of Tunceli (Turkey). The CRP levels in liver and gills were not significantly changed in March and September at every site (p>0.05) except for site 3 in gill tissues (p<0.05). The results of statistical analysis indicate that significant differences in CRP levels were found in liver and gills among sites (p<0.05), but no changes in the gills at March (p>0.05). In March, ADM levels in gills and liver were statistically changed among sites (p<0.05), while no statistical changes were determined in September (p>0.05). The results of this study indicated that CRP levels in the tissues of the C. umbla could be utilized as a significant bioindicator for disease pathogenesis in fish infections.

Danabas, Durali; Yildirim, Nuran Cikcikoglu; Yildirim, Numan; Onal, Ayten Oztufekci; Uslu, Gulsad; Unlu, Erhan; Danabas, Seval; Ergin, Cemil; Tayhan, Nilgun

The aim of this work was to determine the seasonal changes in the activities of selected biomarkers in Capoeta umbla (Heckel, 1843) caught from Uzuncayir Dam Lake (Tunceli, Turkey) and to evaluate the effects of environmental factors on these activities. Fish were sampled on seasonal basis, and superoxide dismutase, catalase, glutathione peroxidase activities and levels of glutathione and malondialdehyde in gills were determined. Significant variations of oxidative stress biomarkers were observed between seasons and sites. The results of this study show that seasonal variations of oxidative stress responses and lipid peroxidation in gills of C. umbla are sensitive to the contaminants present in water of Uzuncayir Dam Lake and selected parameters are in valuable biomarkers for monitoring of water systems, since they give an early warning signal of effects of xenobiotics on aquatic organisms at molecular levels which help to prevent their effects at organismal level. Use of acetylcholinesterase, glutathione S-transferase and cytochrome P450 1A1 in Capoeta umbla as biomarkers for monitoring of pollution in Uzuncayir Dam Lake (Tunceli, Turkey)

Yildirim, Nuran Cikcikogiu; Yildirim, Numan; Danabas, Durali; Danabas, Seual

Uzuncayir Dam Lake is one of the most important water bodies in Tunceli. It is polluted by domestic wastewaters together with natural contamination and pesticides, which have had cumulatively negative effects. This study analyzes the effects of pollution by using the fish species, Capoeta umbla as a sentinel species, with a biomarker approach. The approach comprehends a general biomarker of individual fish health, the condition factor and specific biomarkers of contaminant exposure such as glutathione S-transferase (GST) and cytochrome P450 1A1 (CYP1A1) and acetylcholinesterase (AChE) activity. The general water physico-chemical parameters were measured at each sampling sites during each fish-sampling period. Strong seasonal and locational variations were observed among selected markers. In conclusion, the multibiomarker approach used in the present study clearly revealed differences in the fish health among reference and potentially contaminated sites. The study shows that changes of biomarkers such as acetylcholinesterase, glutathione S-transferase and cytochrome P450 1A1 might be useful for the assessment of environmental contamination in the Uzuncayir Dam Lake (Tunceli, Turkey). Assessment of immunomodulator biomarkers (Tnf-α, Il-1β and Il-6) in liver of Capoeta umbla for biomonitoring of pollution in Uzuncayir Dam Lake (Tunceli, Turkey)

    Yildirim, N. C.; Danabas, S.

This study was aimed to monitor water pullution of Uzuncayir Dam Lake using the changes of IL-6, IL1 beta and TNF-alpha levels in Capoeta umbla (Heckel, 1843) liver tissue at ten stations in March and September 2011. In this study C. umbla (Heckel, 1843) was used as the indicator organism. Tumor necrosis factor-alpha (TNF-alpha), interlekuin1 beta (IL-1 beta) and interleukin-6 (IL-6) levels were determined in samples of the liver tissue by ELISA kit. The lowest mean IL-1 beta levels were found at station 6. The mean IL-1 beta was reached its maximum level at station 2. The difference between the mean levels of IL-6 was found to be significant (p<0.05) amoung stations. The IL-6 levels were significantly increased in September at stations 1, 2, 7 and 8 (p<0.01) compared to the values in March. The mean levels of TNF-alpha were found to be significant (p<0.05) amoung stations. The TNF-alpha levels significantly decreased in September at stations 1 and 9 (p<0.01). TNF-alpha, IL-1 beta and IL-6 levels in C. umbla can be used as early diagnostic indicators against adverse environmental events and useful and reliable bioindicators in determining the pollution of the aquatic ecosystem.

ESTIMATION OF THE POPULATION SIZE OF CAPOETA UMBLA (HECKEL, 1843) IN THE LAKE HAZAR (ELAZIG) BY REMOVAL METHOD

    Yüksel, F.; Alp, A.; Gündüz, F.; Çoban, M. Zulfu; Demirol, F.

Fish populations in smaller aquatic environments are affected by the fishing pressure at a much faster pace. Often, compensation of this effect either takes a long time or is not possible at all. For the purposes of establishing fishing regulations in these water environments, the removal method is able to provide results quickly and almost accurately. The Lake Hazar, which is the environment of our study, is 8,600 hectares in size, and is a natural lake where Capoeta umbla species is caught commercially using gill nets. In this study, stock size of Capoeta umbla in the Lake Hazar (Elazig) was estimated using removal method. In addition, the power of fishing and the status of fishing yield in the lake were evaluated. In our research, it was determined that seven commercial fishing boats caught 25,721 kg of fish in the lake in a fishing season. CPUE was calculated as 1.55 kg (kg/day/a gill net). As a result of the regression analysis, the amount of stock in the lake was the estimated to be 37,781 kg (+/- 14,001 kg). When cumulative catch was formulated as recommended by Ricker (1975), the population size was estimated to be 38,649 kg (+/- 15,007 kg). As a result of our study, the use of removal method was found to be appropriate for small reservoirs with reduced population density due to strong fishing pressure.

Some Heavy Metals Accumulation in Tissues in Capoeta umbla (Heckel, 1843) from Uzuncayir Dam Lake (Tunceli, Turkey)

Ural, Mesut; Yildirim, Numan; Danabas, Durali; Kaplan, Olcay; Yildirim, Nuran Cikcikoglu; Ozcelik, Mehtap; Kurekci, Eylem Funda

Concentrations of metals were determined in the gills, liver, kidney, heart and muscle in Capoeta umbla caught from six stations from the Munzur River system. Metal concentrations in the tissues tended to vary significantly among stations (p < 0.05). Liver (Cu, 10.10 +/- A 0.23-23.03 +/- A 9.37 ppm; Zn, 14.67 +/- A 3.01-21.82 +/- A 2.39 ppm; Cd, 18.04 +/- A 4.56-52.69 +/- A 10.65 ppb and Fe, 28.87 +/- A 6.78-115.11 +/- A 34.87 ppm) and kidney (Cu, 1.80 +/- A 0.25-3.70 +/- A 0.62 ppm; Zn, 20.81 +/- A 0.37-29.36 +/- A 0.70 ppm; Cd, 132.06 +/- A 5.29-639.51 +/- A 20.14 ppb and Fe, 24.40 +/- A 1.98-59.39 +/- A 1.97 ppm) tissues showed higher metal concentrations than other tissues. It seems that metal contamination in the river is too high for the health of fish and the people who eat them. The geographical locations of catch, season, nature of diet, and the size of fish used for analyses might lead to different metal concentration in the same fish species.
