Ellagic Acid Mitigates Lead (Pb)-Induced Toxicity in Cyprinus carpio: A Multi-Biomarker Assessment of Hematological, Immunological, and Oxidative Stress Responses Supported by Molecular Docking

Eroglu, Mucahit; Cakmak, Mehmet Nuri; Pala, Aysegul; Uslu, Harun; Yonar, Serpil Mise; Ispir, Unal; Orhan, Cemal; Yonar, Muhammet Enis

Lead (Pb) is a widespread environmental pollutant that induces systemic toxicity in aquatic organisms primarily through oxidative stress, hematological disruption, and immune dysfunction. This study investigated the protective effects of ellagic acid (EA) against Pb-induced toxicity in common carp (Cyprinus carpio) using a multi-biomarker approach and molecular docking. Fish were assigned to six experimental groups: control, EA-treated, Pb-I, Pb-I + EA, Pb-II, and Pb-II + EA. Fish in the Pb-I and Pb-II groups were exposed to 2.5 and 5 mg/L Pb, respectively, while EA was administered via diet at 100 mg/kg for 14 days. At the end of the exposure period, hematological indices, innate immune parameters, and oxidative stress biomarkers were assessed in blood, liver, kidney, and gill tissues. Pb exposure caused marked hematological impairment, suppressed immune responses, increased malondialdehyde levels, and disrupted antioxidant defense by reducing SOD, CAT, GSH-Px, and GSH levels while increasing GST activity. In contrast, dietary EA supplementation significantly mitigated Pb-induced alterations, improved hematological and immunological responses, reduced lipid peroxidation, restored antioxidant capacity, and normalized GST activity to control levels. Molecular docking analyses further showed that EA interacts with hemoglobin and immunoglobulin M, supporting its potential role in preserving oxygen transport and immune functions under Pb-induced stress. Overall, the findings demonstrate that Pb-induced toxicity involves coordinated disruption of redox homeostasis and immune function, whereas EA exerts a multi-target protective effect through biochemical and molecular mechanisms. This study provides mechanistic insight into chemical-biological interactions and supports the potential application of natural bioactive compounds in mitigating heavy metal-induced toxicity.

A New Metacognitive Confidence Calculation Approach Based on Large Language Models for Natural Language Explanations

Yilmazer Gundem, Merve; Sahin, Simay; Karakose, Mehmet

Traditional methods used to measure metacognitive confidence rely on superficial features such as word count and ambiguity and therefore fail to adequately reflect the level of metacognitive confidence in statements. In this study, in addition to traditional text-based confidence measures, we used a Large Language Model (LLM) to directly analyze confidence and its ratio in statements. LLM-based confidence scores were calculated on statements from the e-SNLI and CoS-E open-source datasets, and the prediction performance obtained with traditional features was compared. The results indicate that traditional surface-level text features explain the variance in LLM-generated confidence scores to a limited extent, and that LLM-based scores are significantly correlated with independent researcher evaluations. This study contributes to the literature by offering a scalable and reproducible analytical framework for measuring metacognitive confidence. It is believed that the findings can be directly applied, particularly in applications where trust is critical, such as automated software test scenario generation, decision support systems, and risk management. The use of LLMs offers a framework for the LLM-based analysis of potential metacognitive confidence signals in natural language explanations by examining the sense of confidence and certainty with which statements are presented, rather than their factual accuracy.

On Wijsman fρ-Statistical Convergence of Order α of Modulus Functions

    Turan, Gulcan Atici; Et, Mikail

In the present paper, we introduce and investigate the concepts of Wijsman f rho-statistical convergence of order alpha and Wijsman strong f rho-convergence of order alpha. These notions are defined as natural generalizations of classical statistical convergence and Wijsman convergence, incorporating the tools of modulus functions and natural density through the function f. We provide a detailed analysis of their structural properties, including inclusion relations, basic characterizations, and illustrative examples. Furthermore, we establish the inclusion relations between Wijsman f rho-statistical convergence and Wijsman strong f rho-convergence of order alpha, showing conditions under which one implies the other. These notions are different in general, while coinciding under certain restrictions on the function f, the parameter alpha, and the sequence rho. The results obtained not only extend some well-known findings in the literature but also open up new directions for further study in the theory of statistical convergence and its applications to analysis and approximation theory.

