Okan K., Z. Özkara, E. Sevindik, M. Sevindik, M. Y. Paksoy (2024). Molecular markers analysis of endemic Bornmuellera hausskn. spp. (Brassıcaceae) ın Türkiye. - Genetika, Vol 56, No.2, 295-304. In this study, molecular characterisation of Türkiye's endemic species Bornmuellera cappadocica (Willd.) Cullen & T.R. Dudley, Bornmuellera glabrescens (Boiss. & Balansa) Cullen & T.R.Dudley, Bornmuellera kiyakii Aytaç & Aksoy and Bornmuellera angustifolia (Hausskn. ex Bornm.) Cullen & T.R.Dudley was carried out using ten RAPD and ten ISSR primers. In RAPD-PCR analysis, 66 bands were obtained and the polymorphism rate was 96.96%. In the ISSR-PCR analysis, 119 bands were obtained and the polymorphism rate was 95.79%. In the UPGMA (Unweighted Pair Group Method with Arithmatic Mean) dendrogram based on RAPD-PCR, B. angustifolia and B. glabrescens were found to be a sister group, and B. kiyakii and B. cappadocica were a sister group. Principal Component Analysis (PCA) analysis based on RAPD-PCR were compatible with the UPGMA dendrogram. In the UPGMA dendrogram based on ISSR-PCR, B. kiyakii and B. glabrescens were found to be sister groups, and B. cappadocica was closely related to this group. PCA analysis based on ISSR-PCR were compatible with the UPGMA dendrogram. As a result, both RAPD and ISSR results a high rate of polymorphism were obtained. The results were compared with previous sequence-based studies, morphological, anatomical and palynological studies.

In this study, molecular characterization of Dittrichia viscosa (L.) Greuter populations collected from Aydın province was carried out based on ISSR technique and chloroplast DNA trnL intron sequences. 10 ISSR primers were used to determine the molecular characterization among the populations. For cpDNA trnL intron amplification, trnC and trnD primers were used. In ISSR analysis, a total of 70 bands were obtained. The polymorphism rate was determined to be approximately 94.28%. According to the ISSR analysis, the UPGMA dendrogram consisted of three groups. For cpDNA trnL intron sequences, phylogenetic analyses were obtained along with genetic distances. For populations, cpDNA trnL intron sequences were determined between 454 and 472 bases. The maximum likelihood phylogenetic tree consist of two clades. In addition, cpDNA trnL intron sequences of some species (Chrysophthalmum Pulicaria Inula Jasonia Stenachaenium Carpesium Blumea Iphiona Limbarda Rhanterium Lifago Duhaldea Chiliadenus) of Asteraceae family from NCBI with Dittrichia viscosa populations phylogenetic tree was constructed. As a result of the study, it was determined that the polymorphism rate of Dittrichia viscosa populations obtained by ISSR-PCR was higher than the results obtained from trnL intron sequences.

In this study, genetic variation and phylogenetic analysis of 13 populations of 6 species belonging to Conringia genus spreading in Turkey were performed using RAPD markers. Genomic DNA isolation from the leaves of the Conringia plant samples was performed via using a commercial kit. Seven RAPD primers were used to identify the genetic diversity between the populations. Polymerase Chain Reaction (PCR) was performed using DNA samples and primers. PCR products were resolved using agarose gel electrophoresis and visualized under UV light. All gel images were analyzed, and the absence and presence of polymorphic bands were scored. The total of 34 DNA bands were detected by seven RAPD primers. PAUP 4.0b10 analysis program was used to calculate phylogenetic tree and genetic distances between the species. The phylogenetic tree was obtained using the UPGMA algorithm and it was composed of two clades. According to the PAUP analysis, the species having the closest distance between each other are C. planisiliqua (Ankara-AyaG) and C. planisiliqua (Ankara-Nallihan) with the value of 0.000 and those having the longest distance are C. grandiflora (Akseki Cukurkoy) and C. orientalis (Elazig-Baskil) with the value of 0.6000. The results suggest that the RAPD markers are useful tools to demonstrate the genetic relationships between populations of the Conringia species.

- MakaleAçık Erişim2020Scopus SJRQ3ANTI-UREASE, TOTAL PHENOLIC CONTENT AND ANTIOXIDANT ACTIVITIES OF SOME Inula L. (ASTERACEAE) TAXA IN TURKEY
Sevindik, Emre; Aydin, Sinem; Paksoy, Mehmet Yavuz; Sokmen, Bahar Bilgin
The genus Inula belongs to family Asteraceae and it is medicinally important because it possesses cytotoxic, anticancer, hepatoprotective, antibacterial and anti-inflammatory properties. The aim of this research was to explore antioxidant and anti-urease activities of fifteen Inula taxa in Turkey. Ethanol extracts of Inula taxa were obtained by Soxhlet extraction. Antioxidant activity was determined through total antioxidant activity, total phenolic content and copper reducing antioxidant capacity (CUPRAC) analyses. The highest total phenolic content and total antioxidant capacity were found in I. orientalis (158.54±0.002 µg GAE/mL) and I. macrocephala (189.16±0.002 µg AAE/mL). CUPRAC activity increases with the increasing concentration of the extracts and the standard. Moreover, all the tested plants exhibited better anti-urease activity than standard thiourea except for I. anatolica and I. thapsoides subsp. thapsoides. Results obtained from this study demonstrate that Inula species might be utilized as a probable natural source in the pharmaceutical and food industries but further and more specific studies are needed to determine active antioxidative and anti-urease compounds in the extracts.
