Effect of the organophosphate insecticide chlorpyrifos exposure on oxidative stress and quality of Salmo coruhensis spermatozoa

Kutluyer, Filiz; Kocabaş, Mehmet; Erişir, Mine; Benzer, Fulya

The use of pesticides has been increasing along with increasing farming activities and has caused deleterious environmental impacts. Non-target organisms in particular, including fish, are affected by pesticides. In this work, the impacts of Chlorpyrifos (CPF) on sperm oxidative stress markers and sperm motility were investigated in vitro. CPF concentrations were 0 μg/L (control), 5 μg/L, 10 μg/L and 15 μg/L. Lipid peroxidation [malondialdehyde (MDA)], nonenzymatic antioxidants [glutathione (GSH)] and enzymatic [superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT)] activities in sperm cells were examined for the determination of oxidative stress status. Our findings showed that motility and survival of sperm cells significantly decreased with exposure to Chlorpyrifos. Biochemical assays revealed that CAT activity and levels of MDA and, GSH increased in spermatozoa based on CPF concentration while activity of GSH-Px and SOD decreased. Consequently, spermatozoa were highly sensitive to CPF exposure. It can be deduced that CPF has the potential to disrupt sperm quality and to cause to oxidative stress in sperm cells of S. coruhensis.

Phenotypic differentiation analysis: A case study in hybridizing Çoruh trout (Salmo coruhensis), Rize trout (Salmo rizeensis) and brown trout (Salmo trutta fario)

Kocabaş, Mehmet; Kutluyer, Filiz; Başçinar, Nadir

The aim of this study was to determine and compare the phenotypic variation in Çoruh trout (Salmo coruhensis), Rize trout (Salmo rizeensis), brown trout (Salmo trutta fario) and their native hybrids (S. rizeensis × S. t. fario; S. t. fario × S. coruhensis; S. rizeensis × S. coruhensis). The numbers and diameters of red and black spots were determined on operculum, fins and above and below the lateral line. The results from this study indicate that there were differences in spotting pattern, colour pattern or fin pigmentation traits between S. rizeensis, S. t. fario, S. coruhensis and their hybrids. Consequently, native S. rizeensis, S. t. fario and S. coruhensis were affected by interaction and phenotypic plastic aspects of traits in S. rizeensis were dominant in their hybrids (S. rizeensis × S. t. fario; S. rizeensis × S. coruhensis).

Kutluyer, Filiz; Kocabaş, Mehmet; Erişir, Mine; Benzer, Fulya

The use of insecticides has been increasing along with increasing agriculture activities and has caused deleterious environmental impacts. Non-target organisms in particular, including fish, are affected by pesticides. In this work, the impacts of cypermethrin (CYP) on sperm oxidative stress markers and sperm motility were investigated in vitro. The CYP concentrations were 0 μg L-1 control, ethanol), 1.025 μg L-1, 2.05 μg L-1 and 4.1 μg L-1. Lipid peroxidation [Malondialdehyde (MDA)], non-enzymatic antioxidants [glutathione (GSH)] and enzymatic [superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT)] activities in sperm cells were examined for determination of oxidative stress status. Our findings showed that motility and survival of sperm cells significantly decreased with exposure to CYP. Biochemical assays revealed that CAT activity and levels of MDA and, GSH increased in spermatozoa based on CYP concentration while activity of GSH-Px and SOD decreased. Consequently, spermatozoa were highly sensitive to CYP exposure. It can be deduced that CYP has the potential to disrupt sperm quality and to cause to oxidative stress in sperm cells of S. coruhensis.

Kocabaş, Mehmet; Kutluyer, Filiz; Benzer, Fulya; Erişir, Mine

The use of pesticides has been increased along with increasing the farming activities and has caused environmental impacts deleteriously. In particular, non-target organisms including fish can be affected by toxic effects of pesticides. Therefore, the impacts of malathion (MTN) on oxidative stress and sperm quality were investigated in vitro. The MTN concentrations used on this study were 0 (control), 75, 100, and 125 μg/L. Lipid peroxidation (MDA), non-enzymatic (GSH), and enzymatic (SOD, GSH-Px, and CAT) activities in spermatozoa were examined for determination of oxidative stress status. Our findings showed that motility rate and period of sperm cells significantly decreased with exposure to MTN. Biochemical assays revealed that CAT activity and levels of MDA, GSH increased in spermatozoa based on concentration while activity of GSH-Px and SOD decreased. Consequently, spermatozoa were highly sensitive to MTN exposure. MTN has disruptive effects on sperm quality and caused to oxidative stress in spermatozoa.