Babacan, Ebru Yüce; Demirpolat, Azize; Çakılcıoğlu, Uğur; Bagcı, Eyüp

The genus Opopanax W. Koch (Apiaceae) is represented by four species in Turkey. The composition of the essential oil of Opopanax genus members (Apiaceae) growing in Turkey was investigated in this study. GC-MS was used to analyze the composition of Opopanax essential oil samples that were taken from their natural environments. The Clevenger apparatus was used to hydrodistill the plant’s aerial parts, and the yields were determined to be between 0.2% v/w (for O. siifolius) and 0.4% (for O. hispidus, O. chironium, and O. persicus). The results and the chemical data provided some information and clues on the chemotaxonomy of the genus Opopanax. In this study, γ-elemene, butanoic acid octyl ester, and cylopropane were the main compounds identified in the essential oils of O. chironium, O. hispidus, and O. persicus. In particular, hexynyl n-valerate was most abundant in the essential oil of O. chironium, cyclopropane in that of O. hispidus, γ-elemene in that of O. persicus, and n-hexadecanoic acid/palmitic acid in that of O. siifolius. In a chemotaxonomic approach, the essential oil analysis of the Opopanax species revealed that these species conformed in a cluster analysis with their morphological classification. The constituents of the essential oils of all examined in the genus Opopanax were determined in this study, which is the most thorough one to date. This study provides new information about the composition of the essential oils of the investigated species.

Adding New Scientific Evidences on the Pharmaceutical Properties of Pelargonium quercetorum Agnew Extracts by Using In Vitro and In Silico Approaches

Chiavaroli, Annalisa; Libero, Maria Loreta; Di Simone, Simonetta Cristina; Acquaviva, Alessandra; Nilofar; Recinella, Lucia; Leone, Sheila; Brunetti, Luigi; Cicia, Donatella; Izzo, Angelo Antonio

Pelargonium quercetorum is a medicinal plant traditionally used for treating intestinal worms. In the present study, the chemical composition and bio-pharmacological properties of P. quercetorum extracts were investigated. Enzyme inhibition and scavenging/reducing properties of water, methanol, and ethyl acetate extracts were assayed. The extracts were also studied in an ex vivo experimental model of colon inflammation, and in this context the gene expression of cyclooxygenase-2 (COX-2) and tumor necrosis factor α (TNFα) were assayed. Additionally, in colon cancer HCT116 cells, the gene expression of transient receptor potential cation channel subfamily M (melastatin) member 8 (TRPM8), possibly involved in colon carcinogenesis, was conducted as well. The extracts showed a different qualitative and quantitative content of phytochemicals, with water and methanol extracts being richer in total phenols and flavonoids, among which are flavonol glycosides and hydroxycinnamic acids. This could explain, at least in part, the higher antioxidant effects shown by methanol and water extracts, compared with ethyl acetate extract. By contrast, the ethyl acetate was more effective as cytotoxic agent against colon cancer cells, and this could be related, albeit partially, to the content of thymol and to its putative ability to downregulate TRPM8 gene expression. Additionally, the ethyl acetate extract was effective in inhibiting the gene expression of COX-2 and TNFα in isolated colon tissue exposed to LPS. Overall, the present results support future studies for investigating protective effects against gut inflammatory diseases.

The study aimed at the metabolite profiling and evaluation of antioxidant and enzyme inhibitory properties of methanol extracts from flowers, leaves, and tubers of unexplored Eminium intortum (Banks & Sol.) Kuntze and E. spiculatum (Blume) Schott (Araceae). A total of 83 metabolites, including 19 phenolic acids, 46 flavonoids, 11 amino, and 7 fatty acids were identified by UHPLC-HRMS in the studied extracts for the first time. E. intortum flower and leaf extracts had the highest total phenolic and flavonoid contents (50.82 ± 0.71 mg GAE/g and 65.08 ± 0.38 RE/g, respectively). Significant radical scavenging activity (32.20 ± 1.26 and 54.34 ± 0.53 mg TE/g for DPPH and ABTS) and reducing power (88.27 ± 1.49 and 33.13 ± 0.68 mg TE/g for CUPRAC and FRAP) were observed in leaf extracts. E. intortum flowers showed the maximum anticholinesterase activity (2.72 ± 0.03 mg GALAE/g). E. spiculatum leaves and tubers exhibited the highest inhibition towards α-glucosidase (0.99 ± 0.02 ACAE/g) and tirosinase (50.73 ± 2.29 mg KAE/g), respectively. A multivariate analysis revealed that O-hydroxycinnamoylglycosyl-C-flavonoid glycosides mostly accounted for the discrimination of both species. Thus, E. intortum and E. spiculatum can be considered as potential candidates for designing functional ingredients in the pharmaceutical and nutraceutical industries.

