In this study, ameliorative effects of ellagic acid (EA) on oxidative stress induced by chlorpyrifos (CPF) in carp, Cyprinus carpio, were investigated. Fish were divided into six groups: C (no treatment), EA (100 mg kg fish−1), CPF-1 (0.040 mg L−1), CPF-2 (0.080 mg L−1), CPF-1 + EA, and CPF-2 + EA. CPF and EA were applied simultaneously for 14 days and, at the end of the study, liver, kidney, and gill samples were collected from fish. On the taken tissue samples, malondialdehyde (MDA) level and some antioxidant enzyme (superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), and glutathione-S-transferase (GST)) activities were evaluated. The results demonstrated statistically significant increases in the MDA levels of the CPF-1 and CPF-2 groups. On the other hand, the MDA levels were significantly decreased by EA administration. Also, CPF exposure caused statistically significant increases in the SOD and GST activities and statistically significant decreases in the CAT and GPx activities. However, treatment with EA reversed negative alterations in the SOD, CAT, GPx, and GST activities. Therefore, the results of this study results showed that simultaneous treatment with EA alleviates CPF-induced oxidative stress in fish.

- MakaleYalnız künye2021Scopus SJRQ1Evaluation of the efficiency of chlorpyrifos-ethyl remediation by Methylobacterium radiotolerans and Microbacterium arthrosphaerae using response of some biochemical biomarkers

This study reveals out detoxifying and antioxidant enzyme response of Gammarus pulex exposed/polluted to chlorpyrifos-ethyl insecticide before and after biodegradation/bioremediation by Methylobacterium radiotolerans and Microbacterium arthrosphaerae. Cytochrome P450 1A1, glutathione S-transferase, catalase, and superoxide dismutase activities in G. pulex exposed to chlorpyrifos-ethyl before and after bioremediation/biodegradation by these two bacteria during 24 and 96 h tested by using commercial ELISA kits. The activity of catalase enzyme was decreased depending on chlorpyrifos-ethyl before and after bioremediation/biodegradation the enzyme activity was increased repeatedly. Superoxide dismutase activity level increased after chlorpyrifos-ethyl exposure in 96 h (p > 0.05). Following bioremediation, superoxide dismutase enzyme activity decreased again during 24 h (p > 0.05) and increased during 96 h (p < 0.05). Statistical differences were not found in cytochrome P450 1A1 enzyme activity before and after the process (p > 0.05). No significant differences were determined during the activity of glutathione S-transferase in 24 h (p > 0.05). The activities of glutathione S-transferase were increased after exposure of chlorpyrifos-ethyl during 96 h. After bioremediation; the activity of glutathione S-transferase increased even more (p < 0.05). The results determined that activities of G. pulex at superoxide dismutase, catalase, and glutathione S-transferase are common biomarkers for revealing out the efficiency of bioremediation of chlorpyrifos-ethyl with these two types of soil bacteria. - MakaleYalnız künye2019Scopus SJRQ2The usability of oxidative stress and detoxification biomarkers in Gammarus pulex for ecological risk assessment of textile dye methyl orange
Cikcikoglu Yildirim, Nuran; Yaman, Metin
The present study was undertaken to determine the toxicity of the methyl orange by using the changes of some antioxidant and detoxification enzyme activities in Gammarus pulex. Lethal Concentration (LC) value of Methyl Orange (MO) was determined. Three sublethal doses of MO (1/4; 1/8 and 1/16 of LC value) were exposed to G. pulex for 24 and 96 h. Superoxide dismutase (SOD), Catalase (CAT), Glutathione peroxidase (GSH-P X ), Cytochrome p450 (CYP1A1), Glutathione S-transferase (GST) activities as well as Glutathione (GSH) and Malondialdehyde (MDA) levels were determined by using The enzyme-linked immunosorbent assay (ELISA) kit. The CAT and CYP1A1 activities were decreased in all the groups exposed to different doses of MO. GST activity and GSH, MDA levels were increased all the groups exposed to different doses of MO. The GSH-P X activities were changed in all the groups. MO affected SOD activity at different levels and in different concentrations. In our study, it has been found that exposure duration didn’t significantly affect the biochemical biomarkers except for GST and GSH. In conclusion, alterations in antioxidant and detoxification enzymes and lipid peroxidation may potentially be used as sensitive biomarkers for risk assessment of dyes in the environment and may contribute to the establishment of discharge regulations.
