Determining The Water Quality of Maruf Dam (Boyabat-Sinop)

    Mutlu, Ekrem; Kutlu, Banu

In this study, the preliminary findings obtained from 3 sampling points, which represent the whole, on Maruf Irrigation Dam, which is located in Boyabat district of Sinop province, for 12 months between September 2015 and August 2016 were examined. The parameters monitored are temperature, dissolved oxygen, pH, electrical conductivity, total hardness, total alkalinity, chemical oxygen demand, biological oxygen demand, and dissolved anions and cations (sodium, potassium, calcium, magnesium, ammonium, nitrate, phosphate, chloride, sulfate, and sulfite). Because of its low dissolved ionic matter content, Maruf Dam was characterized as an alkali dam with mid-hard water and low electrical conductivity. In terms of parameters A and B of Surface Water Quality Management Regulation, the dam is considered high-quality and unpolluted. Besides the irrigation purposes, for which the barrage was constructed, the dam can also be used for aquaculture, animal breeding, and farming needs.

EFFECT OF DIETARY FISH OIL REPLACEMENT WITH PLANT OILS ON GROWTH PERFORMANCE AND GENE EXPRESSION IN JUVENILE RAINBOW TROUT (ONCORHYNCHUS MYKISS)

Kutluyer, Filiz; Sirkecioglu, Ahmet Necdet; Aksakal, Ercument; Aksakal, Feyza Icoglu; Tunç, Abdullah; Günaydin, Esra

An eight-week feeding trial was conducted to evaluate the effects of total (100%) replacement of dietary fish oil with alternative lipid sources in juvenile rainbow trout. Six iso-nitrogenous and iso-lipidic experimental diets were formulated: CO (14%) -cod liver oil; SSO (14%) -safflower seed oil; SBO (14%) -soybean oil; LO (14%) -linseed oil; SBO (7%) + LO (7%) -a blend of soybean oil and linseed oil; and SSO (7%) + LO (7%) -a blend of safflower seed oil and linseed oil. Growth performance [specific growth rate (SGR), weight gain (WG), food conversion ratio (FCR) and survival rate (SR)], growth hormones [growth hormone (GH-I), insulin-like growth factor-I (IGF-I) and insulin-like growth factor-II (IGF-II)], immune transforming growth factor-beta (TGF-beta) and antioxidant [superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GR), catalase (CAT) and glutathione S-transferase (GST)] response, and heat shock protein 70 (HSP70) mRNA levels were determined in muscle and liver. Our data indicated that final weight, weight gain, FCR and SGR showed significant difference among the six dietary treatments (P<0.05) while there were no significant differences in survival rate between the rainbow trout from supplement fed groups and control group. HSP70 mRNA level expression in muscle was higher in fish fed SSO (P<0.05) while highest level in liver was obtained from fish fed SBO compared to the other treatments (P<0.05). There were no significant differences among treatments for TGF-beta mRNA expression level in muscle and liver. In conclusion, growth performance and expression levels of growth hormones, antioxidants, HSP70, except TGF-beta were affected by five separate lipid sources. In addition, LO positively increased growth performance of juvenile rainbow trout by means of preventing oxidative stress and HSP70 and, enhanced expression of growth hormone related gene.

Plant intelligence based metaheuristic optimization algorithms

    Akyol, Sinem; Alatas, Bilal

Classical optimization algorithms are insufficient in large scale combinatorial problems and in nonlinear problems. Hence, metaheuristic optimization algorithms have been proposed. General purpose metaheuristic methods are evaluated in nine different groups: biology-based, physics-based, social-based, music-based, chemical-based, sport-based, mathematics-based, swarm-based, and hybrid methods which are combinations of these. Studies on plants in recent years have showed that plants exhibit intelligent behaviors. Accordingly, it is thought that plants have nervous system. In this work, all of the algorithms and applications about plant intelligence have been firstly collected and searched. Information is given about plant intelligence algorithms such as Flower Pollination Algorithm, Invasive Weed Optimization, Paddy Field Algorithm, Root Mass Optimization Algorithm, Artificial Plant Optimization Algorithm, Sapling Growing up Algorithm, Photosynthetic Algorithm, Plant Growth Optimization, Root Growth Algorithm, Strawberry Algorithm as Plant Propagation Algorithm, Runner Root Algorithm, Path Planning Algorithm, and Rooted Tree Optimization.