A Fuzzy Hybrid MCDM Approach for Assessing the Emergency Department Performance during the COVID-19 Outbreak

    Ortíz-Barrios, Miguel; Jaramillo-Rueda, Natalia; Gul, Muhammet; Yucesan, Melih; Jiménez-Delgado, Genett; Alfaro-Saíz, Juan-José

The use of emergency departments (EDs) has increased during the COVID-19 outbreak, thereby evidencing the key role of these units in the overall response of healthcare systems to the current pandemic scenario. Nevertheless, several disruptions have emerged in the practical scenario including low throughput, overcrowding, and extended waiting times. Therefore, there is a need to develop strategies for upgrading the response of these units against the current pandemic. Given the above, this paper presents a hybrid fuzzy multicriteria decision-making model (MCDM) to evaluate the performance of EDs and create focused improvement interventions. First, the intuitionistic fuzzy analytic hierarchy process (IF-AHP) technique is used to estimate the relative priorities of criteria and sub-criteria considering uncertainty. Then, the intuitionistic fuzzy decision making trial and evaluation laboratory (IF-DEMATEL) is employed to calculate the interdependence and feedback between criteria and sub-criteria under uncertainty, Finally, the combined compromise solution (CoCoSo) is implemented to rank the EDs and detect their weaknesses to device suitable improvement plans. The aforementioned methodology was validated in three emergency centers in Turkey. The results revealed that the most important criterion in ED performance was ER facilities (14.4%), while Procedures and protocols evidenced the highest positive D + R value (18.239) among the dispatchers and is therefore deemed as the main generator within the performance network.

Temperature-Dependent Density and Magnetohydrodynamic Effects on Mixed Convective Heat Transfer along Magnetized Heated Plate in Thermally Stratified Medium Using Keller Box Simulation

Ullah, Zia; Akkurt, Nevzat; Alrihieli, Haifaa F.; Eldin, Sayed M.; Alqahtani, Aisha M.; Hussanan, Abid; Ashraf, Muhammad; Jabeen, Mah

The heat transmission properties along the non-magnetized geometries have been numerically obtainedby various researchers. These mechanisms are less interesting in engineering and industrial processes because of excessive heating. According to current studies, the surface is magnetized and the fluid is electrically conductive, which helps to lessen excessive surface heating. The main objective of the current analysis is to numerically compute the temperature-dependent density effect on magnetohydrodynamic convective heat-transfer phenomena of electrical-conductive fluid flow along the vertical magnetized and heated plate placed in a thermally stratified medium. For the purpose of numerical analysis, the theoretical process governing heat and magnetic intensity along a vertical magnetic plate is examined. By using suitable and well-known similarity transformations for integration, the non-linear coupled PDEs for the aforementioned electrical-conductive fluid flow mechanism are changed and subsequently converted into non-similar formulation. The Keller Box method is used to numerically integrate the final non-similar equations. The MATLAB software program plots the transformed algebraic equations graphically and quantitatively. The behavior of the physical quantities such asvelocity graph, magnetic field graph, and temperature plot along with their slopes that arerate of skin friction, the rate of heat transfer, and the rate of magnetic intensity for different parameters included in the flow model. The novelty of the current work is to compute the magneto-thermo analysis of electrically conducting flow along the vertical symmetric heated plate. First, we secure the numerical solution for steady part and then these results are used to find skin friction, heat transfer, and magnetic intensity. In the current work, the fluid becomes electrically conducing due to a magnetized surface which insulates heat during the mechanism and reduces the excessive heating. The results are excellent and accurate because they are satisfied by its given boundary conditions. Additionally, the current problems have a big impact on the production of polymer materials, glass fiber, petroleum, plastic films, polymer sheets, heat exchangers, catalytic reactors, and electronic devices.

