Electrochemical behaviour of cross-linker glutaraldehyde as a receptor for carbaryl biosensor

    Ipek, Yeliz

Glutaraldehyde (GA) is a famous cross-linker that is used for immobilization of enzyme or biologic material on electrochemical biosensor electrodes. It has an important role in pesticide biosensor studies. Because of the inhibitor effect of pesticides on acetylcholine esterase enzyme (AChE), it is generally immobilized on the electrode by cross-linking method. Glutaraldehyde is used as cross-linker agent to immobilize the enzyme on the electrode surface. Acetylcholine esterase activity is inhibited by pesticides or other neurotoxic compounds. This inhibition helps us to detect the pesticide residual. Although numerous electrochemical pesticide biosensors are based on generally the immobilization of the enzyme in the literature, an enzymeless biosensor based on GA has been developed in this study for the first time. Enzyme biosensors have high cost and needs special storage conditions. In this study, glutaraldehyde detected the carbaryl pesticide as sensitive as an acetylcholine esterase enzyme biosensor. The biosensor electrode was constructed with coating GA on glassy carbon electrode (GCE). Thus, it is a simple, low cost and practical method for pesticide detection in the water samples with low detection limit of 5x10(-9) mol L--(1).

Gul, Muhammet; Celik, Erkan; Aydin, Nezir; Gumus, Alev Taskin; Guneri, Ali Fuat

Multi criteria decision making (MCDM) is one of the research areas of operations research and management science which has widely studied by researchers and practitioners. It finds a compromise solution for evaluating and ranking alternatives from the best to the worst under conflicting criteria with respect to decision maker(s) preferences. In a compromise approach, the VlseKriterijumska Optimizacija I Kompromisno Resenje (VIKOR; that means multi-criteria optimization and compromise solution) continues to be applied satisfactorily across different application areas. This paper conducts a state-of-the-art literature review to categorize, analyze and interpret the current research on VIKOR applications. It also discusses the extensions of VIKOR applied in fuzzy environments. A total of 343 papers are classified into 13 different application areas and a number of sub-application areas. Furthermore, all papers are also categorized with respect to publication year, published journal, country of origin, application type (real case study vs empirical study), and version of fuzzy sets used. This comprehensive literature review provides an insight for researchers and practitioners on VIKOR applications in terms of showing current state and potential areas for future attempts to be focused in the future. Hardness removal from waters by using citric acid modified pine cone

Altundogan, Hamdi Soner; Topdemir, Aykut; Çakmak, Meltem; Bahar, Nurdan

In this study, an effective cation exchanger was developed from the pine cone by citric acid modification and its hardness removal properties from the waters were investigated. For this purpose, ground pine cone samples were subjected to a citric acid modification following NaOH saponification. Both of the raw (RPC) and modified pine cone (MPC) samples were subjected to standardized hardness removal tests by shaking with hard waters. These tests showed that citric acid modification significantly increases the cation exchange capacity of the pine cone. Most suitable size fraction of pine cone was determined as -16 + 30 mesh (600 mu m < x < 1200 mu m). Also, citric acid modified product obtained from this fraction was used in a continuous system to remove hardness from water. The results of this study showed that the MPC can be used for hardness removal from waters as a cheap, durable and environment -friendly material. Finally, regeneration experiments showed that the MPC can be used for hardness removal from water repeatedly by a simple HCl regeneration. (C) 2015 Taiwan Institute of Chemical Engineers. Kundmannia anatolica HUB.-MOR. and Kundmannia syriaca BOISS. are two endemic species of Apiaceae diversified in Turkey. In the present study, the essential oil composition and antimicrobial activity of these endemic species collected from Southeast Anatolia region of Turkey were investigated. Essential oils of aerial parts were obtained by hydro distillation method and analyzed by GC-MS/FID. Acetone, chloroform and methanol extracts of aerial parts of the plants were tested against seven bacteria and three yeasts by disc diffusion method for antimicrobial assays and minimum inhibition concentrations (MIC) were determined. Spathulenol, curdione, apritone, germacrene-d and- (-) globulol were the major components of K. anatolica. In the essential oil of K. syriaca, spathulenol, germacrene-d, bioallethrin, 12-norcyercene-B and beta methadol were determined as the main compounds. The most abundant compound of K anatolica and K. syriaca was spathulenol with 17.40%-19.53% ratio in the GC-MS and 19.7%, 24.99% ratio in GC-FID results, respectively. Aerial parts extracts of K. anatolica and K. syriaca exhibited remarkable antimicrobial activity to all tested microorganisms except for Candida albicans. (C) 2015 Elsevier B.V. All rights reserved.