Marine Fungi Against Aquaculture Pathogens and Induction of the Activity via Co-Culture

Özkaya, Ferhat Can; Peker, Zerife; Camas, Mustafa; Camas, Anil Sazak; Altunok, Muhammet

Ethyl acetate extracts from 70 strains of marine-derived fungi, isolated from sponges, were screened against four fish pathogenic bacteria. Sixteen of the fungal isolates exhibited antibacterial activity against at least one pathogen. All the strains showed antibacterial activity against Vagococcus salmoninarum. Three isolates showed strong inhibition against all test pathogens and they may therefore be remarkable as a broad-spectrum antimicrobial. Nine isolates showed antibacterial activity against Lactococcus garvieae, six were against Vibrio anguillarum, and four isolates inhibited the growth of Yersinia ruckeri. Co-culture of the fungus with the mix of the pathogen bacteria led to the induction of bioactivity in two isolates. Aspergillus iizukae produced strong and broad-spectrum antibacterial activity against pathogens with minimum inhibitory concentrations of 0.8-406 mu gmL(-1). The screening results confirmed the sponge associated fungi as a source of environmentally friendly antimicrobials that may contribute the sustainable development of aquaculture. In addition, co-culture technique is a promising strategy to increase antimicrobial activity that can stimulate the production of novel compounds, and increases the antimicrobial activities of marine derived fungi.

Anticancer and Cytotoxic Activities of [Cu(C6H16N2O2)2][Ni(CN)4] and [Cu(C6H16N2O2) Pd(CN)4] Cyanidometallate Compounds on HT29, HeLa, C6 and Vero Cell Lines

Aydin, Ali; Korkmaz, Sengul Aslan; Demir, Veysel; Tekin, Saban

Background: In cancer, apoptosis relevant proteins-such as CaM kinase, Bcl-2 or P53, topoisomerase I, cell migration feature and DNA/BSA-macromolecules represent significant targets for current chemotherapeutics. Objective: We recently reported two coordination compounds-[Cu(C6H16N2O2)(2)][Ni(CN)(4)] (1) and [Cu(C6H16 N2O2) Pd(CN)(4)] (2)-together with their IR spectra, magnetic properties, thermal analyses and crystal structures. Herein, we describe the ability of these complexes to induce apoptosis in relevant proteins and stimulate topoisomerase I activity, cell migration velocity and DNA/BSA binding properties. Method: The in vitro antiproliferative effects and cell toxicity of both compounds were investigated through pharmacological measurement techniques, and interactions between both compounds and CT-DNA/BSA were studied with UV-Vis spectroscopy and fluorescence spectroscopy. Results & Conclusion: Studies on cells revealed that 2 (i) demonstrated a high antiproliferative effect, which was higher toward HeLa and C6 cancer cells than toward healthy Vero cells; (ii) impaired the migration of HeLa cells; (iii) altered the P53-Bcl-2 ratio in favor of apoptosis; (iv) strongly bound to DNA/BSA macromolecules; and (v) inhibited human topoisomerase I and KpnI or BamHI restriction endonucleases. In conclusion, this preliminary information demonstrates that 2 may represent a promising antiproliferative agent and a potential candidate for a therapeutic approach against HeLa.

Evaluation of physical and mechanical characteristics of siderite concrete to be used as heavy-weight concrete

    Esen, Yuksel; Dogan, Zulfu Murat

The objective of this study is to investigate the usability of siderite aggregates mentioned above as a heavy-weight aggregate in the heavy-weight concrete production and their effects on physical properties of the heavy-weight concrete mixture. For this purpose, normal-weight aggregate content was decreased at the rate of 20%, 40%, 60%, 80% and 100% by volume of the concrete mixture prepared and mixtures were prepared by its substitution with siderite aggregate at the same rates. Based on the increase in siderite aggregate in the concrete mixture, a considerable increase in unit weight, compressive strength and tensile strength of concrete was obtained. Plate-shaped concrete specimens obtained from combined normal-weight and heavy-weight aggregates were exposed to radiation with different X-ray source (118 keV, 164 keV) and Co-60 (1250 keV) beam source at cekmece Nuclear Facility. It is clearly seen that the concrete mixture containing siderite aggregate improved 70% radiation absorption characteristics compared to that of the concrete mixture without siderite aggregate. SPATIAL AND TEMPORAL DISTRIBUTION OF PHYTOPLANKTON IN KARKAMIS DAM LAKE (SANLIURFA/TURKEY)