Stability Analysis of Buoyancy Magneto Flow of Hybrid Nanofluid through a Stretchable/Shrinkable Vertical Sheet Induced by a Micropolar Fluid Subject to Nonlinear Heat Sink/Source

Khan, Umair; Zaib, Aurang; Ishak, Anuar; Alotaibi, Abeer M.; Eldin, Sayed M.; Akkurt, Nevzat; Waini, Iskandar; Madhukesh, Javali Kotresh

The utilization of hybrid nanofluids (HNs) to boost heat transfer is a promising area of study, and thus, numerous scientists, researchers, and academics have voiced their admiration and interest in this area. One of the main functions of nanofluids is their dynamic role in cooling small electrical devices such as microchips and associated gadgets. The major goal of this study is to perform an analysis of the buoyancy flow of a shrinking/stretching sheet, whilst considering the fascinating and practical uses of hybrid nanofluids. The influence of a nonlinear heat source/sink induced by a micropolar fluid is also inspected. Water-based alumina and copper nanoparticles are utilized to calculate the fine points of the fluid flow and the features of heat transfer. The governing equations are framed with acceptable assumptions and the required similarity transformations are used to turn the set of partial differential equations into ordinary differential equations. The bvp4c technique is used to solve the simplified equations. Dual solutions are presented for certain values of stretching/shrinking parameters as well as the mixed convective parameter. In addition, the shear stress coefficient in the first-branch solution (FBS) escalates and decelerates for the second-branch solution (SBS) with the superior impact of the magnetic parameter, the mass transpiration parameter, and the solid nanoparticles volume fraction, while the contrary behavior is seen in both (FB and SB) solutions for the larger values of the material parameter.

Significance of Free Convection Flow over an Oscillating Inclined Plate Induced by Nanofluid with Porous Medium: The Case of the Prabhakar Fractional Approach

Raza, Ali; Khan, Umair; Eldin, Sayed M.; Alotaibi, Abeer M.; Elattar, Samia; Prasannakumara, Ballajja C.; Akkurt, Nevzat; Abed, Ahmed M.

Given the importance and use of electrically conducted nanofluids, this work aims to examine an engine-oil-based nanofluid including various nanoparticles. In the current study, a fractional model for inspecting the thermal aspect of a Brinkman-type nanofluid, composed of (molybdenum disulfide (MOS2) and graphene oxide (GO) nanoparticles flows on an oscillating infinite inclined plate, which characterizes an asymmetrical fluid flow, heat, and mass transfer. Furthermore, the Newtonian heating effect, magnetic field, and slip boundary conditions were taken into account. The objectives for implementing the Prabhakar-like fractional model are justified because this fractional algorithm has contemporary definitions with no singularity restrictions. Furthermore, the guided fractional model was solved using the Laplace transform and several inverse methods. The obtained symmetrical solutions have been visually analyzed to investigate the physics of several relevant flow parameters on the governed equations. Some exceptional cases for the momentum field are compiled to see the physical analysis of the flowing fluid symmetry. The results show that the thermal enhancement can be progressively improved with the interaction of the molybdenum disulfide-engine oil-based nanofluid suspension, rather than with the graphene oxide mixed nanoparticle fluid. Furthermore, the temperature and momentum profiles enhance due to the factional parameters for molybdenum disulfide and the graphene oxide-engine oil-based nanofluid suspension. This study’s graphical and numerical comparison with the existing literature has shown a very close resemblance with the present work, which provides confidence that the unavailable results are accurate. The results show that an increase improved the heat transmission in the solid nanoparticle volume fractions. In addition, the increment in the mass and heat transfer was analyzed in the numerical evaluation, while the shear stress was enhanced with the enhancement in the Prabhakar fractional parameter α.

Ozturk, Refiye Beyza; Zengin, Gokhan; Sinan, Kouadio Ibrahime; Montesano, Domenico; Zheleva-Dimitrova, Dimitrina; Gevrenova, Reneta; Uba, Abdullahi Ibrahim; Çakılcıoğlu, Uğur; Kaplan, Alevcan; Jugreet, Sharmeen

