Aim: In this study, the phytochemical composition of the seeds of twelve different local grape (Vitis vinifera L.) varieties traditionally cultivated in the Southeastern Anatolia Region (Diyarbakır, Türkiye) was investigated using a holistic approach. The main objective of the study was to evaluate the associations of the effects of berry skin colour, the ripening stage and utilization type on the fatty acid profile, tocopherol and phytosterol contents, vitamin K and D levels, antioxidant capacity (DPPH) and lipid peroxidation (MDA) of grape seeds. Method: The seeds of grape varieties belonging to four different colour groups, grown on their own roots, were harvested during the 2017 growing season and analysed. Fatty acid composition was determined by gas chromatography (GC), vitamin and phytosterol contents by HPLC-UV, MDA content by the thiobarbituric acid reactive substance (TBARS) method, and antioxidant capacity by the DPPH radical scavenging assay. Findings: Linoleic acid was identified as the predominant fatty acid in all varieties (53.68–67.06%), followed by oleic acid (13.80–27.32%). α-Tocopherol was identified as the predominant tocopherol form (1.597–4.703 mg kg−1), whilst β-sitosterol was the phytosterol component present at the highest level (915.67–3340.42 µg g−1). Statistically significant varietal differences were detected for most measured phytochemical parameters. Group-wise differences associated with berry skin colour and ripening stage were observed for selected traits. Overall, varietal differences appeared to be more pronounced than those associated with berry skin colour or ripening stage. It was observed that the differences in phytochemical composition between the various commercial classifications of grapes were very limited. Result: The findings reveal that the seeds of the local grape varieties studied exhibit a rich phytochemical profile and represent an important source of bioactive compounds that could be utilised, particularly in the functional food, nutraceutical and cosmetics industries. The ‘Tahannebi’ variety stood out as the one with the highest values across many parameters. Differences observed among samples of the same cultivar collected from different vineyards suggest that vineyard-specific conditions may contribute to variation in selected phytochemical traits. Future multi-year and multi-location studies are needed to confirm the stability of these phytochemical profiles and to further clarify genotype × environment interactions.

Growing tomatoes in season and organically are an important factor in terms of carrying higher levels of antioxidant molecules. This study aimed to demonstrate the protective effect of extracts obtained from naturally grown tomatoes, which are widely used as food, against oxidative damage caused by hydrogen peroxide on yeast cells (Saccharomyces cerevisiae L.). Lipophilic vitamins, phytosterol content, gluathione and oxidised gluathione content were analyzed by Shimadzu brand high performance liquid chromatography, while fatty acid analysis was performed by Shimadzu GC 2010 Plus Gas Chromatography. Protein content was analyzed spectrophotometrically. The study groups were designed as control, hydrogen peroxide, tomato and tomato + hydrogen peroxide. In all groups added with hydrogen peroxide and tomato extract compared to the hydrogen peroxide group; it was observed that vitamin D2, α-Tocopherol, β-Sitosterol and Stigmasterol contents increased significantly. Again, it was observed that ergosterol level increased significantly in the D + hydrogen peroxide group compared to the hydrogen peroxide group. When the fatty acid analysis results were examined, it was determined that in all groups added to the cultured yeast with hydrogen peroxide and tomato extracts (except the B + hydrogen peroxide group), there were increases in 16:0, 16:1, n-7, 17:0, 17:1 and 18:0 fatty acid levels compared to the hydrogen peroxide group. On the other hand, when its effect on protein and gluathione levels was examined, more significant increases were determined in all groups with tomato extract addition compared to the hydrogen peroxide group. At the end of the study, it was observed that tomato extracts had positive effects against hydrogen peroxide toxicity in yeast cells, although at different levels in terms of fatty acid, vitamin and phytosterol contents.
- MakaleAçık Erişim2026Scopus SJRQ4Protective effects of Sodium Tetraborate on fatty acid profiles in rat hearts subject to Lead toxicity; [Efecto protector del tetraborato de sodio sobre los perfiles de ácidos grasos en el corazon de ratas, expuestas a toxicidad por plomo]
Aydin, Sevinç
This study was designed to observe the effects of sodium tetraborate on preventing lead metal toxicity damage to fatty acids in rat heart tissue. After the heart tissues were homogenized with Tris-HCl buffer, the separated pellet part was prepared for fatty acid analysis. The fatty acids in the obtained lipid extract were converted to methyl ester form and analyzed with SHIMADZU GC 2010 PLUS gas chromatography. When comparing the lead group with the lead + sodium tetraborate group, partial increases were observed in saturated fatty acid levels such as myristic acid (14:0), pentadecanoic acid (15:0), and palmitic acid (16:0), while varying increases were seen in some monounsaturated fatty acid levels such as palmitoleic acid (16:1, n-7), and oleic acid (18:1, n-9). Moreover, demonstrated varying levels of increases in all polyunsaturated fatty acids excluding the arachidonic acid (20:4, n-6) and adrenic acid (22:4, n-6) fatty acids. As the most notable fatty acid change in our study, arachidonic acid (20:4, n-6) fatty acid increasing in the lead group while decreasing in the sodium tetraborate group demonstrates that sodium tetraborate decreased the negative effects of oxidative stress on fatty acids. The results of this study provide supporting evidence for the potential use of sodium tetraborate in drug design to prevent the toxic effects of lead.