The in vitro effect of Lambda-cyhalothrin on quality and antioxidant responses of rainbow trout Oncorhynchus mykiss spermatozoa

Kutluyer, Filiz; Erisir, Mine; Benzer, Fulya; Ögretmen, Fatih; Inanan, Burak Evren

There is little information in the scientific literature about effect of in vitro exposure of fish spermatozoa to pesticides. In vitro effect of Lambda-cyhalothrin (La) on sperm quality and oxidative stress has not been fully explored yet. The effects of LCT, which is a synthetic pyrethroid insecticide, on quality and oxidative stress of spermatozoa were investigated in vitro due to extensively use to control a wide range of insect pests in agriculture, public health, and homes and gardens. To explore the potential in vitro toxicity of La, fish spermatozoa were incubated with different concentrations of LCT (0.6, 1.2 and 2.4 mu g/L) for 2 h. Reduced glutathione (GSH), glutathione peroxidase (GSH-Px), catalase (CAT) and malondialdehyde (MDA) in spermatozoa were analyzed for determination of oxidant and antioxidant balance. Our results indicated that the percentage and duration of sperm motility significantly decreased with exposure to LCT. Activity of GSH-Px and MDA (P < 0.05) and GSH levels (P < 0.05) increased in a concentration-dependent manner while CAT activity decreased (P < 0.05). In conclusion, the oxidant and antioxidant status and sperm quality were affected by increasing concentrations of LCT. (C) 2015 Elsevier B.V. All rights reserved.

Use of acetylcholinesterase, glutathione S-transferase and cytochrome P450 1A1 in Capoeta umbla as biomarkers for monitoring of pollution in Uzuncayir Dam Lake (Tunceli, Turkey)

Yildirim, Nuran Cikcikogiu; Yildirim, Numan; Danabas, Durali; Danabas, Seual

Uzuncayir Dam Lake is one of the most important water bodies in Tunceli. It is polluted by domestic wastewaters together with natural contamination and pesticides, which have had cumulatively negative effects. This study analyzes the effects of pollution by using the fish species, Capoeta umbla as a sentinel species, with a biomarker approach. The approach comprehends a general biomarker of individual fish health, the condition factor and specific biomarkers of contaminant exposure such as glutathione S-transferase (GST) and cytochrome P450 1A1 (CYP1A1) and acetylcholinesterase (AChE) activity. The general water physico-chemical parameters were measured at each sampling sites during each fish-sampling period. Strong seasonal and locational variations were observed among selected markers. In conclusion, the multibiomarker approach used in the present study clearly revealed differences in the fish health among reference and potentially contaminated sites. The study shows that changes of biomarkers such as acetylcholinesterase, glutathione S-transferase and cytochrome P450 1A1 might be useful for the assessment of environmental contamination in the Uzuncayir Dam Lake (Tunceli, Turkey). Geo-environmental site investigation for Tunceli, Turkey municipal solid waste disposal site

Önal, Ayten Oztufekci; Demirbilek, Deniz; Demir, Veysel

The environmental and geological condition of a current solid waste disposal site in the city of Tunceli, Turkey was investigated. Improper geological structure and soil properties of this unsanitary solid waste storage site as well as leachate from this site caused significant pollution in air, soil, and surface water. Natural soil characteristics and geology of the site are presented in this paper. Petrographic properties of vulcanized rubber rocks composed of tuff and volcanic sandstone at the bottom of storage sites and the mineralogical composition of the base composed of these rocks were examined. Permeability of the natural soil and index properties were determined using soil mechanics experiments (moisture content, specific gravity, Atterberg limit tests, sieve analysis, hydrometer analysis, falling-head permeability test, and standard proctor test) conducted on disturbed samples collected from excavated sites and the relationship between mineralogical composition and soil hydraulic behavior was investigated. Natural soil samples of six different parts of the site indicated that, the soil class in four samples is silty sands, sand-silt mixtures, one is clayey sands, sand-clay mixtures and one is clayey silts with slight plasticity. The values obtained using permeability tests for subsoil samples vary from 10(-5) to 10(-8) cm/s. Low permeability (10(-7) and 10(-8) cm/s) at the south part of the site increases to 10(-5) cm/s at the natural soil of the North side. Extensively faulted and fissured volcanic sandstone levels at the site give potential to waste leachate to reach deep levels. The soil composition, hydraulic conductivity, topographic conditions, and other location criteria in the current solid waste disposal site showed that the current disposal site is not suitable for this purpose. Uniform Big Bang-Chaotic Big Crunch optimization

    Alatas, Bilal

This study proposes methods to improve the convergence of the novel optimization method, Big Bang-Big Crunch (BB-BC). Uniform population method has been used to generate uniformly distributed random points in the Big Bang phase. Chaos has been utilized to rapidly shrink those points to a single representative point via a center of mass in the Big Crunch phase. The proposed algorithm has been named as Uniform Big Bang-Chaotic Big Crunch (UBB-CBC). The performance of the UBB-CBC optimization algorithm demonstrates superiority over the BB-BC optimization for the benchmark functions. (C) 2010 Elsevier B.V. All rights reserved.