The bioactive content, antioxidant properties, and enzyme inhibition properties of extracts of Alcea fasciculiflora from Turkey prepared with different solvents (water, methanol, ethyl acetate) and extraction methods (maceration, soxhlet, homogenizer assisted extraction, and ultrasound assisted extraction) were examined in this study. UHPLC-HRMS analysis detected or annotated a total of 50 compounds in A. fasciculiflora extracts, including 18 hydroxybenzoic and hydroxycinnamic acids, 7 Hexaric acids, 7 Coumarins, 15 Flavonoids, and 3 hydroxycinnamic acid amides. The extracts had phenolic and flavonoid levels ranging from 14.25 to 24.87 mg GAE/g and 1.68 to 25.26 mg RE/g, respectively, in the analysis. Both DPPH and ABTS tests revealed radical scavenging capabilities (between 2.63 and 35.33 mg TE/g and between 13.46 and 76.27 mg TE/g, respectively). The extracts had reducing properties (CUPRAC: 40.38–78 TE/g and FRAP: 17.51–42.58 TE/g). The extracts showed metal chelating activity (18.28–46.71 mg EDTAE/g) as well as total antioxidant capacity (phosphomolybdenum test) (0.90–2.12 mmol TE/g). DPPH, ABTS, FRAP, and metal chelating tests indicated the water extracts to be the best antioxidants, while the ethyl acetate extracts had the highest overall antioxidant capacity regardless of the extraction technique. Furthermore, anti-acetylcholinesterase activity was identified in all extracts (0.17–2.80 mg GALAE/g). The water extracts and the ultrasound-assisted ethyl acetate extract were inert against butyrylcholinesterase, but the other extracts showed anti-butyrylcholinesterase activity (1.17–5.80 mg GALAE/g). Tyrosine inhibitory action was identified in all extracts (1.79–58.93 mg KAE/g), with the most effective methanolic extracts. Only the ethyl acetate and methanolic extracts produced by maceration and homogenizer aided extraction showed glucosidase inhibition (0.11–1.11 mmol ACAE/g). These findings showed the overall bioactivity of the different extracts of A. fasciculiflora and provided an overview of the combination of solvent type and extraction method that could yield bioactive profile and pharmacological properties of interest and hence, could be a useful reference for future studies on this species.

This study presents the evaluation of biological activities and chemical profiling of Oenanthe aquatica (L.) Poir. and Oenanthe silaifolia M. Bieb. The phytochemical profile, antioxidant, enzyme inhibitory, cytotoxic and antiviral activities of the methanolic and aqueous extracts were investigated. The aqueous extract of O. aquatica possessing the highest content of phenolics (60.85 mg gallic acid equivalent/g extract), also exhibited the strongest radical scavenging potential against 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (79.46 and 148.66 mg Trolox equivalent/g extract, respectively), the highest reducing ability (207.59 and 107.27 mg Trolox equivalent/g extract, for cupric reducing antioxidant capacity and ferric reducing antioxidant activity, respectively), metal chelating potential (33.91 mg ethylenediaminetetraacetic acid equivalent/g extract) and total antioxidant ability (1.60 mmol Trolox equivalent/g extract). Liquid chromatography-electrospray ionization-quadrupole time-of-flight-mass spectrometry (LC-ESI-QTOF-MS/MS) permitted tentative identification of compounds from simple organic acids, phenolic acids, coumarins, flavonoids and their glycosides in O. aquatica and O. silaifolia extracts. The methanolic extract of O. aquatica substantially depressed acetylcholinesterase (3.67 mg galantamine equivalent/g extract), tyrosinase (126.66 mg kojic acid equivalent/g extract), and α-amylase (0.83 mmol acarbose equivalent/g extract) enzymes. The methanolic extract of O. silaifolia showed highest enzymatic inhibitory property against butyrylcholinesterase, and its aqueous extract depressed α-glucosidase activity (0.26 mmol acarbose equivalent/g extract). All tested extracts exerted selective toxicity towards cancer cell lines, and the highest anticancer potential was found for O. aquatica aqueous extract on FaDu and HeLa cells with CC50 of 57.36 and 47.16 µg/mL, respectively. Significant antiviral activity against HSV-1 (HHV-1) was found for both aqueous extracts in concentrations of 1000 µg/mL, which inhibited the HSV-1 cytopathic effect (CPE) in virus infected VERO cells and reduced the virus infective titer by more than 3 log (logCCID50 /mL). This study has produced critical scientific data on O. aquatica and O. silaifolia, which are potential contenders for the development of novel phyto-pharmaceuticals.

