Docosahexaenoic acid (DHA) was the major polyunsaturated fatty acid (PUFA) in the sperm plasma membrane, and the levels of PUFAs decreased in treated groups during the cryopreservation process. The supplementation of TT (Tribulus terrestris) to Cyprinus carpio sperm at 200 µg∙L-1 positively affected the maintenance of motility parameters and FAs levels after the freeze-thaw cycle, and TT can be used as an herbal medicine instead of synthetic pharmaceuticals to improve the productive performance in aquaculture.

Kaya, Seyma Ozer; Ozcan, Sinan; Kaya, Emre; Gur, Seyfettin; Koprucu, Kenan; Sonmez, Mustafa; Turk, Gaffari; Erisir, Mine

Background: This study was conducted to determine the effect of various fatty acid ratios added to diets of rainbow trout (Oncorhynchus mykiss, Walbaum) on some sperm parameters and parameters related to arginase activity. Methods: A total of 99 male rainbow trout were used in this study, with the fish averaging 3 years old. Semen was collected by abdominal massage. Four different experimental groups were created in the study: a control group without n-3 series fatty acids added to their diets, and groups with n-3 series fatty acids added at ratios of 1% (D1), 2% (D2), and 3% (D3), respectively. Results: While no significant difference in semen volume was observed between the D1 and D3 groups and the control group, semen volume was significantly higher in the D2 group. Compared with the control group, the time required for sperm motility to decline to both 50% and 0% was significantly prolonged in the D1 and D3 groups. Seminal plasma arginase activity did not differ significantly in the D2 and D3 groups compared with the control group but was significantly higher in the D1 group. Conclusions: Consequently, considering the doses used in the study, fatty acid supplementation was found to lead to changes in sperm quality parameters, specifically volume (only 2%) and arginase activity (only 1%). Given that the rate of decrease in sperm motility was later in groups D1 and D2, it was concluded that the added fatty acids may be beneficial.

Kocabaş, Filiz Kutluyer; Kizak, Volkan; Özçiçek, Esin; Kocabaş, Mehmet; Yilmaz, Ökkeş

Freshwater snails are a nutritious food source, providing vital amino acids, minerals, and protein with low lipid levels. Hence, the adaptation ability, growth rate, fatty acid (FA) profile, and shell content of Viviparus contectus were determined. Biometric measurements were recorded for 6 months. Over the 6-month cultivation period, specific growth rates were recorded as 0.32% · day-1 for shell length and 0.92% · day-1 for body weight. The FA profile of the freshwater snail included a total of 32 compounds with saturated, monounsaturated, and polyunsaturated FAs represented by 13, 10, and 9 types, respectively. Monounsaturated fatty acids (MUFAs) exhibited the highest concentration, followed by saturated fatty acids (SFAs) s at 35.14% and polyunsaturated fatty acids (PUFAs) at 14.57%. Stearic acid (C18:0), erucic acid (C22:1 n-9), and arachidonic acid (C20:4 n-6) were determined as the most abundant FAs within the saturated, monounsaturated, and polyunsaturated FAs classes, respectively. The energy-dispersive X-ray spectroscopy spectra analysis revealed that the shell powder composition consisted of the elements, such as carbon, oxygen, calcium, iron, manganese, and silicium. Overall, V. contectus showed a clear increase in growth over the cultivation period, and future studies are needed to obtain the best culture conditions and intensive production of the species.

Effect of Ferula sp. on sperm cryotolerance, quality, and fatty acid composition in common carp (Cyprinus carpio)

Ölçülü, Abdullatif; Özçiçek, Esin; Kutluyer Kocabaş, Filiz; Kızak, Volkan; Kocabaş, Mehmet; Yılmaz, Ökkeş

Ferula sp. is a perennial herbaceous plant and is used in traditional medicine owing to their antimicrobial, antiviral, aphrodisiac and spermatogenic activities in the world. Hence, Ferula sp. was used in the present study for cryopreservation protocol as a herbal supplement. The impact of different concentrations [0 mM (Control), 30 µg L−1, 60 µg L−1, 90 µg L−1] of Ferula sp. extract on the quality and fatty acid (FAs) composition was assessed in fresh and cryopreserved sperm of Cyprinus carpio. Optimal supplementation of Ferula sp. at a concentration of 60 µg L−1 in the freezing extender significantly improved post-thawed sperm quality of C. carpio. Sperm motility parameters were assessed using the sperm class analyzer system. The fatty acid (FAs) content in sperm was analyzed using gas chromatography. The results revealed that the increases in motility rate (80.0 ± 3.54%) at 60 µg L−1 were increased significantly (p < 0.05). Significant differences in sperm parameters [(VCL: curvilinear velocity, VSL: straight line velocity, VAP: average path velocity, and BCF: beat cross frequency)] were observed between the different treatments (p < 0.05). The Monounsaturated fatty acids (MUFAs) showed a slight increase in comparison to fresh spermatozoa during cryopreservation. DHA (Docosahexaenoic acid), LA (Linoleic acid) and ARA (Arachidonic acid) were identified as the major PUFAs (polyunsaturated fatty acid) in the sperm plasma membrane, and however the total PUFAs reduced all treated compared to fresh sperm samples after post-thawing. Overall, the supplementation with Ferula sp. to extender at a concentration of 60 µg L−1 positively influenced the motility parameters and FAs levels during cryopreservation and, Ferula sp. may act as a promising protective substance for the conservation of valuable fish genetic resources.

