The Lamiaceae family, encompassing diverse plant species, holds significant value in food, medicine, and cosmetics. Within this family, Pentapleura subulifera and Cyclotrichium glabrescens, relatively unexplored species, were investigated for their chemical composition, antioxidant capacity, and enzyme-inhibiting effects. The chemical composition of hexane, methanolic, and aqueous extracts from P. subulifera and C. glabrescens were analyzed using LC-ESI-MS/MS and the non-polar hexane fraction was investigated via GC-MS. The antioxidant potential of the extracts was determined through radical scavenging, reducing power and metal chelating assays. Additionally, inhibitory activity against six enzymes – acetylcholinesterase (AChE), butyrylcholinesterase (BChE), tyrosinase, amylase, and glucosidase – was examined. The aqueous extract of P. subulifera and the methanolic extract of C. glabrescens exhibited elevated phenolic content at 129.47 mg gallic acid equivalent (GAE)/g and 55.97 mg GAE/g, respectively. Chemical profiling of the constituents of the two plant species resulted in the identification of a total of twenty compounds. The majority of which belonged to flavonoids and quinic acid derivatives, primarily concentrated in the methanol and aqueous extracts. Among all antioxidant assays, the aqueous extracts of P. subulifera demonstrated superior antioxidant activity, with the highest recorded activity of 404.93 mg trolox equivalent (TE)/g in the cupric reducing antioxidant capacity (CUPRAC) test. Meanwhile, the hexane extract of C. glabrescens exhibited the highest AChE inhibitory activity at 2.71 mg galanthamine equivalent (GALAE)/g, followed by the methanol extract of P. subulifera at 2.41 mg GALAE/g. These findings unequivocally establish the notable antioxidant and enzyme inhibitory activity of P. subulifera and C. glabrescens extracts, underscoring their potential as a source of valuable natural antioxidants.

Cytoprotective, antimutagenic/antirecombinogenic and antibacterial properties of Lallemantia iberica extracts

Yilmaz Kardas, Begumhan; Diken, Mehmet Emin; Bayhan, Hamza; Acar, Mikail; Dogan, Serap

BACKGROUND: Plants have important chemical compounds to protect organisms from many mutagens but they may also have dangerous toxic substances that must be tested before use. Lallemantia iberica has been used by local people for treatment of some diseases but it was never tested for its cell protective and antimutagenic/antirecombinogenic properties. In this study, it was aimed to determine the cytoprotective, antimutagenic/antirecombinogenic, antibacterial, antioxidant and phenolic profile of L. iberica by MTS assay, micronucleus test/somatic mutation recombination test (SMART), disc diffusion method, ABTS/DPPH tests and liquid chromatography (LC), respectively. RESULTS: MTS assay results showed that both of the extracts supported the normal growth of healthy human lymphocytes and there was no significant difference between extracts. Although there was no significant increase in micronucleus concentration (‰) of the cultures treated with ethanol or methanol extracts, methanol extract caused slightly lower micronucleus concentration (12.64 ± 3.65‰) than the ethanol extract (24.46 ± 4.50‰). SMART results showed that ethanol and methanol extracts prevented harmful mitotic recombination. Lallemantia iberica also showed antibacterial activity against all of the strains tested, and the largest inhibition zones were observed with Pseudomonas aeruginosa (17.9 mm) and Enterobacter aerogenes (13.8 mm). Methanol extracts showed better antioxidant activities (ABTS IC50: 104.07 μg mL−1, DPPH IC50: 58.8 μg mL−1) than ethanol extracts (ABTS IC50: 344.03 μg mL−1, DPPH IC50: 233.7 μg mL−1). According to the LC results, the most abundant phenolic compounds were rutin hydrate (1796.4 ± 62.9 mg kg−1) and p-coumaric acid (228.1 ± 7.60 mg kg−1). CONCLUSION: Lallemantia iberica extracts had cell-protective, antimutagenic/antirecombinogenic, antibacterial and strong antioxidant characteristics in relation to their rich phenolic content.

Selvi, Selami; Polat, Rıdvan; Çakilcioğlu, Uğur; Celep, Ferhat; Dirmenci, Tuncay; Ertuğ, Zafer Füsun