Inverted Solar Stills: A Comprehensive Review of Designs, Mathematical Models, Performance, and Modern Combinations

Hussein, Ahmed Kadhim; Rashid, Farhan Lafta; Abed, Azher M.; Al-Khaleel, Mohammad; Togun, Hussein; Ali, Bagh; Akkurt, Nevzat; Malekshah, Emad Hasani; Biswal, Uddhaba; Al-Obaidi, Mudhar A.

Fresh water is scarce, making it a worldwide issue. In order to address global freshwater demand, a filtration technique is needed. Solar distillation (SD) that purifies brackish and sub-terranean water has been proven to be a promising green technology. It produces distilled water which can be used as potable water for drinking and other purposes. The designs, operations, and configurations of several inverted solar stills are thoroughly examined in this article. All techniques for solar water distillation to separate saltwater from freshwater have the same fundamental idea. It has been demonstrated that for a specified mass of water in the top basin, the daily output of an inverted absorber double-basin solar still (IASS) grows with increasing water depth in the bottom basin. Nevertheless, as the water depth in the bottom basin rises from 1 cm to 5 cm, the output of an inverted absorber double-basin solar system falls by about 27%. At water depths of 4, 6, and 8 cm, the daily yields from inverted absorber solar stills paired with a refrigeration cycle (RIASS) were 6.4, 10.08, and 9.5 L/day, respectively. The results also showed a substantial rise in water temperature in the inverted absorber solar still as a result of lower bottom heat loss and larger absorptivity. In this study, the emphasis on energy, efficiency, and environmental concerns remains with the modified multi-wick basin-type inverted absorber solar panel. The performance of the inverted trickling solar still was assessed with a 60 ppm average salinity under various sun radiation, feed water flow rate, and existence conditions for natural convection. It was found that the still could produce 2.55 L/day m2 of fresh water, a 15% increase. Brackish water with a salinity of 6000 ppm was used to test the still in November at tilt angles of 47◦ and 32◦ . Condensate production at the specified angles was 2.8 and 2 L/d, respectively. A total output of 6.907, 5.681, and 4.650 kg/m2 /day was also generated using the modified multi-wick basin-type inverted absorber solar still (MMWBIASS) at water depths of 1, 2, and 3 cm, respectively. With black cotton wicks, the MMWBIASS had a total thermal efficiency of 34.04%, 28.17%, and 23.61%.

Experimental and Numerical Analysis of Forced Convection in a Horizontal Tube Partially Filled with a Porous Medium under Local Thermal Equilibrium Conditions

Amoli, Behzad Siavash; Ajarostaghi, Seyed Soheil Mousavi; Saffar-Avval, Majid; Abardeh, Reza Hosseini; Akkurt, Nevzat

The objective of the present work is to analyze experimentally and numerically the laminar forced convection flow in a horizontal pipe partially filled with a porous medium under constant heat flux and to study the influence of the eccentricity of the porous medium on the results. In a numerical analysis, the governing equations are solved in three dimensions. To simplify the grid generation and the satisfaction of the boundary conditions, conformal mapping is applied to convert the cross-section of the tube in the fluid domain (space between two eccentric circles) into a rectangle, and the equations are solved in a computational domain in this domain. The Darcy–Brinkman–Forchheimer model is applied to simulate the hydrodynamic behavior of the flow in the porous region. Thermal equilibrium between solid and fluid is assumed for the energy equation. A FORTRAN program was developed to solve the equations using the finite volume method and the SIMPLE algorithm. Velocity profile, pressure drop and average Nusselt number are studied in a wide range of Darcy numbers, thickness of porous mediums and eccentricities. The results show that the eccentricity of the porous material reduces the heat transfer coefficient and the pressure drop simultaneously; of course, the reduction in the heat transfer coefficient is less noticeable when the thickness of the porous medium is smaller. For example, at RP = 0.5, when the eccentricity of the porous medium increases up to E = 0.4, the average Nusselt number decreases by 66%, and this reduction for a smaller porous thickness decreases to 11%. The maximum pressure drop reduction for Da = 10−5 and E = 0.4 is 25%.