Sonmez, Feray; Kutlu, Banu; Sesli, Ahmet

The environmental factors that affect the seasonal change of the phytoplankton community and composition in Karkamis Dam Lake were investigated on samples taken from five stations between January 2015 and December 2015. It was found that the phytoplankton of Karkamis Dam Lake consisted of 85 taxa belonging to Bacillariophyta (56 taxa), Chlorophyta (15 taxa), Chrysophyta (1 taxon), Cyanophyta (10 taxa), Dinophyta (2 taxa) and Euglenophyta (1 taxon). Members of Bacillariophyta and Chrysophyta increased well during spring, summer and also at the end of fall whilst Chlorophyta grew well only in summer and fall. Cyanophyta became dominant at the end of spring and in summer. However Dinophyta was remarkably conspicuous in winter as well as in spring and fall in terms of prevalence. Excessive reproduction of Cyanobacteria, which is a characteristic aspect of mesotrophic lakes, was observed in Karkamis Dam Lake in both May and July. Results of physical and chemical measurements and analysis indicated that the lake was mesotrophic based on PCA and Carlson trophic state index. Phytoplankton composition was found to be affected by environmental factors such as temperature, pH, dissolved oxygen, phosphate and nitrate through the year. Using these data, a strong model was developed for sustainable management in Karkamis Dam Lake.

MONTHLY VARIABILITY OF OXIDANT AND ANTIOXIDANT BIOMARKERS OF FRESHWATER CRAYFISH (ASTACUS LEPTODACTYLUS ESCH., 1823)

Barim-Oz, Ozden; Yilmaz, Seval; Sahin, Hulya; Benzer, Fulya

In the present study was determined the effects of monthly variability on enzymatic antioxidant (superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px)), antioxidant (reduced glutathione (GSH)) and the oxidant status (nitric oxide (NO), lipid peroxidation (MDA)) in hepatopancreas, muscle, gills and gonad tissues of freshwater crayfish (A. leptodactylus) the monthly collected throughout the year from Keban Dam Lake of Turkey. Results indicate that variations in oxidants and antioxidants with relation to month were not only tissue specific but also were gender specific. The levels of oxidant parameters were generally higher in hepatopancreas in comparison to muscle, gills and gonad of crayfish. Moreover, in tissues were also determined the different oxidants and antioxidants responses in moulting, mating, breeding and egg laying periods.

Exploring thermodynamics and kinetic parameters of immobilized catalase enzyme via adsorption on krill clay

Nas, Mehmet Salih; Gür, Aycan; Gür, Tugba; Yönten, Vahap

The thermodynamic parameters and immobilization kinetics of catalase enzyme via adsorption onto krill clay were performed in aqueous solution in batch system. The adsorbent was characterized by X-ray fluorescence, scanning electron microscope and Brunauer-Emmett-Teller N-2 adsorption measurements. The effect of some parameters to the adsorption process such as contact time, stirring speed, initial enzyme concentration, initial ionic strength, temperature and pH were analyzed. Maximum adsorption capacity (q(m)) showed a great dependence on pH and initial enzyme concentration. It was found at pH 5.5, and initial enzyme concentration, 0.0075 g L-1 values. Pseudo-first-order, pseudo-second-order and intraparticle diffusion were used to fit the kinetics data on kinetic models. Furthermore, thermodynamic activation parameters such as enthalpy, entropy, Gibbs free energy and activation energy (Delta H degrees, Delta S degrees, Delta G degrees and E-a) which reveal that catalase enzyme adsorption occur spontaneously and in endothermic nature were determined. The obtained results confirmed that the applicability of krill clay is an efficient immobilizing supporter for catalase enzyme and krill clay material can be used as a very effective adsorbent in immobilization of this enzyme from aqueous solutions.

