Glyphosate disrupts sperm quality and induced DNA damage of rainbow trout (Oncorhynchus mykiss) sperm

Akça, Ayşe; Kocabaş, Mehmet; Kutluyer, Filiz

As a widespread pollutant, glyphosate (GLY) adversely affects the aquatic environment and can impair the reproductive ability and functions of fish. The purpose of the current study was to assess in vitro effect of GLY on rainbow trout (Oncorhynchus mykiss) sperm cells. The sperm cells were exposed to different GLY concentrations (2.5, 5, 10 mg/L). Sperm motility parameters were analyzed with computer assisted sperm analysis. DNA fragmentation (%) was measured by the comet assay using fluorescence microscopy. With increased GLY concentration, sperm motility and duration decreased after exposure. DNA fragmentation (% DNA in tail) in sperm cells was higher in treatments containing GLY than control (p < 0.05). Consequently, sperm cells are sensitive to low doses of GLY, and this can negatively affect natural populations.

THE EFFECT OF PENDIMETHALIN HERBICIDE ON ANTIOXIDANT SYSTEM IN SAFFLOWER

    Beker-Akbulut, Gulcin

Pendimethaline is a dinitroaniline herbicide compound which selectively controls weeds. It is a cell division and growth inhibitor. In this study, pendimethalin herbicide was applied to Dincer and Remzibey cultivars of Carthamus tinctorius L. in 0.004-0.01 M concentration range after germination. The effects of pendimethalin upon peroxidase (POD), ascorbate peroxidase (APX), superoxide dismutase (SOD), catalase (CAT) reduced glutathione (GSH), glutathione reductase (GR), glutathione-Stransferase (GST) and total phenolic content were investigated on the 24, 48 and 72 hours following the treatment. POD, APX, SOD, CAT and GST activities were increased on the 24, 48 and 72 hours in Dincer while decreased at 72 hour in Remzibey. GSH and GR activity were decreased at 72 hour in Dincer and Remzibey. Total phenolic content decreased at 48 and 72 hours in Dincer and Remzibey. In this study, we determined that pendimethalin herbicide affected the antioxidant enzyme activities in Dincer and Remzibey.

PERSISTENCE OF TWO SULPHONYLUREA HERBICIDES IN WHEAT PLANTED FIELD, THRACE REGION IN TURKEY

    Erguven, G. O.; Yildirim, N.

This study is concerning the environmental fate of chlorsulfuron and tribenuron-methyl herbicide in a wheat planted field at Thrace Region in Turkey. Use of herbicides containing these active ingredients is advised in the Marmara region in Turkey for cereal agriculture. The study was conducted in wheat planted fields where these herbicides were used. Over nine months, in each two or three week periods, soil samples were obtained from selected sampling points and herbicide residuals were determined by gas chromatography with electron capture detector (GC-ECD) and high performance liquid chromatography-mass spectrometry (HPLC-MS). As a result of these studies, persistence of chlorsulfuron was determined as approximately 1.2% at the second half of June, 2016 and tribenuron methyl seemed to be approximately 1% at the end of May, 2016. The results of the present study showed that chlorsulfuron is generally adsorbed weakly by soil when compared to tribenuron-methyl. pH, organic material, climatic factors and clay content of soil were the main factors influencing the adsorption and desorption properties of the herbicides. The results of this study demonstrated that chlorsulfuron and tribenuron-methyl residues in the soil might depend on their different chemical structures, soil temperature, precipitation and humidity.

ROLE OF SOME ISOLATED SOIL CULTURES ON REDUCTION OF HERBICIDE TRIFLURALIN

    Erguven, Gokhan Onder; Bayhan, Hurrem

The study is about the removal of herbicide Trifluralin as COD parameter and monitoring the population dynamic of isolated cultures. These cultures were isolated from soil samples taken from these areas for their identifications before the use of herbicide. Removal of Trifluralin was determined by microbiological degradation with these produced bacteria and fungi in liquid media. With these growing bacteria and fungi, removal rate by bioremediation method studies were carried out. In addition to biodegradation mechanism, with the measurements of the turbidity; the colony number of microorganisms is monitored with isolated bacteria and fungi species. The removal efficiency for Bacillus simplex was 95% and Clostridium tetani species was 86%, by the fungi Penicillium simplicissimum, and Metacordcyceps chlamydosporia species, were 59% and 80% respectively in five days. Bioremediation study results have shown differences with respect to species in liquid media. Degradation time of these herbicides in liquid culture media with isolated cultures varied between 5 min. and 135 min.