Antioxidant and Enzyme Inhibitory Properties, and HPLC–MS/MS Profiles of Different Extracts of Arabis carduchorum Boiss.: An Endemic Plant to Turkey

Uba, Abdullahi Ibrahim; Zengin, Gokhan; Montesano, Domenico; Cakilcioglu, Ugur; Selvi, Selami; Ulusan, Musa Denizhan; Caprioli, Giovanni; Sagratini, Gianni; Angeloni, Simone; Jugreet, Sharmeen

In this study, six extracts of Arabis carduchorum prepared from solvents of varying polarity (water, methanol, ethyl acetate, dichloromethane, and n‐hexane) were investigated for their chemical profiles and total phenolic and flavonoid contents (TPC/TFC) using HPLC–MS/MS and spectrophotometric/colorimetric assays, respectively, along with the assessment of their antioxidant and enzyme inhibitory effects. In general, the polar extracts (methanol, water, and infusion) showed higher TPC/TFC than the other extracts (range:13.73–26.62 mg GAE/g; 2.66–52.95 mg RE/g, respectively). The total antioxidant capacity of the polar extracts ranged from 0.90–1.85 mmol TE/g in phosphomolybdenum assay, while the free radical scavenging potential ranged from 4.36–32.13 mg TE/g and 12.58–67.73 mg TE/g in DPPH and ABTS assays, respectively. While the water and infuse extract inhibited only butyrylcholinesterase (2.68 and 2.39 mg GALAE/g, respectively), the other extracts were found to inhibit both acetyl‐ and butyryl‐cholinesterases (0.96–2.73 mg GALAE/g and 1.86–5.11 mg GALAE/g, respectively). On the contrary, all extracts exhibited anti‐tyrosinase (40.24– 59.22 mg KAE/g) and anti‐amylase (0.09–0.63 mmol ACAE/g) activities. Overall, this study highlighted the effect of solvent polarity on the extraction of bioactive compounds and the biological properties of the extracts of A. carduchorum; hence, the results obtained could be used to guide optimum extraction in future studies.

Magnesium picolinate improves bone formation by regulation of rank/rankl/opg and bmp-2/runx2 signaling pathways in high-fat fed rats

Sahin, Emre; Orhan, Cemal; Balci, Tansel Ansal; Erten, Fusun; Sahin, Kazim

Magnesium (Mg) deficiency may affect bone metabolism by increasing osteoclasts, de-creasing osteoblasts, promoting inflammation/oxidative stress, and result in subsequent bone loss. The objective of the present study was to identify the molecular mechanism underlying the bone protective effect of different forms of Mg (inorganic magnesium oxide (MgO) versus organic magnesium picolinate (MgPic) compound) in rats fed with a high-fat diet (HFD). Forty-two Wistar albino male rats were divided into six group (n = 7): (i) control, (ii) MgO, (iii) MgPic, (iv) HFD, (v) HFD + MgO, and (vi) HFD + MgPic. Bone mineral density (BMD) increased in the Mg supplemented groups, especially MgPic, as compared with the HFD group (p < 0.001). As compared with the HFD + MgO group, the HFD + MgPic group had higher bone P (p < 0.05) and Mg levels (p < 0.001). In addition, as compared to MgO, MgPic improved bone formation by increasing the levels of osteo-genetic proteins (COL1A1 (p < 0.001), BMP2 (p < 0.001), Runx2 (p < 0.001), OPG (p < 0.05), and OCN (p < 0.001), IGF-1 (p < 0.001)), while prevented bone resorption by reducing the levels of RANK and RANKL (p < 0.001). In conclusion, the present data showed that the MgPic could increase osteogenic protein levels in bone more effectively than MgO, prevent bone loss, and contribute to bone formation in HFD rats.

Curcumin positively affects performance during exercise and subsequent recovery. However, curcumin has limited bioavailability unless consumed in larger doses. In the current study, we examined the impact of a new formulation of curcumin, Next‐Generation Ultrasol Curcumin (NGUC), which is relatively more bioavailable than natural curcumin on exhaustion time, grip strength, muscle damage parameters, and serum and muscle proteins. A total of 28 rats were randomly grouped as control (C, non‐supplemented), exercise (E, non‐supplemented), E+NGUC100 (supplemented with 100 mg/kg BW NGUC), and E+NGUC200 (supplemented with 200 mg/kg NGUC). Grip strength and exhaustion time were increased with NGUC supplementation (p < 0.0001). Creatine kinase (CK), lactate dehydrogenase (LDH), lactic acid (LA), myoglobin, malondialdehyde (MDA) concentrations were reduced in serum, and muscle tissue in NGUC supplemented groups (p < 0.05). In contrast, NGUC supplementation elevated the antioxidant enzyme levels compared to the non‐supplemented exercise group (p < 0.01). Additionally, inflammatory cytokines were inhibited with NGUC administration (p < 0.05). NGUC decreased PGC‐1α, p‐4E‐BP1, p‐mTOR, MAFbx, and MuRF1 proteins in muscle tissue (p < 0.05). These results indicate that NGUC boosts exercise performance while reducing muscle damage by targeting antioxidant, anti‐inflammatory, and muscle mass regulatory pathways.

