Sevindik, Emre; Gelir, Seda Nur; Demircan, Şelale; Çiftçi, İbrahim; Paksoy, Mehmet Yavuz

Absract: In this study, chloroplast DNA (cpDNA) ndhF sequence analysis was used to determine the sequence analysis and genetic diversity of Chrysophthalmum Schultz Bip. species distributed in Türkiye. Genomic DNA was isolated from green leaves and cpDNA ndhF region was amplified by PCR. The results of the PCR, the forward and reverse sequences were edited using the Sequencher 5.4.6 programme and the contig sequences were obtained. To determine the phylogenetic relationships between species, MEGA 6.0 was used to construct a maximum likelihood (ML) tree. Using the same program, nucleotide composition, phylogenetic trees, genetic distance matrix, and genetic diversity analyses were performed. In addition, the physicochemical properties and 3D structure of ndhF proteins were determined. From the results of the ndhF analyses, the nucleotide length of Chrysophthalmum species was found to vary between 471 and 625. The genetic matrix value was determined to be between 0.000 and 0.009. In the phylogenetic tree constructed using ndhF sequences from Anvillea DC., Pulicaria Gaertn., Limbarda Adans, Inula L., Carpesium L., Pentanema Cass., Dittrichia Greuter, Rhanteriopsis Rauschert and Telekia Baumg species from NCBI, C. montanum was grouped with Inula, Pentanema and Rhanteriopsis species. C. gueneri was grouped with C. dichotomum and Pentanema alanyense. Previous phylogenetic studies were in agreement with our results. In addition, the ndhF results supported that Chrysophthalmum species are not a monophyletic group.

An Artificial Neural Network Modeling of Solar Drying of Mint: Energy, Exergy, and Drying Kinetics

Gülçimen, Fevzi; Karakaya, Hakan; Durmuş, Aydın

Abstract: Solar drying is an environmentally friendly method for preserving agricultural products; however, limited studies have jointly examined energy–exergy performance and data-driven prediction methods under varying airflow conditions, which restricts performance optimization. This study aims to investigate the effects of inlet airflow rate on energy utilization ratio, energy generation, exergy loss, exergy efficiency, and drying kinetics of mint leaves, and to evaluate the predictive capability of artificial neural network modeling. Thin-layer drying experiments were conducted in a forced convective solar dryer equipped with a newly designed solar air collector at different air mass flow rates. Energy and exergy analyses were conducted using experimental thermodynamic data, and energy utilization and exergy loss were modeled with a feed-forward back-propagation artificial neural network. The energy utilization ratio ranged from 7.45 to 87.1% and increased with decreasing airflow rate. The average exergy losses for air mass flow rates of 0.012, 0.026, and 0.033 kg s–1 were 16.2, 8.2, and 6.88 W, respectively. Both experimental and artificial neural network results showed that increasing airflow rate reduced energy utilization ratio and exergy loss. The artificial neural network predictions exhibited very close agreement with experimental data. The results show that artificial neural networks can accurately predict energy utilization ratio and exergy loss in a forced convective solar dryer. Airflow rate is a key parameter for thermodynamic performance, and the integrated experimental–ANN approach offers a robust tool for optimizing and designing solar drying systems.

Antioxidant Enzyme Responses to Lead Exposure in Daphnia magna

    Sahilli, Yeliz Cakir

Abstract: Lead (Pb), a pervasive environmental contaminant, poses significant toxicological risks to freshwater ecosystems. Daphnia magna, a widely used model organism in aquatic toxicology, provides valuable insights into the sublethal effects of heavy metal exposure. This study investigated the effects of Pb exposure on antioxidant defense mechanisms in D. magna. Test organisms were exposed to aqueous Pb solutions at varying concentrations (0, 10, 20, 30 μg/L) for 96 h. The enzyme activities of glutathione peroxidase (GPx), catalase (CAT), and superoxide dismutase (SOD) were measured using enzyme-linked immunosorbent assay (ELISA). SOD activity did not differ significantly between the exposed and control groups. However, CAT activity showed a significant decrease in all Pb-exposed groups compared to controls. GPx activity exhibited a non-significant decrease. In conclusion, Pb exposure may induce oxidative stress by generating reactive oxygen species (ROS) and inhibiting the function of sulfhydryl-containing antioxidant enzymes. Changes in antioxidant enzyme activity may serve as early biomarkers in ecotoxicological assessments of Pb pollution in aquatic environments. It is suggested that future studies incorporate molecular-level endpoints and chronic exposure scenarios to better elucidate the long-term impacts of Pb on aquatic invertebrates.

Production and Partial Characterisation of Bifunctional Cellulase Genes of Anaerobic Gut Fungus Orpinomyces sp. GMLF19

    Yazdic, F.C.; Akyol, I.; Ozkose, E.

