The current study focused on the ameliorating impact of TRP (5 mM) on Salmo abanticus spermatozoa exposed to sub‑lethal concentrations of GLY (2.5, 5, and 10 mg/L). Sperm quality parameters, oxidative stress indices, and DNA damage were analyzed in the post‑exposure sperm. Our data reveal that GLY has an adverse effect on motility rate and duration, sperm kinetics parameters, antioxidant status and DNA damage, and triggers antioxidant defense mechanisms at the highest dose (10 mg/L) and it has insufficient therapeutic effect. Overall, GLY poses potential hazards to male reproductive cells and may adversely affect subsequent generations of natural populations.

Sahilli, Yeliz Çakir; Korkmaz, Volkan
Abstract: Glyphosate (GLY) is one of the most widely used herbicides in modern intensive agriculture to enhance crop production. However, the chemical additives in commercial formulations and the active ingredient itself can adversely affect biological and chemical processes in aquatic organisms. Therefore, the aim of this study was to evaluate the effects of sub-lethal concentrations of GLY (0, 0.23, 0.58, and 1.17 μg/L) on oxidative stress biomarkers—namely lipid peroxidation (measured as malondialdehyde, MDA), and antioxidant enzymes (superoxide dismutase, SOD; catalase, CAT; and reduced glutathione, GSH)—in Daphnia magna after 24 and 96 h of exposure. The results showed that SOD activity increased in all treatment groups compared to the control at both time points while CAT activity decreased in all groups. GSH and TBARS (thiobarbituric acid reactive substances) levels were significantly elevated after 24 and 96 h in all exposed groups. Overall, D. magna was sensitive to sub-lethal concentrations of GLY, and changes in SOD and CAT activities, as well as GSH and TBARS levels, these biomarkers can serve as useful indicators for detecting glyphosate-induced oxidative stress and water pollution.

- Yüksek Lisans TeziAçık Erişim2023Toprak ortamında herbisit biyoremediasyon veriminin Gammarus pulex'in bazı biyokimyasal paramatreler kullanarak araştırılması
- MakaleYalnız künye2021Scopus SJRQ3Glyphosate disrupts sperm quality and induced DNA damage of rainbow trout (Oncorhynchus mykiss) sperm
Akça, Ayşe; Kocabaş, Mehmet; Kutluyer, Filiz
As a widespread pollutant, glyphosate (GLY) adversely affects the aquatic environment and can impair the reproductive ability and functions of fish. The purpose of the current study was to assess in vitro effect of GLY on rainbow trout (Oncorhynchus mykiss) sperm cells. The sperm cells were exposed to different GLY concentrations (2.5, 5, 10 mg/L). Sperm motility parameters were analyzed with computer assisted sperm analysis. DNA fragmentation (%) was measured by the comet assay using fluorescence microscopy. With increased GLY concentration, sperm motility and duration decreased after exposure. DNA fragmentation (% DNA in tail) in sperm cells was higher in treatments containing GLY than control (p < 0.05). Consequently, sperm cells are sensitive to low doses of GLY, and this can negatively affect natural populations.

- MakaleYalnız künye2019Scopus SJRQ2The effect of a glyphosate-based herbicide on acetylcholinesterase (AChE) activity, oxidative stress, and antioxidant status in freshwater amphipod: Gammarus pulex (Crustacean)
Pala, Ayşegül
This study had determined the effect of glyphosate-based herbicide (GBH) on acetylcholinesterase (AChE) enzyme activity, oxidative stress, and antioxidant status in Gammarus pulex. Firstly, the 96-h LC50 value of glyphosate on G. pulex was determined and calculated as 403 μg/L. Subsequently, the organisms were exposed to sub-lethal concentrations (10, 20, and 40 μg/L) of the determined GHB for 24 and 96 h. The samples were taken from control and GBH-treated groups at 24 and 96 h of study and analysed to determine the malondialdehyde (MDA) and reduced glutathione (GSH) levels, the AChE, superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) enzyme activities. In the G. pulex exposed to GBH for 24 and 96 h, the MDA level increased significantly (p < 0.05). The GSH level, the AChE, the CAT, and the GPx activities decreased compared with the control group (p < 0.05). G. pulex exposure to GBH for 24 h showed a temporary reduction in the SOD. GBH exposure led to oxidative stress in the G. pulex as well as affected the cholinergic system of the organism. These results indicated that the parameters measured may be important indicators of herbicide contamination in G. pulex.

- MakaleAçık Erişim2018Scopus SJRQ4Effect of glyphosate on some protective systems in Zea mays L; [Glifosatın mısır (Zea mays L.)’da bazı koruyucu sistemlere etkisi]
Beker Akbulut, Gulcin; Yigit, Emel; Bayram, Dilek
Glyphosate is an herbicide that is applied after non-selective germination and affects plant growth. In this study, glyphosate was applied to Zea mays L. after germination, at a concentration range from 0.017 to 0.145 M in a growth chamber. The effects of this herbicide on some antioxidant enzymes, lipid peroxidation, total chlorophyll and total carbohydrate content were investigated on days 1, 5 and 10 following the application. Results showed that peroxidase (POD), ascorbate peroxidase (APX), superoxide dismutase (SOD), catalase (CAT) and glutathione-S-transferase (GST) were increased in the groups treated with glyphosate on days 5 and 10, but the activities of reduced glutathione (GSH) and glutathione reductase (GR) were decreased on day 10. Malondialdehyde (MDA) content indicating lipid peroxidation was increased on days 1, 5 and 10. Changes in total chlorophyll and total carbohydrate contents were found to be time-dependent. These increases and decreases in the antioxidant activities, MDA content, total chlorophyll and total carbohydrate content in Zea mays L. were determined as a symptom of cytotoxicity caused due to glyphosate.
