Oxidative Stress Responses induced by Yttrium in Gammarus pulex at Increasing Temperatures

    Aydın, Ayşe Nur; Serdar, Osman

In parallel with the increase in the need for raw materials used in industry, the area and amount of use of Rare Earth Elements are also increasing. Yttrium (Y), which is used as a raw material source, is thought to form residues in the environment. Gammarus pulex individuals were selected as a model organism to investigate the effects of Yttrium on the environment and aquatic organisms. G. pulexes were exposed to Y at different temperatures (16, 18, and 20 °C) and concentrations (25, 100, and 500 µg/L) for 96 hours and samples were taken at 24 and 96 hours for biochemical analyses. For biomarker analysis, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities, thiobarbituric acid (TBARS) and glutathione (GSH) levels were performed using CAYMAN ELISA kit. Study data revealed that SOD and CAT activities increased significantly with increasing temperature and exposure concentration, while GPx activity showed an irregular response; in contrast, no significant change was observed in TBARS levels at 16 °C, while the increase in TBARS and significant changes in GSH levels at 18 and 20 °C indicate that oxidative stress becomes more pronounced at high temperatures. Nanoparticles (NPs) are increasingly used across various fields of science and technology, including food science from crop harvesting to final food consumption. With their widespread application in daily life, the environmental effects of nanoparticles have become an important area of academic investigation. In this study, Gammarus pulex species was selected as research material/bioindicator species to investigate the effects of zinc oxide (ZnO), copper oxide (CuO), and iron(III) oxide (Fe2O3) on aquatic life. To conduct the experiments, separate setups were established for each of the nanoparticles (ZnO, CuO, and α-Fe2O3). Following ten organisms for each concentration group (10, 20, and 40 ppm) with corresponding control groups, exposures were performed for 24 and 96 hours. Samples were collected at both time points and biochemical analyses were conducted using homogenization procedures. The results were evaluated statistically. According to the findings, SOD activity increased significantly after exposure to ZnO, CuO, and Fe2O3. CAT activity did not show any change under ZnO NP exposure, whereas a decrease was observed under CuO and Fe2O3 NP exposures. However, a significant increase in TBARS levels and a significant increase in GSH levels were observed following nanoparticle exposure. Based on the study data, exposure to ZnO, CuO, and Fe2O3 appears to induce oxidative stress in G. pulex.

Pollutants such as copper nanoparticles (CuNP), which form residues in the environment, especially in water environments, cause serious damage to the environment and the organisms living in the environment. In this study, Navicula cryptocephala var. veneta was selected as a model organism. The EC50 values of CuNP on Navicula cryptocephala var. veneta were determined as 4.94 mg\L and 3 different application concentrations were determined considering this ratio. The experiments were carried out in 3 replicates and samples were taken at 24 and 96 hours. At the end of 120 hours, samples were taken to determine the elimination values. CuNP accumulation amounts in the samples were measured by electrothermal atomic absorption spectrophotometer (ETAAS). For biomarker analysis, thiobarbituric acid (TBARS) and glutathione (GSH) levels and superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities were performed using CAYMAN ELISA kit. According to the study data, as CuNP concentration and application time increased, the amount of accumulation in Navicula cryptocephala also increased. Increasing-decreasing changes were observed in elimination. CuNP caused oxidative stress in N. cryptocephala and caused changes in TBARS and GSH levels. In addition, it caused increases in SOD activities and statistically significant decreases (P<0.05) in CAT and GPx activities.

Biochemical Responses of Zebra Mussel Dreissena polymorpha (Pallas, 1771) (Bivalvia: Dreissenidae) against Cadmium