Medicinal plants have been used by humans for the treatment of various diseases for thousands of years from past to present. Members of the Lamiaceae family are among the most preferred medicinal plants due to the wide variety of secondary components they contain, particularly essential oils. In this review, Master’s and PhD theses and books based on ethnobotanical studies investigated between 1960 and 2021 as well as internationally recognized databases (PubMed, Scopus, Google Scholar, Web of Science, SciFinder, Springer and Elsevier) were used to determine the medicinal uses of Lamiaceae taxa among the people in Turkey. As a result of the study, it was found that a total of 221 taxa (192 species) belonging to 29 genera, 51 of which are endemic, have medicinal/therapeutic uses. The local names of these taxa, the usage which are used among the people, the diseases in which they are used, and the geographical regions given in the present study. The genera of taxa most commonly used for medicinal purposes by the local people are Salvia (37 taxa), Sideritis (25 taxa), Stachys (22 taxa), Thymus (16 taxa) and Origanum (13 taxa). It was observed that the most frequently used two methods while preparing these plants for use was infusion (61%), and decoction (24%). Studies conducted on National (TURK, BHP), Regional (EU) and International Pharmacopoeias (WHO) and various monographs (AHP, COMISSION E, EMA, ESCOP, PDR) have revealed that there are 29 taxa included in these pharmacopoeias and monographs whose purpose of use are similar to the diseases that local people are trying to treat. Diseases that people use plants for therapeutic purposes are grouped into 12 categories. It was determined that these plants were widely used for therapeutic purposes in gastric disorders as well as otolaryngology and respiratory system diseases. This review briefly discusses whether some taxa commonly used by the local people in the treatment of diseases can be useful in the treatment of the disease in question, in the light of scientific studies.

Kılınç, Namık; Açar, Mikail; Tuncay, Salih; Karasakal, Ömer Faruk

Background: The 2019 novel coronavirus disease (COVID-19) has caused a global health catastrophe by affecting the human population around the globe. Unfortunately, there is no specific medi-cation or treatment currently available for COVID-19. Objective: It is extremely important to find effective drug treatment in order to put an end to this pandemic period and return to normal daily life. In this context and considering the urgency, rather than focusing on the discovery of novel compounds, it is critical to explore the effects of existing herbal agents with proven antiviral properties on the virus. Methods: Molecular docking studies were carried out employing three different methods, Glide extra precision (XP) docking, induced fit docking (IFD), and molecular mechanics/generalized born surface area (MM/GBSA), to determine the potential antiviral and antibacterial effects of 58 phytochemicals present in Rosmarinus officinalis, Thymbra spicata, Satureja thymbra, and Stachys lavandulifolia plants against the main protease (Mpro) and angiotensin-converting enzyme 2 (ACE2) enzymes. Results: 7 compounds stood out among all the molecules, showing very high binding affinities. Accord-ing to our findings, the substances chlorogenic acid, rosmarinic acid, and rosmanol exhibited extremely significant binding affinities for both Mpro and ACE2 enzymes. Furthermore, carnosic acid and alpha-cadinol showed potent anti-Mpro activity, whereas caffeic acid and carvacrol exhibited promising anti-ACE2 activity. Conclusion: Chlorogenic acid, rosmarinic acid, rosmanol, carnosic acid, alpha-cadinol, caffeic acid, and carvacrol compounds have been shown to be powerful anti-SARS-CoV-2 agents in docking simulations against Mpro and ACE2 enzymes, as well as ADME investigations.

In vitro biological activity of Salvia fruticosa Mill. infusion against amyloid β-peptide-induced toxicity and inhibition of GSK-3β, CK-1δ, and BACE-1 enzymes relevant to Alzheimer's disease

Gürbüz, Perihan; Dokumacı, Alim Hüseyin; Gündüz, Miyase Gözde; Perez, Concepcion; Göger, Fatih; Paksoy, Mehmet Yavuz; Yerer, Mükerrem Betül; Ömür Demirezer, L.

Salvia species have been traditionally used to improve cognition and have been proved to be a potential natural treatment for Alzheimer's disease. Salvia fruticosa Mill. (Turkish sage or Greek sage) demonstrated to have anticholinergic effects in vitro. The aim of this study was to understand the mechanism underlying the neuroprotective effects of S. fruticosa infusion and its representative compound rosmarinic acid, which was detected by LC-DAD-ESI-MS/MS. The protective effects of the S. fruticosa infusion (SFINF) and its major substance rosmarinic acid (RA) on amyloid beta 1–42 -induced cytotoxicity on SH-SY5Y cells together with p-GSK-3β activation were investigated. Their in vitro inhibitory effects against glycogen synthase kinase 3β, β-secretase, and casein kinase 1δ enzymes were also evaluated. The results showed that treatment with the all tested concentrations, SFINF significantly decreased Aβ 1–42-induced cytotoxicity and exhibited promising in vitro glycogen synthase kinase 3β inhibitory activity below 10 µg/mL (IC50 6.52 ± 1.14 µg/mL), in addition to β-secretase inhibition (IC50 86 ± 2.9 µg/mL) and casein kinase 1δ inhibition (IC50 121.57 ± 4.00). The SFINF (100 µg/mL and 250 µg/mL) also activated the expression of p-GSK-3β in amyloid beta 1–42 treated SH-SY5Y cells. The outcomes of this study demonstrated that the S. fruticosa infusion possessed activity to prevent amyloid beta 1–42 -induced neurotoxicity and provided proof that its mechanism may involve regulation of p-GSK-3β protein.

