Anti-oxidants are vital for protecting sperm and can mitigate the negative effects associated with cryopreservation. This study was conducted to evaluate the protective effects of adding anti-oxidants (vitamins and amino acids) to extender in sex-reversed rainbow trout (Oncorhynchus mykiss) sperm. The collected sperm was diluted at the ratio of 1: 5 by the extenders supplemented with different anti-oxidants including 1.00 mM of ascorbic acid and L-tryptophan, 2.00 mM of cysteine and α-tocopherol, and their combination. After dilution, the semen was aspirated into 0.50 mL straws, and the straws were placed on the tray, frozen for 10 min, and plunged into liquid nitrogen. Straws were thawed in a 30.00 ˚C water bath for 15 sec. The Sperm Class Analyzer System was used to evaluate sperm kinematics. The activity of anti-oxidant enzymes and lipid peroxidation were determined as oxidative stress indices. Our data indicated that the incorporation of anti-oxidants and amino acids increased sperm motility duration. The elevated activity of glutathione peroxidase, superoxide dismutase, and catalase in post-thaw samples indicates that the anti-oxidant system in sex-reversed rainbow trout sperm likely plays a crucial role in protecting membrane compounds from oxidation. In conclusion, the combination of 1.00 mM L-tryptophan and ascorbic acid to the extender media caused a prolonging effect in sperm motility after thawing and they have the potential to serve as effective agents for improving sperm cryosurvival.

The profile of fatty acids in the gametes of brook trout (Salvelinus fontinalis) ofduring the spawning period under culture conditions

Kocabas, Mehmet; Kutluyer-Kocabas, Filiz; Basçinar, Nadir; Özçiçek, Esin; Kirmizikaya-Özmen, Gorkem; Yilmaz, Okkes

The fertilization and subsequently embryo and larvae development are linked to gamete quality and FA (fatty acid) content is one of the specific factors affecting egg or sperm quality. Hence, FA profile of gametes (egg and sperm) was documented in brook trout (Salvelinus fontinalis) under cultivation. FAs were detected and measured using gas chromatography. These results indicated that a total of 26 and 22 FAs were determined in egg and sperm, respectively. The levels of PUFAs were determined in nearly identical quantities in both egg (47.57%) and sperm (47.53%). DHA (Docosahexaenoic acid), LA (Linoleic acid), EPA (Eicosapentaenoic acid) and ARA (Arachidonic acid) were predominant PUFAs for egg and sperm. Interestingly, vaccenic acid (C18:1 n-11) (24.07%) was major FAs in egg. In conclusion, the analysis of fatty acids in broodstock gametes contributes valuable insights into the dietary requirements necessary for effective aquaculture breeding programs.

Effect of Sperm Extender and Dilution Ratio on the Sperm Motility, Fertility, and Hatching Rates of Depik Fish Rasbora Tawarensis (Pisces: Cyprinidae) Eggs

Muchlisin, Zainal Abidin; Aldila, Dhea Febby; Sarah, Putri Inten; Eriani, Kartini; Hasri, Iwan; Fadli, Nur; Muhammadar, Abdullah Abbas; Handayani, Luvi Syafrida; Maulida, Siti; Siti-Azizah, Mohd Nor

There is no known artificial breeding method for depik fish Rasbora tawarensis. Sperm extender and the dilution ratio are crucial factors in the artificial breeding of fish. The current study aimed to assess the effect of several common fish sperm extenders and its dilution ratio on sperm characteristics and fertilization of depik fish eggs. Two series of experiments were performed to find the best extender and its dilution ratio. In the first experiment, five types of extenders were tested: Ringer's solution, physiological solution, coconut water, sugarcane water, and roomie water. The second experiment studied four dilution levels of sperm and extender (v/v): 1:20, 1:30, 1:40, and 1:50. Every treatment was conducted in four replicates and the experiments were conducted consequently so that the findings in the first experiment were applied in the second. The first experiment showed that Ringer's is the best extender for depik sperm with sperm motility, fertilization and hatching rates of 71.00%, 69.30%, and 53.66%, respectively; and the second experiment revealed that Ringer's at dilution ratio 1:40 gave higher sperm motility, fertilization, and hatching rates of 74.66, 70.33, and 59.00%, respectively. In conclusion, Ringer's solution was the most favorable extender for sperm which yielded best fertilization results at 1:40 dilution ratio (sperm:extender, v/v) in depik fish.

Combined effects of physicochemical variables (pH and salinity) on sperm motility: characterization of sperm motility in European sea bass Dicentrarchus labrax

    Ögretmen, F.; Inanan, B. E.; Kutluyer, F.

Gamete activation in fish is an important step in terms of artificial fertilization of oocytes, cryopreservation studies and other experimental manipulations. Salinity and pH differences in activation media affect to sperm motility and fertilizing ability. These experiments were therefore designed to investigate the combined effects of pH (range 5.0-9.0) and salinity (20, 30, 37, and 45 parts per thousand) of activation media on sperm motility of European sea bass Dicentrarchus labrax. The best results were obtained at salinity 37 parts per thousand and a pH of 9.0. Our results also demonstrated that non-progressive motility at salinity 45 parts per thousand was observed in the range of 5.0-9.0 pH. In conclusion, spermatozoa can be motile at a wide range of pH and salinity values although the percent of motile spermatozoa and motility duration are negatively affected by low pH values.