Abstract: Fiber-degrading properties of anaerobic gut fungus, Orpinomyces sp. GMLF19, were investigated when it was grown on various carbon sources, namely carboxymethylcellulose, barley, oat, corncob and wheat straw. Fungi were developed under anaerobic conditions, and carboxymethylcellulase (CMCase), lichenase, and xylanase activities were determined by using DNS method. Meanwhile, the decreasing order of the enzyme activity of Orpinomyces sp. GMLF19 was observed to be lichenase > CMCase > xylanase. Relative activities of these enzymes in the isolate GMLF19 were found to be the highest when the culture was grown for 5 days. Cellulase gene domain was reproduced from chromosomal DNA and transferred into Escherichia coli with the aid of pCT plasmid vector and CMCase, lichenase and xylanase activities were determined using DNS method. The gene showed relatively higher CMCase activity and moderate lichenase activity while no xylanase activity was observed suggesting that the gene is bi-functional. The cell-associated activity of overexpressed Cel19 enzyme (E. coli cells containing this transferred gene were designated as C19) was relatively higher than extracellular activity. It was also recorded that this gene has 2 dockerin domains and 1 catalytic domain with the length of 1.585 bp, and it coded 50.59 kDa size of the protein which belongs to glycosyl hydrolase family 6.

Kocabaş, Filiz Kutluyer; Kocabaş, Mehmet; Kara, Cemil; Bozali, Nuri; Gürlek, Mustafa Emre

Abstract: Peregriana labiata (Rossmässler, 1835) is a broadly distributed Palaearctic freshwater snail belonging to the family Lymnaeidae and it is an intermediate host of Fascioloides manga causing Fasciolosis as a snail-borne trematode disease. The disease negatively influences the livestock sector and human health with increasing prevalence in the last decades. As a helpful tool, geographical distribution of P. labiata was assessed in Kelkit Basin (North-East Anatolia) as a risk area for fasciolosis transmission. 85 sampling sites were sampled from different habitats in Kelkit Basin (NE Anatolia). The first evidence of P. labiata occurrence was revealed in the basin. In addition, the effects of altitudes on abundance, distribution and shell characters of P. labiata were investigated in several ecosystems. The highest values for SL (standard length) and W (weight) were recorded as 11.80 ± 0.10 mm and 0.17 ± 0.14 g at an altitude of 1233 m, respectively. The highest occurrence of P. labiata was observed at altitudes of 728 m (17%) and 1223 m (16%). PCA analysis showed a strong correlation between SpH (spire height) and AW (aperture width). Overall, the broad range of samples gathered from different altitudes strengthened the idea that the shell characteristics vary with altitude.

Oxidative and Antioxidant Responses in Daphnia magna Following Exposure to Sub-Lethal Glyphosate Concentrations

    Sahilli, Yeliz Çakir; Korkmaz, Volkan

Abstract: Glyphosate (GLY) is one of the most widely used herbicides in modern intensive agriculture to enhance crop production. However, the chemical additives in commercial formulations and the active ingredient itself can adversely affect biological and chemical processes in aquatic organisms. Therefore, the aim of this study was to evaluate the effects of sub-lethal concentrations of GLY (0, 0.23, 0.58, and 1.17 μg/L) on oxidative stress biomarkers—namely lipid peroxidation (measured as malondialdehyde, MDA), and antioxidant enzymes (superoxide dismutase, SOD; catalase, CAT; and reduced glutathione, GSH)—in Daphnia magna after 24 and 96 h of exposure. The results showed that SOD activity increased in all treatment groups compared to the control at both time points while CAT activity decreased in all groups. GSH and TBARS (thiobarbituric acid reactive substances) levels were significantly elevated after 24 and 96 h in all exposed groups. Overall, D. magna was sensitive to sub-lethal concentrations of GLY, and changes in SOD and CAT activities, as well as GSH and TBARS levels, these biomarkers can serve as useful indicators for detecting glyphosate-induced oxidative stress and water pollution.

Kocabaş, Mehmet; Bozali, Nuri; Kocabaş, Filiz Kutluyer; Kara, Cemil; Gürlek, Mustafa Emre

Abstract: Non-indigenous species in an ecosystem increase the potential of novel ecological relationships and can dramatically affect community composition and ecosystem roles. This study examined the spatial distribution of Dreissena caputlacus and Dreissena polymorpha gallandi within the Kelkit Basin located in Northeastern Anatolia. A total of 75 sampling locations representing various habitats were surveyed in the Kelkit Basin. The presence of D. p. gallandi and D. caputlacus was identified for the first time in the basin. The effects of altitude on the population structure, geographic distribution, and shell traits of D. p. gallandi and D. caputlacus were also assessed in a range of ecosystems. The highest shell length (SL) for D. caputlacus was recorded at 566 m altitude with values of 25.58 ± 5.80 mm while the highest SL value for D. p. gallandi was 24.15 ± 5.13 mm at 610 m altitude. Principal Component Analysis (PCA) revealed a strong association among shell length (SL), shell height (SH), shell width (SW), and weight (W). In conclusion, the diverse altitudinal distribution of samples strengthened the evidence that shell features differ with changes in altitude.

