The purpose of this study was to examine the changes to occur in egg production, egg quality traits, hatchability, and oxidative stress parameters of laying quails fed with high calorie diets and effect of milk thistle (Silybum marianum) seed on these changes. A total of 75 45-day-old quails including 60 females and 15 males were used in the study. The quails were divided into 5 groups with three repetitions. 4 females and 1 male were used in each repetition. The groups of the study were arranged as following; control group (C) consuming corn-soybean based basal diet, oil group (SFO) in which 5% sun flower oil was added into basal diet, oil + milk thistle group (SFO+MT) in which 5% sun flower oil + 1% milk thistle seed was added into basal diet, syrup group (CS) in which 10% corn syrup was added into basal diet, and syrup + milk thistle group (CS+MT) in which 10% corn syrup + 1% milk thistle was added into basal diet. Total egg production was found to be significantly higher in SFO and SFO+MT groups of present study (P<0.05). Feed intake, total egg weight and feed conversion were found similar among the groups (P>0.05). The differences among the groups in albumen rate, yolk rate, dried shell rate and shape index were not statistically significant (P>0.05). The best yolk colour was obtained in SFO+MT and CS groups (P<0.01). There were no statistically differences among the groups in hatchability, hatchability of fertile and embrionic mortality (P>0.05), but the difference in examined fertility were statistically significant (P<0.05). Malonyl dialdehyde (MDA) values of blood, liver and heart tissues were similar among the groups of study (P>0.05). MDA level of kidney significantly increased in CS group, however addition of milk thistle seed into diet was reduced MDA level of kidney in CS+MT group (P<0.01). High calorie diets caused changes in antioxidant system in blood and tissues. Consequently, high-calorie basal diets especially addition of SFO improved only egg production and fertility rate. However feeding quails with corn syrup had significantly higher lipid peroxidation in kidney. Supplementation of milk thistle seed into CS group may protect kidney against free radical damage.

This study has been carried out to investigate the presence of Yersinia ruckeri, the causative agent of Enteric Red Mouth (ERM) disease, in rainbow trout fish breeding farms located in Elazig and Malatya provinces in Eastern Turkey. For this purpose, bacteriological culture and a specific polymerase chain reaction (PCR) have been compare with examine blood and intestine samples collected from adult and young rainbow trout fish as well as water samples collected from all the farms. Y. ruckeri was isolated and identified from eight of the 17 fish farms giving a proportion of 52.9% at fish farm. The disease was noted in five of the rainbow trout farms located in Elazig and three of the 11 rainbow trout farms located in Malatya. The agent was detected in five of the six young fish breeding farms. The isolation percentages of the causative agent at fish level were calculated as 25.7% (131/510) in adult and 31.7% (19/60) in young fish, giving an overall proportion of 26.3% (150/570). All the isolates have been successfully identified as Y ruckeri by species-specific PCR. No isolation could be made from water samples. It is therefore concluded that Y. ruckeri infection poses a significant threat to the fish farms in the study area and necessary precautions should urgently be taken in order to minimize economical losses due to ERM disease. A specific PCR can be utilized successfully in aquaculture for rapid identification of bacterial agents which will help to prevent spread of infectious diseases and will therefore contribute to the productivity of fishery sector.

Staphylococcal food poisoning is one of the most prevalent causes of gastroenteritis throughout the world. This study has three steps. In the first step of the study, different food matrices were analyzed for the presence of Staphylococcus aureus. In the second step, types of staphylococcal enterotoxins (SET) from these foods were analyzed. Lastly, antibiotic resistance of S. aureus isolated was determined using conventional and PCR methods. For this purpose, 300 different food samples were evaluated for contamination with S. aureus. From these samples 221 S. aureus strains were isolated. The same strains were evaluated for antibiotic resistance and existence of mecA gene. All strains were at least resistant to 2 antibiotics, but no MRSA strain was detected. Staphylococcal Enterotoxins (SE) were determined with VIDAS and ELISA from the foods of concern. The results of the analysis held with Vidas SET2 showed 72 (24%) of 300 samples and with ELISA these enterotoxins showed difference in distribution.

The present study was undertaken to investigate the presence of Escherichia coli O157:H7 in cakes (plain, with cacao, with fruit) marketed in Elazig. A total of 200 cake samples as 100 plain, 50 with cacao, and 50 with fruit were analysed. Following the enrichment, the samples were plated on to CT-SMAC agar. A total of 208 typical colonies (colourless) as 138 colonies from 65 plain cakes, 25 colonies from 8 cakes with cacao, and 45 colonies from 19 cakes with fruit were isolated. These colonies were subjected to further analysis given in the materials and methods section for identification of serotype O157:H7 of E. coli. However, no colonies representing E. coli O157 were detected in any of the samples. Therefore, analysis was not proceeded for determining H7 in any of the isolates. E. coli O157:H7 was not detected in any of the cake samples analyzed. Although this finding is a good news for public health, considering other pathogens, which may exist in cakes, it is concluded and reemphasized that strict hygienic practices should be applied at every stage of cake production.

- MakaleYalnız künye2010Scopus SJRQ3Disinfection of Gilthead Sea Bream (Sparus aurata), Red Porgy (Pagrus pagrus), and Common Dentex (Dentex dentex) Eggs from Sparidae with Different Disinfectants
Can, Erkan; Saka, Sahin; Firat, Kuersat
The aim of this study is to determine suitable conditions for egg disinfection of gilthead sea bream (Sparus aurata), red porgy (Pagrus pagrus), and common dentex (Dentex dentex) with glutaraldehyde, hydrogen peroxide and iodine. The eggs were disinfected with 200 ppm glutaraldehyde (T= 2, 4, 8, 16 min), 300 ppm hydrogen peroxide and iodine (T= 5, 10, 15, 20 min) at four different contact times. The hatching rate was determined in triplicates for each treatment. Also, bacteria colonies were counted on tryptic soy agar (TSA) and thiosulphate citrate bile salt sucrose agar (TCBS). At the end of the experiment, bacterial load and hatching rate were assessed together. For glutaraldehyde treatment, optimal hatching rates and bacterial colonies were estimated as CT (Disinfectant concentration in mg.l(-1)* exposure time in minute) 800 for sea bream eggs, CT 1600 for red porgy eggs and CT 800-1600 for common dentex eggs. Also, for hydrogen peroxide treatments, they were determined as CT 3000 for sea bream and red porgy and CT 4500 for common dentex. Finally, in iodine treatments, these parameters were measured as CT 4500-6000 for all species' eggs. However, in our study, it is clear that the glutaraldehyde showed the optimum disinfection effect against the microorganisms because the bacteria could be eliminated completely.
