Experiments were designed to test the influence of L-tryptophan supplementation of the extender on the sperm motility parameters and bacterial flora of brook trout (Salvelinus fontinalis) and rainbow trout (Oncorhynchus mykiss). The extender containing 0.3 M glucose in 10% Dimethyl sulfoxide (DMSO) was supplemented with 0, 0.5, 1, 2, and 4 mM L-tryptophan. Sperm samples stored for up to 12 days at 4°C for brook trout were motile while motility was not observed after six days of storage for rainbow trout. Diluted sperm samples were spread-plated onto Plate Count Agar (PCA) (Total Bacteria Count), Rose Bengal Agar (RBC) (Yeast-Mold Count), Violet Red Bile Dextrose (VRBD) (Enterobacteriaceae count) and Mannitol Salt Agar (MSA) (Micrococcus/Staphylococcus count) and counts were performed in duplicate and sampling occurred on day 0, 2, 4, 6, and 12 of cold storage. L-tryptophan supplementation of the extender yielded a positive effect, significantly prolonging sperm motility in S. fontinalis and O. mykiss compared to the control group (p < 0.05). The ideal results were obtained above a concentration of 0.5 mM for both species. Total bacteria count in fresh sperm (undiluted samples) was not detectable and not detected in sperm samples treated with different L-tryptophan concentrations. Yeast-mold, Enterobacteriaceae and Micrococcus/Staphylococcus were not detected from fresh and treated sperm brook trout and rainbow trout. On the basis of the results, L-tryptophan-based extender is useful for maintaining sperm quality in short-term storage. L-tryptophan had a negative effect on the bacterial flora. The results of the current study encourages further studies related to long-term storage and reproduction management. Abbreviations: DMSO: dimethyl sulfoxide; PCA: plate count agar; RBC: rose Bengal agar; VRBD: violet red bile dextrose; MSA: mannitol salt agar; SCA: sperm class analyzer; CFU: colony-forming unit.

- MakaleAçık Erişim2019Scopus SJRQ2l-Arginine Improves Sperm Motility in Threatened Coruh Trout Salmo coruhensis (Teleostei: Salmonidae)

l-arginine is a versatile amino acid and precursor of nitric oxide that provides protection for preventing free radical damage. Herein, experiments were designed to clarify the effect of l-arginine supplementation on the sperm motility of a threatened trout, Coruh trout Salmo coruhensis. Activation solution was supplemented with levels of 0 mM (Control), 2 mM, 4 mM, and 8 mM l-arginine, and motility and survival of sperm cells were assessed. The selected concentrations were determined according to results of our preliminary experiments. The sperm motility rates at l-arginine concentrations of 0, 2, 4, and 8 mM were 73.33 ± 5.77%, 76.67 ± 5.75%, 80.01 ± 0.01%, and 90.00 ± 0.05%, respectively; notility durations were 21.67 ± 2.08, 32.33 ± 3.79, 32.67 ± 1.15, and 37.00 ± 6.38 s, respectively. Significant effect of l-arginine supplementation was determined on the percentage and duration of motile spermatozoa (p < 0.05). The maximum increment was evoked at the greatest concentration (8 mM). Here, we showed that l-arginine supplement can improve sperm motility of S. coruhensis.

- MakaleAçık Erişim2018Scopus SJRQ3Determination of Lipid Peroxidation and Fatty Acid Compositions in the Caviars of Rainbow Trout and Carp

In this study, roes obtained from carp (Cyprinus carpio carpio) that were caught in Keban Dam Lake (Turkey) and cultured rainbow trout (Oncorhynchus mykiss) were processed as caviar, and the products were stored at + 4 C°. Lipid peroxidation and the compositions of fatty acids of these products were assessed during production and storage period. The result of fatty analyses showed that palmitic acid (16:0), stearic acid (18:0), palmitoleic acid (16:1), oleic acid (18:1), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), which are all essential fatty acids, were determined in the caviars of the roes obtained from rainbow trout and carp. According to results of chemical analyses, there were no significant differences in thiobarbituric acid reactive substances (TBARs) between rainbow trout and carp caviars (p > 0.05), but malondialdehyde (MDA) levels were found to be statistically significant (p < 0.01). It can be concluded that caviars obtained from both rainbow trout and carp roes are rich in fatty acids and take a part in a balanced and healthy diet.

- MakaleAçık Erişim2017Stability of fish protein hydrolysate from heads of gilthead sea bream (Sparus aurata), european sea bass (Dicentrarchus labrax) and rainbow trout (Oncorhyncus mykiss) during storage
Erol, Nida Demirtas; Erdem, Omer Alper; Çakli, Sukran
In the present study, protein hydrolyzate was obtained from the head region from the wastes produced during filleting of seabream (Sparus aurata), seabass (Dicentrarchus labrax) and trout (Oncorhyncus mykiss) cultured in Turkey. After the functional and antioxidant properties of the obtained products were determined, the post-storage stability of the protein hydrolysates stored frozen for 6 months was determined by analysis. In this study, degree of hydrolysis, functional properties (protein solubility, emulsion properties, foaming properties, color properties, water holding capacity (WHC)), antioxidant properties (DPPH (1,1 -diphenyl-2-pic-ryhydrayl) free radical scavenging capacity, antioxidative activity, metal chelating) and determination of aminoacid sequencing were carried out in protein hydrolysate. To evaluate the effect of storage time on fish protein hydrolysates stored at -18 degrees C after production, the above-mentioned analyzes including functional and antioxidative properties were repeated at the end of the 6 month storage period. Results were interpreted using statistical analysis. According to the functional and antioxidative properties of fish protein hydrolyzate, the highest antioxidant activity was determined in seabream head hydrolyzate. The production yield of all fish protein hydrolysates is 5%. The functional and antioxidant properties of fish protein hydrolysates varied according to fish species.