13 species of Gundelia species plants are endemic in T & uuml;rkiye and are frequently used by the public for both nutrition and medicinal objectives. In this study, three parts (flower, leaf, and stem) of three species of Gundelia ( G.dersim , G.glabra, and G.munzurensis) were biochemically investigated. DPPH and ABTS analyses for antioxidant activity were performed on methanol extracts of plant parts. As a result of both analyses in parallel, the flower part of G.munzurensis had the highest %Inhibition value (DPPH:91.85 +/- 0.78; ABTS:97.38 +/- 0) (P<0.001). In total phenolic (29.09 mg GAE/g) and total flavonoid (5.59 mg CE/g) measurements, the highest amount was found statistically quite significant (P<0.001) in the leaf part of G.munzurensis. Gallic, vanillic, ferulic, romantic, and cinnamic acids, which are phenolic acids and of the flavonoids, rutin, catechin, naringin, and naringenin, were determined in all plant parts. Vitamin D3, alpha-tocopherol, ergosterol, stigmasterol, and beta-sterol were detected in parts of all plant species. While palmitic (16:0), palmitoleic (16:1), stearic (18:0), oleic (18:1), linoleic (18:2), and linolenic (18:3) fatty acids as the main fatty acids were detected in all parts of the three Gundelia species, lauric (12:0), tridecanoic (13:0), myristic (14:0), pentadecanoic (15:0), cis-10-pentadecanoic (15:1), heptadecanoic (17:0), cis-10heptadecanoic (17:1), and eicosanoid (21:0) acids were also identified at different concentrations. Considering the biochemical properties of Gundelia species, it shows that they can be consumed as food and also used pharmacologically.

İncele - MakaleAçık Erişim2024Scopus SJRQ3Lipid content and wastewater treatment potential of Chlorella vulgaris and Scenedesmus obliquus isolated from Uzuncaylr Dam Lake
Can, Şafak Seyhaneyildiz; Can, Erkan; Yilmaz, Kadir
Production of microalgae for biodiesel, one of the alternative renewable energy sources, is costly due to nutrient media. Industrial and domestic wastewater contains nitrogen, phosphorus and other nutrients that are the primary food source for algae. Releasing this environmentally harmful effluent into receiving bodies of water, such as the sea or a freshwater reservoir, without prior treatment causes serious problems. Combining microalgae cultivation with wastewater treatment is a promising strategy for improving wastewater and reducing the cost of nutrient media required for algae production. In this study, wastewater obtained from a wastewater treatment facility was diluted with clean water to 0.25%, 50%, and 75% concentrations, and Chlorella vulgaris and Scenedesmus obliquus were cultured in these nutrient media for 20 days. As a result, microalgae increased their biomass and lipid content, while consuming nitrite, nitrate, phosphate, and ammonium from the wastewater as nutrients.

- MakaleYalnız künye2021ANTIOXIDANT ACTIVITIES AND SOME BIOCHEMICAL CONTENTS OF THE SEEDS OF THE OKUZGOZU AND BOGAZKERE (Vitis vinifera L.) GRAPE VARIETIES GROWN IN TURKEY
Aydin, Sevinc
The Vitis vinifera L. grape species is a fruit that is the most prevalently grown in the whole world and has economic significance. It is known that, due to its climate conditions, Turkey has rich resources in terms of Vitis vinifera L. With this study, we aimed to determine the phenolic, vitamin and fatty acid contents of the Okuzgozu and Bogazkere varieties of Vitis vinifera L. and see the effects of these contents on antioxidant activity. Fatty acids in the lipid extract were analyzed by gas chromatography after conversion into methyl esters. For flavonoid and vitamin analyses, a high-performance liquid chromatography device (HPLC-UV. Shimadzu) was used. Antioxidant activity was determined by Radical Scavenging Activity (DPPH) and Lipid Peroxidation (MDA) experiments, As a result of the study, some saturated and unsaturated fatty acid, phenolic compound and vitamin contents of the two varieties were obtained. It was observed that the amounts of unsaturated fatty acids, whose effects on antioxidant activity are known, in the Okuzgozu variety were significantly higher than those in the Bogazkere variety. The results on phenolic and vitamin compounds revealed higher amounts in the Okuzgozu variety in comparison to Bogazkere as in the case of fatty acids. The finding that these contents and antioxidant activity were in proportion suggested that the antioxidant effect of the Okuzgozu variety is higher. If the substances that provide this effect are correctly isolated from the plant, they may be used beneficially in both the food and pharmaceutical sectors.