Effects of subchronic exposure to zinc nanoparticles on tissue accumulation, serum biochemistry, and histopathological changes in tilapia (Oreochromis niloticus)

Kaya, Hasan; Duysak, Muge; Akbulut, Mehmet; Yilmaz, Sevdan; Guerkan, Mert; Arslan, Zikri; Demir, Veysel; Ates, Mehmet

Zinc nanoparticles (ZnNPs) are among the least investigated NPs and thus their toxicological effects are not known. In this study, tilapia (Oreochromis niloticus) were exposed to 1 and 10 mg/L suspensions of small size (SS, 40-60 nm) and large size (LS, 80-100 nm) ZnNPs for 14 days under semi-static conditions. Total Zn levels in the intestine, liver, kidney, gill, muscle tissue, and brain were measured. Blood serum glucose (GLU), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and lactate dehydrogenase (LDH) were examined to elucidate the physiological disturbances induced by ZnNPs. Organ pathologies were examined for the gills, liver, and kidney to identify injuries associated with exposure. Significant accumulation was observed in the order of intestine, liver, kidney, and gills. Zn levels exhibited time- and concentration-dependent increase in the organs. Accumulation in kidney was also dependent on particle size; NPs SS-ZnNPs were trapped more effectively than LS-ZnNPs. No significant accumulation occurred in the brain (p>0.05) while Zn levels in muscle tissue increased only marginally (p0.05). Significant disturbances were noted in serum GOT and LDH (p<0.05). The GPT levels fluctuated and were not statistically different from those of controls (p>0.05). Histopathological tubular deformations and mononuclear cell infiltrations were observed in kidney sections. In addition, an increase in melano-macrophage aggregation intensity was identified on the 7th day in treatments exposed to LS-ZnNPs. Mononuclear cell infiltrations were identified in liver sections for all treatments. Both ZnNPs caused basal hyperplasia in gill sections. Fusions appeared in the gills after the 7th day in fish treated with 10 mg/L suspensions of SS-ZnNPs. In addition, separations in the secondary lamella epithelia were observed. The results indicated that exposure to ZnNPs could lead to disturbances in blood biochemistry and cause histopathological injuries in the tissues of O. niloticus. (c) 2016 Wiley Periodicals, Inc. Hazardous Waste Management in Turkey

Akkoyunlu, Atilla; Avsar, Yasar; Erguven, Gokhan Onder

The objective of this study is to analyze the time processes of hazardous waste management (HWM) in Turkey. There should be a holistic evaluation of a country's hazardous waste management program in terms of theoretical qualifications, best practice applications, and, of course, problems and their solutions. This paper offers a similar approach to evaluate HWM in compliance with the European Union under the laws of environmental applications. In 2010, 786,418 tons of hazardous waste (HW) were produced in Turkey. Of this, around 18% is sent to municipal garbage dumps, 0.6% is burned, and the remaining 81.4% is disposed of by other methods (such as controlled landfill or haphazard throwaways). Comparative morphological, karyological and palynological investigations of three endemic Centaurea L. species (Asteraceae) in Turkey

Hayta, Sukru; Tasar, Neslihan; Kiran, Yasar; Baser, Birol

Centaurea derderiifolia Wagenitz., Centaurea drabifolia Sm. subsp. floccosa (Boiss.) Wagenitz & Greuter and Centaurea kotschyi (Boiss. & Heldr.) Hayek var. floccosa (Boiss.) Wagenitz, which are endemic taxa from Turkey belonging to Centaurea sect. Cheirolepis, were subjected to morphological, karyological and palynological investigations. The somatic chromosome numbers, pollen morphology and detailed morphometric properties were examined. The somatic chromosome numbers were determined as 2n = 2x = 36 in C. derderiifolia, C. kotschyi var. floccosa, and C. drabifolia subsp. floccosa. Karyotype analysis indicated that chromosomes of these Centaurea taxa generally have median region (m), submedian region (sm) and rarely median point (M) karyotypes. The pollen morphology of these endemic taxa of the genus Centaurea was investigated by light and scanning electron microscopy. The pollen grains of the examined species are mainly isopolar and radially symmetric, subprolate, tricolporate. Tectum ornamentation is scabrate and scabrate-perforate, amb triangular. The pollen grains of all taxa studied are 3-zonocolporate. The colpus is usually long, acute at ends and with smooth membrane. Pollen grains belonging to Cheirolepis section reveal homogeneity in their characters.

