Water quality changes and goldfish growth (carassius auratus) in microgreen aquaponic and recirculating systems

    Kizak, Volkan; Kapaligoz, Serdar

Goldfish (Carassius auratus) growth was studied comparatively in microgreen aquaponic (APS) and sump filter systems (SFS), and the ef fects of these two different filter systems on water quality parameters were investigated. Arugula (Eruca vesicaria) was used in APS and produced as microgreens. NH3-N, NO2-N, NO3-N, POj and NH4 were measured at specific periods for 60 days. No statistical differences were found between both systems in terms of food conversion ratio (FCR), specific growth rate (SGR), survival and mean body weight of goldfish. In the beginning of the experi-ment, water pH level was around 8.5 while trend towards declining at the end of period. Nitrification process was successful in both systems, and declin ing pH towards from basic medium to approaching neutral levels made positive contribution to perfor mance of nitrifying bacteria. Although there was a significant decrease in the level of NH4, the level of NO3-N increased continously after the 30,h day in APS. According to these results, arugula micro-greens used mostly NH4 instead of NO3-N in higher pH level. Goldfish-arugula microgreen aquaponic system is similar with SFS in terms of keeping water quality in optimal conditions and any adverse effect on goldfish growth and welfare was not ob served in both systems. Although in high pH, it was also seen that arugula seedlings can be produced efficiently as microgreens.

CRYOPRESERVATION OF GOLDFISH (Carassius auratus) SPERMATOZOA: EFFECTS OF EXTENDER SUPPLEMENTED WITH TAURINE ON SPERM MOTILITY AND DNA DAMAGE

Kutluyer, Filiz; Ögretmen, Fatih; Inanan, Burak Evren

BACKGROUND: Amino acids, present in seminal plasma at high concentration, protect spermatozoa against cell damage during cryopreservation. OBJECTIVE: Experiments were designed to analyze the effect of semen extender supplemented with taurine on post-thawed sperm motility and duration, as well as DNA damage. MATERIALS AND METHODS: Extenders were supplemented with 1, 2 or 4 mM taurine. Semen samples were diluted at the ratio of 1:9 with the extenders. Diluted samples were aspirated into 0.25 ml French straws and 0.1 ml pellets. DNA damage was assessed with the comet assay after cryopreservation. RESULTS: The percentage and duration of sperm motility were significantly increased by taurine. Additionally, sperm motility and the motility period in pellets were higher than in straws. The best concentration of taurine was 4 mM, and the highest post-thaw motility rate (72.50 +/- 3.54%) and duration (17.50 +/- 0.71 s) were obtained from the extender with 4 mM in pellets. DNA damage was decreased after taurine supplementation. CONCLUSION: Pellets could be used for goldfish sperm cryopreservation. The addition of 4 mM taurine increases the post-thaw motility and decreases DNA damage on goldfish semen.

EFFECTS OF SEMEN EXTENDER SUPPLEMENTED WITH L-METHIONINE AND PACKAGING METHODS (STRAWS AND PELLETS) ON POST-THAW GOLDFISH (Carassius auratus) SPERM QUALITY AND DNA DAMAGE

Kutluyer, Filiz; Ögretmen, Fatih; Inanan, Burak Evren

BACKGROUND: Amino acids protect spermatozoa against cell damage during cryopreservation due to have antioxidant property and found in seminal plasma at high concentration. OBJECTIVE: The aim of the present work was to analyse the effect of extender supplementation with L-methionine on post-thawed sperm motility, duration and DNA damage and also it was tested the feasibility of using straws and pellets method for the cryopreservation of goldfish (Carassius auratus) sperm. MATERIALS AND METHODS: Extenders were supplemented with different L-methionine concentrations of 1 mM; 1.5 mM; 3 mM; 6 mM. Semen samples diluted at the ratio of 1:9 by the extenders were subjected to cryopreservation. After dilution the semen was aspirated into 0.25 ml straws and 0.1 ml pellets, the straws and pellets were placed on the tray, frozen in nitrogen vapor and plunged into liquid nitrogen. DNA damage was determined with comet assay after cryopreservation.. RESULTS: Our results indicated that an increase in the concentration of L-methionine caused a significant increase in the motility rate and duration of sperm in goldfish (C. auratus) (p<0.05). In addition, duration and percentage of motility in pellets were higher than in straws. Comparing all concentrations of L-methionine, the best concentration of L-methionine was 1.5 mM. Highest post-thaw motility (45.00 +/- 7.07%) and duration of motility (17.00 +/- 0.71s) were obtained with the extender at concentration 1.5 mM in pellets. Addition of the extender with L-methionine was reduced DNA damage compared to control group. CONCLUSION: Consequently, pellets could be use for goldfish sperm cryopreservation and the tested amino acid affected the motility parameters, and semen extenders could be supplement with L-methionine.

Accumulation and Toxicity of CuO and ZnO Nanoparticles Through Waterborne and Dietary Exposure of Goldfish (Carassius auratus)

Ates, Mehmet; Arslan, Zikri; Demir, Veysel; Daniels, James; Farah, Ibrahim O.

Dietary and waterborne exposure to copper oxide (CuO) and zinc oxide (ZnO) nanoparticles (NPs) was conducted using a simplified model of an aquatic food chain consisting of zooplankton (Artemia salina) and goldfish (Carassius auratus) to determine bioaccumulation, toxic effects, and particle transport through trophic levels. Artemia contaminated with NPs were used as food in dietary exposure. Fish were exposed to suspensions of the NPs in waterborne exposure. ICP-MS analysis showed that accumulation primarily occurred in the intestine, followed by the gills and liver. Dietary uptake was lower, but was found to be a potential pathway for transport of NPs to higher organisms. Waterborne exposure resulted in about a 10-fold higher accumulation in the intestine. The heart, brain, and muscle tissue had no significant Cu or Zn. However, concentrations in muscle increased with NP concentration, which was ascribed to bioaccumulation of Cu and Zn released from NPs. Free Cu concentration in the medium was always higher than that of Zn, indicating CuO NPs dissolved more readily. ZnO NPs were relatively benign, even in waterborne exposure (p0.05). In contrast, CuO NPs were toxic. Malondialdehyde levels in the liver and gills increased substantially (p<0.05). Despite lower Cu accumulation, the liver exhibited significant oxidative stress, which could be from chronic exposure to Cu ions. (c) 2014 Wiley Periodicals, Inc. Environ Toxicol 30: 119-128, 2015.