Akbaba, Emel; Aydin, Sevinc; Sanyurek, Nesrin Karaca
Vitis vinifera L. is cultivated throughout the World including Turkey which is one of the largest producers of grape. Grape is consumed as fresh or dry fruit as well as used in the wine industry in which good quality of grapes are preferred. Grape is also a part of the traditional Mediterranean diet which is linked to a low risk of cardiovascular and degenerative diseases. Kohnu is a geographically labeled type of grape cultivated in the eastern cities of Turkey. In this study, Kohnu grapes harvested from two different cities of Turkey were compared through phenolic compounds, phytosterols, vitamins, fatty acids, and antioxidative properties. The concentrations of phenolic compounds in the sample from Elazig were obtained to be significantly higher as compared to the sample from Malatya. Especially vanillic acid was found to be much higher in Elazig samples. The concentrations of fatty acids especially palmitic acid, palmitoleic acid, oleic acid, and linoleic acid were obtained significantly higher in Elazig samples as compared to Malatya samples. Vitamin and phytosterol concentrations of the samples from Elazig were also determined as higher as compared to Malatya samples. Significantly higher antioxidant activity of Kohnu grape seeds from Elazig was obtained via DPPH radical scavenging activity and MDA levels. The antioxidant profile of Kohnu grape seeds of Elazig samples could be related to phenolic composition and vitamins.

Kuzgun, N.K.
In this study, fish chips were produced using Luciobarbus esocinus, their fatty acids and amino acids profiles were determined and they were sensorially evaluated. For the preparation of experimental samples, L. esocinus meat was minced and added to a basic chips mixture. After that, the mixture was divided into in two: while pepper sauce was added to one half, the other half had no sauce added. The mixtures with sauce and without sauce were shaped into chips. Two samples of each batch of the chips prepared were cooked using two different methods (baking and frying). In this way four different groups were obtained. Analysis of fatty acids showed that the pre-frying and the pre-baking process resulted in a decrease in all fatty acids except for 18:1n-9 and 18:2n-6. Analysis of amino acids showed that isoleucine, lysine, threonine and, proline were present in the highest quantities respectively. After frying and baking the amounts of leucine, isoleucine, lysine, arginine, phenylalanine, and threonine decreased while the amounts of the valine and histidine remained at the same level. The essential amino acids for humans were found in the fish meat and in the fish chip groups, except for tryptophan. The general acceptability score of the fish chips was highest for group A which consisted of chips with sauce that had been fried (9.28±0.69).

- MakaleYalnız künye2011Scopus SJRQ3FATTY ACIDS PROFILES OF RAINBOW TROUT (ONCORHYNCHUS MYKISS WALBAUM, 1792), FED WITH ZEOLITE (CLINOPTILOLITE)
Danabas, D.
In this trial, the lipid contents and fatty acid compositions (% of total fatty acids) of rainbow trout fed with four different ratios of clinoptilolite were studied. The fatty acid compositions of fish in groups ranged from 26.81% to 27.93 % saturated fatty acids, 25.35-31.435 % monounsaturated and 32.99-40.185 % polyunsaturated. Among them, those occurring in the highest proportions were oleic acid (C18:1n9, 19.85-22.27 %), palmitic acid (C16:0, 15.60-16.56 %), linoleic acid (C18:2n6, 11.43-18.88 %), cis-4,7,10,13,16,19-docosahexaenoic acid (C22:6n3, 13.36-15.52 %), stearic acid (C18:0, 4.25-4.75 %), palmitoleic acid (C16:1, 3.53-4.59 %), cis-5,8,11,14,17-eicosapentaenoic acid (C20:5 n3, 3.11-3.39 %), and myristic acid (C14:0, 2.56-2.85 %). The findings demonstrated that fatty acid compositions of the groups depend on feed, age, environmental conditions, and effects of feed additives like clinoptilolite. In this respect, it demonstrated that clinoptilolite can be added to fish feeds