Homoploid Hybridization and Its Role in Emergence and Diversity of the Genus Origanum L. (Lamiaceae)

Dirmenci, Tuncay; Arabacı, Turan; Özcan, Taner; Yazıcı, Türker; Martin, Esra; Çelenk, Sevcan; Açar, Mikail; Üzel, Devrim

Hybrid specimens can be occurred as same ploidy level (homoploidy) or not (polyploidy) with its parents. Homoploid hybrids are originated without any change in their ploidy level (2n) and polyploid hybrids are produced with the change in ploidy level (3n, 4n etc.). According to recent studies, the frequency of natural hybridization (homoploid and polyploid) has been estimated to 25% of all flowering plants. Recent studies show that the genus Origanum has homoploid hybrids. Hybridization is a common phenomenon among the species of Origanum in overlapping distribution habitats (natural or cultivated areas). Ietswaart suggested that the hybridization process is a significant factor in the speciation of Origanum, and Origanum is probably caused by the hybridization of the old tribe Saturejeae (now tribe Mentheae) genera. Based on the hypothesis of Ietswaart, we can discuss the emergence of the genus Origanum, its intrageneric speciation and its hybrids having the possibility for speciation via hybridization using morphological and cytological evidence. During the field studies, it was seen that many populations of wide-distributed species were overlapping, and some Origanum putative hybrid specimens were also observed in these overlapping areas with morphological differences from their parents. All studied Origanum species and hybrids have 2n = 30 chromosome numbers to define homoploidy. Palynological characters were revealed, and intermediate pollen structure (size, P/E ratio etc.) was seen in the hybrids between their parents. In addition, DNA extractions were done from all the hybrids and their parents. nrITS DNA was sequenced. Hybrid specimens have some heterozygous DNA loci in comparison to their parents. This study presents some evidence about homoploid the hybridization of the genus Origanum put forward by Ietswaart. If habitat, ecological and geographic isolations occur between the hybrids and their parents, these hybrids can be speciation via homoploid hybridization in the future.

Comparative Micromorphological and Anatomical Investigations on the Subsection Distantes R. Bhattacharjee (Stachys L./Lamiaceae)

    Açar, Mikail; Satil, Fatih

In this study; comparative anatomical and micromorphological studies were performed on the subsection Distantes of the Stachys L. genus (Lamiaceae). Also, it was determined distinctive anatomical characters for the taxa of subsection in the study. In anatomical studies, free-hand cross-sections and the paraffin embedding method was applied with the microtome. Micromorphological studies were performed in LM (Light Microscope) and SEM (Scanning Electron Microscope). In anatomical studies, besides of similarities in stem and leaf structures there were also some differences in taxa. In micromorphological studies, the trichome and nutlet characteristics were found to be useful characters on taxa and at species and subspecies level. As a result of the study, the presence of sclerenchyma like a bundle in the stem structure and the presence and types of glandular trichomes and ornamentation on the surface of nutlet, the presence of the wing and ventral structure in the nutlet were important characters that can be used to distinguish taxa. According to the numerical findings, trichome and nutlet micromorphological characters were more important than anatomical characters.

A rearranged homoploid hybrid species of Origanum (Lamiaceae): O. ×munzurense Kit Tan & Sorger

Dirmenci, Tuncay; Özcan, Taner; Açar, Mikail; Arabacı, Turan; Yazıcı, Türker; Martin, Esra

Origanum munzurense Kit Tan & Sorger (Sect. Brevifilamentum Ietsw.) was described by Kit Tan & Sorger in 1984, and it is an accepted species related to O. acutidens (Hand.-Mazz.) Ietsw. belonging to sect. Brevifilamentum. It is morphologically intermediate between O. acutidens and O. vulgare L. subsp. gracile (K.Koch) Ietsw., and its type locality lies in the overlap zone between the geographic distributions of these two taxa. We hypothesized that O. munzurense is in fact a hybrid between the two aforementioned taxa and we tested this hypothesis with the general morphology, chromosome features, and nuclear (ITS1–ITS2) and chloroplast (rpl32) DNA sequences of specimens of O. munzurense and its putative parental species, O. vulgare subsp. gracile and O. acutidens. The somatic chromosome number of O. munzurense, O. vulgare subsp. gracile and O. acutidens were counted as 2n = 30. Origanum munzurense presented polymorphic sites in the nuclear ITS region, but the chloroplast markers did not allow us to identify the putative maternal of O. munzurense. Our results suggest that O. munzurense is indeed a hybrid, and its rearrangement as a new homoploid hybrid (O. ×munzurense) would be more accurate. These present results support previous reports for the speciation of Origanum members via hybridization.