The improved Bernoulli sub-equation function method is used in extracting some new exponential function solutions to the (2+1)-dimensional Broer-Kaup-Kupershmidt system. It is of vital effort to look for more solutions of the (2+1)-dimensional Broer-Kaup-Kupershmidt system, which are very helpful for coastal and civil engineers to apply the nonlinear water models in a harbor and coastal design. All the obtained solutions satisfied the (2+1)-dimensional Broer-Kaup-Kupershmidt system. The two-and three-dimensional shapes of all the obtained solutions in this paper are also presented. All the computations and the graphics plots in this study are carried out with the aid of the Wolfram Mathematica 9.

First microsatellite markers for Scaligeria lazica Boiss. (Apiaceae) by next-generation sequencing: population structure and genetic diversity analysis

Baldemir, Ayse; Topçu, Hayat; Paksoy, Mehmet Yavuz; Motalebipour, Elmira Ziya; Kafkas, Salih

The Apiaceae family includes a few agronomic and medicinal species, one of which is Scaligeria lazica Boiss. In this study, the genetic diversity of S. lazica was analyzed based on novel simple sequence repeat (SSR) markers using next-generation sequencing (NGS). A total of 15.17G clean Illumina data set was obtained and dinucleotide repeats were the most abundant repeats in S. lazica. Of the tested 150 SSR primer pairs, 139 ones produced amplification and 84 ones were polymorphic. Forty polymorphic SSR loci were used in genetic diversity analysis of 40 S. lazica accessions from four locations. A total of 264 alleles were amplified with an average of 6.6 alleles per locus. The polymorphism information content (PIC) was 0.60, while the observed homozygosity (Ho) and expected heterozygosity (He) values were 0.47 and 0.66, respectively. According to cluster and structure analysis, all accessions were grouped into four different clusters according to their collection sites. The SSR markers developed in this study can be tested for other Scaligeria species due to their high transferability and can be used for genetic studies in genus Scaligeria DC.

Occupational health and safety risk assessment in hospitals: A case study using two-stage fuzzy multi-criteria approach

Gul, Muhammet; Ak, M. Fatih; Guneri, Ali Fuat

Occupational Health and Safety (OHS) is a basis to reduce occupational accidents in an acceptable level, and it covers employee health, safety, and welfare in the workplaces. Hospitals as the largest employer group in healthcare industry in Turkey face major hazards categorized as chemical, biological, physical, ergonomic, and psychosocial risks. Although Turkey demonstrates rapid economic growth, OHS practices have not been fully put into practice, and necessary attention has not been paid to the health industry. For this reason, this paper aims to assess risks for health staff, contribute for planning of health services, and enhance regulations. A case study was carried out in a leading hospital in Turkey using a two-stage fuzzy multi-criteria approach, which provides more consistency in decision-making process and gives an appropriate final rank of hazard types. Fuzzy Analytic Hierarchy Process (FAHP) is used in weighing five risk parameters, which are severity, occurrence, undetectability, sensitivity to maintenance non-execution, and sensitivity to personal protective equipment (PPE) non-utilization. The fuzzy VIKOR (FVIKOR) approach is then applied for prioritization of hazard types in each department of the hospital. On conclusion of the hazard control hierarchy, measures are overtaken for the hazards, and areas open for improvement are presented.

Palaeoseismic history of the eastern part of the North Anatolian Fault (Erzincan, Turkey): Implications for the seismicity of the Yedisu seismic gap

Zabci, Cengiz; Akyüz, H. Serdar; Sançar, Taylan

The North Anatolian Fault showed a remarkable seismic activity especially between 1939 and 1999, when the westward migrating earthquake sequence created surface ruptures more than 1000 km, leaving unbroken only the Marmara segments, to the west, and the Yedisu Segment, to the east along the main strand of the fault. To understand the palaeoseismicity of the Yedisu Seismic Gap, we undertook trench investigations close to the village of Balaban SarA +/- kaya, on the western part of the Yedisu Segment. We found evidence for at least five surface faulting earthquakes, from which only two are correlated with the 18 July 1784 CE and 27 June 1583 CE historical events. Although the surface rupture of the 1784 CE was reported by other trench studies, the evidence of 1583 CE event is presented for the first time. In consideration with other historical earthquakes, affecting the region east of Erzincan, we suggest that this particular section of the North Anatolian Fault may be in a seismically quiescent period, following a cluster of earthquakes in its near history. In order to test this hypothesis, further studies are needed to increase our knowledge on the temporal and spatial seismic behaviour of the Yedisu Segment, which has potential to create an earthquake with M (w) similar to 7.2 in the near future.