Alişer, Ahmet Bedrettin; Aydin, Rahmi; Serdar, Osman

The biochemical reactions of zebra mussels Dreissena polymorpha (Pallas, 1771) exposed to different concentrations of cadmium (Cd) in the acute period (24 and 96 hours after treatment) were studied. Four variants of experimental treatment were examined: 0 μg/L Cd (control), 10 μg/L Cd, 20 μg/L Cd and 40 μg/L Cd. Changes in the enzyme activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione (GSH) and thiobarbituric acid reactive substances (TBARS) levels were determined using ELISA kits. In D. polymorpha exposed to the three experimental concentrations of cadmium, there was a statistically significant decrease (p<0.05) in SOD enzyme activity with increasing concentration or exposure times. No change was observed in CAT enzyme activity with increasing concentrations. The decrease in the activity depending on the exposure times was not statistically significant (p>0.05). As for GPx enzyme activity, a significant increase was detected after Cd application compared to the control (p<0.05). TBARS and GSH levels increased significantly in all Cd application groups compared to the control at 24 and 96 hours. The effects of different concentrations of Cd on D. polymorpha in the acute period were associated with changes in SOD, CAT, GPx enzyme activities. TBARS and GSH levels were effective parameters in determining the oxidative damage caused by Cd.

Neurotoxic Effect of New Generation Insecticide with Flubendiamide Active Ingredient in Gammarus pulex (Linnaeus, 1758) (Amphipoda: Gammaridae) at Different Temperatures

    Söylemez, Hilal; Serdar, Osman

This study aimed to determine the relationship between temperature and neurotoxicity of a new generation insecticide, the active ingredient of which is flubendiamide, on non-target aquatic organisms (Gammarus pulex (L., 1758)). Firstly, LC50 values of flubendiamide on G. pulex were determined at 16, 18 and 20 °C as 256.83 ± 19.59 µgL-1, 169.38 ± 34.17 µgL-1 and 155.84 ± 17.22 µgL-1, respectively. We found that the acute toxicity sensitivity of the test organism to the pesticide containing flubendiamide increased with increasing temperature. Secondly, G. pulex was exposed to non-lethal concentrations of flubendiamide pesticide for 96 hours at the above temperatures. Finally, the changes in acetylcholinesterase (AChE) enzyme activity were determined for each temperature using ELISA kits. The exposure time between the 24th and 96th h at the same temperature was found to be statistically insignificant (p>0.05). It was determined that the inhibition value of AChE enzyme activity on the test organism of the pesticide with active ingredient flubendiamide increased depending on the temperature increase. Considering the changes in AChE enzyme activities in G. pulex at different temperatures against flubendiamide insecticide, it was concluded that it can be used as a useful biomarker.

Electrocoagulation induced treatment of indigo carmine textile dye in an aqueous medium: the effect of process variables on efficiency evaluated using biochemical response of Gammarus pulex

Tanyol, Mehtap; Yildirim, Nuran Cikcikoglu; Alparslan, Dilek

Indigo carmine removal from aqueous solutions was investigated by the electrocoagulation process and biochemical responses of Gammarus pulex to solutions before and after treatment were studied. Electrocoagulation experiments in the first stage were carried out with central composite design (CCD) using response surface methodology (RSM). Optimization of process variables that maximize indigo carmine removal by electrocoagulation process was carried out by using a second-order model within the studied test range of various process variables. According to the optimization results, optimum conditions for obtaining 82.55% maximum indigo carmine removal were pH 5.86, a current density of 13.31 mA/cm2, an indigo carmine concentration of 20.01 mg/L, and a time period of 115.80 min. Under optimum conditions for indigo carmine removal, the amount of sludge formed was 0.928 kg/m3, electrode consumption was 0.0305 kg/m3, and energy consumption was 7.461 kWh/m3. The operating cost was calculated as 0.79 US$/m3 under treatment conditions where optimum indigo carmine removal was achieved. In the second stage, G. pulex was exposed to treated and untreated synthetic indigo carmine solutions obtained from experiments according to optimum conditions in order to investigate the biochemical response for 24 and 96 h. Treatment efficiency was evaluated with superoxide dismutase (SOD), catalase (CAT), glutathione S-transferase (GST), and cytochrome P450 1A1 (CYP1A1) biomarkers. Results clearly showed that the electrocoagulation process was a simple and effective treatment method for the decolorization of wastewaters containing indigo carmine dye. The bioassays used in the present study provided good credibility for checking the detoxification of treated and untreated indigo carmine solutions.