A deep learning model for automated sleep stages classification using PSG signals

Yildirim, Ozal; Baloglu, Ulas Baran; Acharya, U Rajendra

Sleep disorder is a symptom of many neurological diseases that may significantly affect the quality of daily life. Traditional methods are time-consuming and involve the manual scoring of polysomnogram (PSG) signals obtained in a laboratory environment. However, the automated monitoring of sleep stages can help detect neurological disorders accurately as well. In this study, a flexible deep learning model is proposed using raw PSG signals. A one-dimensional convolutional neural network (1D-CNN) is developed using electroencephalogram (EEG) and electrooculogram (EOG) signals for the classification of sleep stages. The performance of the system is evaluated using two public databases (sleep-edf and sleep-edfx). The developed model yielded the highest accuracies of 98.06%, 94.64%, 92.36%, 91.22%, and 91.00% for two to six sleep classes, respectively, using the sleep-edf database. Further, the proposed model obtained the highest accuracies of 97.62%, 94.34%, 92.33%, 90.98%, and 89.54%, respectively for the same two to six sleep classes using the sleep-edfx dataset. The developed deep learning model is ready for clinical usage, and can be tested with big PSG data.

Application of Computational Intelligence Methods for the Automated Identification of Paper-Ink Samples Based on LIBS

Rzecki, Krzysztof; Sośnicki, Tomasz; Baran, Mateusz; Niedźwiecki, Michał; Król, Małgorzata; Łojewski, Tomasz; Acharya, U. Rajendra; Yildirim, Özal; Pławiak, Paweł

Laser-induced breakdown spectroscopy (LIBS) is an important analysis technique with applications in many industrial branches and fields of scientific research. Nowadays, the advantages of LIBS are impaired by the main drawback in the interpretation of obtained spectra and identification of observed spectral lines. This procedure is highly time-consuming since it is essentially based on the comparison of lines present in the spectrum with the literature database. This paper proposes the use of various computational intelligence methods to develop a reliable and fast classification of quasi-destructively acquired LIBS spectra into a set of predefined classes. We focus on a specific problem of classification of paper-ink samples into 30 separate, predefined classes. For each of 30 classes (10 pens of each of 5 ink types combined with 10 sheets of 5 paper types plus empty pages), 100 LIBS spectra are collected. Four variants of preprocessing, seven classifiers (decision trees, random forest, k-nearest neighbor, support vector machine, probabilistic neural network, multi-layer perceptron, and generalized regression neural network), 5-fold stratified cross-validation, and a test on an independent set (for methods evaluation) scenarios are employed. Our developed system yielded an accuracy of 99.08%, obtained using the random forest classifier. Our results clearly demonstrates that machine learning methods can be used to identify the paper-ink samples based on LIBS reliably at a faster rate.

Big social network data and sustainable economic development

    Can, Umit; Alatas, Bilal

New information technologies have led to the rapid and effective growth of social networks. The amount of data produced by social networks has increased the value of the big data concept, which is one of the popular current phenomena. The immediate or unpredictable effects of a wide array of economic activities on large masses and the reactions to them can be measured by using social media platforms and big data methods. Thus, it would be extremely beneficial to analyze the harmful environmental and social impacts that are caused by unsustainable business applications. As social networks and big data are popular realms currently, their efficient use would be an important factor in sustainable economic development. Accurate analysis of people's consumption habits and economic tendencies would provide significant advantages to companies. Moreover, unknown consumption factors that affect the economic preferences of individuals can be discovered and economic efficiency can be increased. This study shows that the numerous solution opportunities that are provided by social networks and big data have become significant tools in dynamic policy creation by companies and states, in solving problems related to women's rights, the environment, and health.