Effect of Cobalt on Sperm Motility in an Endangered Trout Species, Salmo coruhensis

    Kocabaş, Mehmet; Kutluyer, Filiz

Experiments were designed to examine the in vitro effect of cobalt on sperm motility of the endangered trout species (Salmo coruhensis). Sperm samples were diluted in an immobilizing solution, and activated in a motility-activation solution that was supplemented with cobalt at concentrations of 1, 10, 100 and 1000 mg/L. The percentage of motile sperm and duration of motility were determined. Cobalt concentrations of 1–100 mg/L had a positive effect on the percentage of motile sperm and duration of motility compared to the control group, while a concentration of 1000 mg/L resulted in decreases in these parameters (p < 0.05). The percentages of motile sperm at cobalt concentrations of 0, 1, 10, 100 and 1000 mg/L were 83.33% ± 0.25%, 88.33% ± 0.34%, 89.00% ± 0.57%, 90.00% ± 0.87% and 42.50% ± 0.45%, respectively; and the time durations over which the sperm remained motile were 72.00 ± 0.63, 74.83 ± 0.28, 77.40 ± 0.47, 81.14 ± 0.78, and 50.25 ± 0.67 s, respectively. This study has shown that sperm motility and duration were significantly enhanced (p < 0.05) at cobalt concentrations of 1, 10 and 100 mg/L, relative to controls, and significantly decreased at 1000 mg/L.

The probiotics are very important for aquaculture because of their role in enhancing the fish health status, improving disease resistance, growth performance, and body composition, reducing malformations, improving gut morphology and the overall microbial balance. The objective of the presently reported study was to investigate effects of kefir as a probiotic product on the digestive and hepatic enzyme activities, as well as glucose level of Coruh trout, Salmo coruhensis Turan, Kottelat et Engin, 2010. The experiment was performed with four treatments: control group (no kefir), D1, D2, and D3 (kefir-supplemented diet: 10, 20, and 40 g . kg(-1) fish body mass, respectively). The activity of: alanine aminotransferase (ALT), aspartate aminotranferase (AST), lactate dehydrogenase (LDH), lipase, amylase-as well as the glucose levels were determined after 4 months. The results indicated that glucose levels (P < 0.05) and digestive enzymes (lipase and amylase) in the blood serum decreased in fish fed diets containing kefir (P > 0.05). On the contrary, the activity of liver enzymes (ALT, AST, and LDH) increased insignificantly (P > 0.05). In conclusion, digestive and liver enzyme activities were affected by quantitative changes in dietary kefir insignificantly but further research is needed to establish whether the liver and digestive enzyme physiology of Coruh trout can be affected by kefir-supplemented diets in a long-term trial.

Can, Erkan; Kurtoglu, Ilker Zeki; Benzer, Fulya; Erisir, Mine; Kocabas, Mehmet; Kizak, Volkan; Kayim, Murathan; Çelik, Huseyin Tugrul

The objective of the present work is to examine the effects of different doses (0, 10, 20 and 40 g kg(-1) fish diet mass) of kefir on growth performance and oxidant-antioxidant status in the blood and liver tissues of Coruh trout, Salmo coruhensis, in different periods (2 and 3 months). In this study, survival was more than 88.3%, and irrespective of dietary kefir levels at the end of the study. There were no significant differences in SGR (Specific growth rate), FCR (Feed conservation rate), CF (Condition factor) among fish fed diets with 0, 10, 20 and 40 g kg(-1) kefir, however, these growth values were significantly different in terms of durations between 2-month and 3-month treatment (P<0.05). Our results indicated that TAS (Total Antioxidant Status) and TOS (Total Oxidant Status) in blood tissues reduced with kefir treatment at the end of third month (P<0.05). Malondialdehyde (MDA) levels in liver decreased in all groups compared with control group and a considerable extent decrease was observed in 40 g dose of kefir treatment at the end of third month. The data obtained from this experiment indicated that the same dose of kefir was more effective at the end of 3-month treatment than 2-month treatment (P<0.001). Although there was no statistical difference among groups, an increase in the glutathione peroxidase (GSH-Px) enzyme activity was observed in all groups compared to control groups. While catalase (CAT) activity decreased in all groups compared to control group at the end of second month (P<0.01), the decrease was insignificant level at the end of third month. In glutathione (GSH) levels, statistical differences were no observed in all groups compared to the control group with 2-month treatment while there were significant increases with 3-month treatment (P<0.001). It was concluded that kefir could play an antioxidant role and its effectiveness depended on dosage and time of application in Coruh trout, S. coruhensis.