Comparative evaluation of toxicological effects and recovery patterns in zebrafish (Danio rerio) after exposure to phosalone-based and cypermethrin-based pesticides

Korkmaz, Volkan; Güngördü, Abbas; Ozmen, Murat

This study evaluated the toxic effects and recovery patterns in zebrafish (Danio rerio) after exposure to phosalone-based (PBP) and cypermethrin-based (CBP) pesticides. Initially, the 96 h LC50 values of the pesticides were calculated as being 5.35 µg of active ingredient (AI) L−1 for CBP and 217 µg AI L−1 for PBP based on measured concentrations. Accordingly, experimental groups were exposed to three sublethal concentrations of pesticides for 96 h, separately, and then zebrafish were transferred to pesticide-free conditions for 10 and 20 days recovery periods. Biochemical markers were assessed including carboxylesterase (CaE), acetylcholinesterase (AChE), glutathione S-transferase (GST), lactate dehydrogenase, glutathione peroxidase, catalase, alanine and aspartate aminotransferase (ALT, AST) activities after the exposure and recovery periods. Also, the pesticide concentrations in test water were quantified by high-performance liquid chromatography (HPLC) analysis. Our results showed that AChE and CaE activities were significantly inhibited and GST was induced by both pesticides after 96 h exposure. For PBP exposure, the decreases for GST induction and CaE inhibition showed a partial recovery in pesticide-free conditions. However, the decreases in AChE activity for CBP exposure and insufficient increases in same enzyme activity for PBP exposure after 20 days in pesticide-free conditions indicated that the projected recovery period was not enough to the recovery of AChE activities and for the improvement of fish health.

Changes in antioxidant defense system in gills of Capoeta umbla caught from Uzuncayir Dam Lake, Turkey

Danabas, Durali; Yildirim, Nuran Cikcikoglu; Yildirim, Numan; Onal, Ayten Oztufekci; Uslu, Gulsad; Unlu, Erhan; Danabas, Seval; Ergin, Cemil; Tayhan, Nilgun

The aim of this work was to determine the seasonal changes in the activities of selected biomarkers in Capoeta umbla (Heckel, 1843) caught from Uzuncayir Dam Lake (Tunceli, Turkey) and to evaluate the effects of environmental factors on these activities. Fish were sampled on seasonal basis, and superoxide dismutase, catalase, glutathione peroxidase activities and levels of glutathione and malondialdehyde in gills were determined. Significant variations of oxidative stress biomarkers were observed between seasons and sites. The results of this study show that seasonal variations of oxidative stress responses and lipid peroxidation in gills of C. umbla are sensitive to the contaminants present in water of Uzuncayir Dam Lake and selected parameters are in valuable biomarkers for monitoring of water systems, since they give an early warning signal of effects of xenobiotics on aquatic organisms at molecular levels which help to prevent their effects at organismal level. Assessment of immunomodulator biomarkers (Tnf-α, Il-1β and Il-6) in liver of Capoeta umbla for biomonitoring of pollution in Uzuncayir Dam Lake (Tunceli, Turkey)

    Yildirim, N. C.; Danabas, S.

This study was aimed to monitor water pullution of Uzuncayir Dam Lake using the changes of IL-6, IL1 beta and TNF-alpha levels in Capoeta umbla (Heckel, 1843) liver tissue at ten stations in March and September 2011. In this study C. umbla (Heckel, 1843) was used as the indicator organism. Tumor necrosis factor-alpha (TNF-alpha), interlekuin1 beta (IL-1 beta) and interleukin-6 (IL-6) levels were determined in samples of the liver tissue by ELISA kit. The lowest mean IL-1 beta levels were found at station 6. The mean IL-1 beta was reached its maximum level at station 2. The difference between the mean levels of IL-6 was found to be significant (p<0.05) amoung stations. The IL-6 levels were significantly increased in September at stations 1, 2, 7 and 8 (p<0.01) compared to the values in March. The mean levels of TNF-alpha were found to be significant (p<0.05) amoung stations. The TNF-alpha levels significantly decreased in September at stations 1 and 9 (p<0.01). TNF-alpha, IL-1 beta and IL-6 levels in C. umbla can be used as early diagnostic indicators against adverse environmental events and useful and reliable bioindicators in determining the pollution of the aquatic ecosystem.