Özçiçek, Esin; Kocabaş, Filiz Kutluyer; Kocabaş, Mehmet; Yilmaz, Ökkeş

Abstract: As an important challenge restricting hatchery production, gamete quality and subsequent larvae and juvenile are influenced by broodstock nutritional status (i.e., protein, essential amino acids, fatty acid profile and minerals). Hence, the fatty acid (FA) profiles of muscle and gametes in threatened Abant trout (Salmo abanticus) from wild was elucidated and quantitated using the gas chromatography. Our results indicated that the saturated fatty acids (SFAs) in muscle and sperm (38.00 and 60.98%) of S. abanticus occurred in higher quantities than unsaturated fatty acids. The egg of S. abanticus in the wild is characterized by high (mostly n-6 HUFA, 33.59%). Tricosanoic acid (C23:0) and docosahexaenoic acid (DHA, C22:6 n-3) were not determined in sperm of S. abanticus. Overall, the current study provides FA composition in the reproduction cells and muscle of mature S. abanticus and the gametes and muscle had similar general FA profiles with the dominance of C18:1 n-9 (Oleic acid), C16:0 (Palmitic acid), C4:0 (Butyric acid), C18:2 n-6 (Linoleic acid, LA) and C20:5 n-3 (Eicosapentaenoic acid, EPA).

Synthesis, Characterization and Biocompatibility of Cross-linked Dicarboxylic Acids from Waste Potato Peel Starch

    Paşa, S.; Gürler, N.; Erdoğan, Ö.; Cevik, O.

Abstract: People have become conscious about recycling or reusing many things they use in the 21st century because diminishing vital resources make it necessary, both personally and socially. The evaluation of waste is also critical in terms of national economies. In this study, starch dicarboxylic compounds were synthesized using waste potato peels (WPP) in the presence of sodium hypophosphite monohydrate as a catalyst for the formation of the ester cross-links. Adipic acid, succinic acid, and glutaric acid were employed with waste potato starch (WPS) to obtain cross-linked samples: WPS-A, WPS-S, and WPS-G, respectively. The obtained compounds were characterized by attenuated total reflectance Fourier transform infrared (FTIR-ATR), scanning electron microscopy with energy-dispersive X-ray analysis (SEM-EDX), X-ray diffraction (XRD), and their cytotoxicity properties were investigated. The biocompatibility of the obtained WPS-dicarboxylic acid compounds against NIH-3T3 and L929 murine fibroblast cells with MTT assay was determined. Studies have shown that these compounds do not harm healthy cells and are found to be bioavailable.

Synthesis and Characterization of Hydroxyapatite/TiO2 Coatings on the β-Type Titanium Alloys with Different Sintering Parameters using Sol-Gel Method

    Dikici, B.; Niinomi, M.; Topuz, M.; Say, Y.; Aksakal, B.; Yilmazer, H.; Nakai, M.

In this study, hydroxyapatite (HA) based composite films were successfully syntheses on the β-type Ti29Nb13Ta4.6Zr (TNTZ). The solutionized TNTZ substrates coated with HA and HA/Titania (TiO2) bioactive composite coatings by sol-gel method under various sintering parameters related to sintering temperatures and heating ramp rates. Microstructural observations of the coatings revealed that apatite was formed on the substrates. The hardness values of the coatings increase with increasing both the sintering temperature and the TiO2 concentration in the coatings layer. However, it was found that the heating ramp rate of the sintering was not affecting the hardness values so much. Also, the hardness values of the HA/TiO2 composite coatings at all sintering temperatures were higher than only HA coated TNTZ samples due to the existence TiO2 phases in the HA matrix. Results indicating that the doping of HA with TiO2, improve the physical consistency between the coating layer and the substrates and provide a better inter-particle bonding due to the existence TiO2 phases in the HA.

400 MHz NMR Study of Isotope Effects on Albumin in H2O/D2O Solutions

    Korunur, S.; Zengin, B.; Yılmaz, A.

Abstract: Chemical shift measurements were performed for several percentages of H2O added to D2O, in several H2O/D2O mixtures without and with a single concentration of albumin, and also in a mixture of 0.1H2O/0.9D2O containing increasing concentrations of albumin. The chemical shifts of pure mixtures decreases with increasing fraction of pure water. This is closely related to isotope effects. The chemical shift corrected for isotope effects increases with percentage of H2O. The chemical shift obtained for the mixture of 0.1H2O/0.9D2O increases with increasing concentration of albumin. These increments are consistent with fast chemical exchange in H2O/D2O mixture. In conclusion, present study reveals the presence of isotope effect on albumin in H2O/D2O solutions, and demonstrates fast chemical exchange in D2O solutions with albumin in a different way.