Identification and Quantification of Fatty Acid Profile of Muscle and Gametes in Threatened Abant Trout (Salmo abanticus) from Wild

Özçiçek, Esin; Kocabaş, Filiz Kutluyer; Kocabaş, Mehmet; Yilmaz, Ökkeş

Abstract: As an important challenge restricting hatchery production, gamete quality and subsequent larvae and juvenile are influenced by broodstock nutritional status (i.e., protein, essential amino acids, fatty acid profile and minerals). Hence, the fatty acid (FA) profiles of muscle and gametes in threatened Abant trout (Salmo abanticus) from wild was elucidated and quantitated using the gas chromatography. Our results indicated that the saturated fatty acids (SFAs) in muscle and sperm (38.00 and 60.98%) of S. abanticus occurred in higher quantities than unsaturated fatty acids. The egg of S. abanticus in the wild is characterized by high (mostly n-6 HUFA, 33.59%). Tricosanoic acid (C23:0) and docosahexaenoic acid (DHA, C22:6 n-3) were not determined in sperm of S. abanticus. Overall, the current study provides FA composition in the reproduction cells and muscle of mature S. abanticus and the gametes and muscle had similar general FA profiles with the dominance of C18:1 n-9 (Oleic acid), C16:0 (Palmitic acid), C4:0 (Butyric acid), C18:2 n-6 (Linoleic acid, LA) and C20:5 n-3 (Eicosapentaenoic acid, EPA).

Özçiçek, Esin; Can, Erkan; Yılmaz, Ökkeş

In this study, fatty acids, fat-soluble vitamins (A, D, E, K vitamins) and cholesterol contents of wild-caught and farm-raised rainbow trout (Oncorhynchus mykiss) were compared. The amounts of docosahexaenoic acid (22:6 n-3, DHA) in wild and farmed forms were 21.56%–18.14%, 18.79%–16.21% and 14.34%–11.49% in April, May and June, respectively. Vitamin E, which is one of the fat-soluble and antioxidant vitamins, showed highest level among the vitamins. Vitamin E levels in both wild and farmed fish were 5.11–4.26, 4.83–2.89 and 0.99–1.95 mg/100 g in April, May and June, respectively. The substantial difference between wild and farmed rainbow trout was found in May (p < 0.05). In conclusion, both wild and farmed rainbow trout from Keban Dam Lake (Pertek Region) can be considered as good sources of n-3 polyunsaturated fatty acids (PUFA). However, it was observed that the n-3 PUFA level in wild form was higher than the culture fish within the investigated monthly periods. On the other hand, farmed fish demonstrated higher levels of n-6 PUFA over the wild individuals.

FATTY ACID CONTENT AND FREE RADICAL SCAVENGING (DPPH) ACTIVITIES OF SOME OF BRYOPHYTES EXTRACTS

Aydinl, Sevinc; Alatas, Mevlut; Batan, Nevzat

In this study, DPPH and fatty acid contents were investigated of some taxa belonging to Neckeraceae (Alleniella complanata, Alleniella besseri, Exsertotheca crispa) and Hypnaceae (Hypnum cupressiforme, Hypnum resupinatum, Hypnum andoi) family collected from Kamilet Valley (Artvin-Arhavi) in 2017. DPPH free radical scavenging activity was determined according to the method developed by Brand-Williams et al. The fatty acids in extracts were analyzed via gas chromatography after transforming them into methyl esters. According to the obtained results, it was determined that the DPPH scavenging effects of taxa from Hypnaceae family were significantly higher than those of taxa from Neckeraceae family, and that the free radical scavenging effects of taxa from Hypnaceae family varied depending on the amount of extract. The saturated fatty acids content of the Neckeraceae and the unsaturated fatty acids of the Hypnaceae were found to be richer (p<0.001). The high DPPH activity in our study, especially in Hypnum species, suggested that Hypnum species were richer in terms of phenolic content. These compounds can be used in the pharmaceutical, cosmetic and food sectors, if both of these species are phenolic and fatty acid content can be obtained as pure.

THE EFFECTS HYDROXYUREA DERIVATIVE SCHIFF BASES ON FATTY ACIDS AND LYPOPHILIC VITAMINS IN LIVER OF RATS

Turkoglu, Semra; Parlak, Akif E.; Yildirim, Isil; Karagozoglu, Yusuf; Alayunt, Naci O.

Schiff bases and their metal complexes have antiviral, antimicrobial, antitumoral, radical scavenging and antioxidant activities. In this study, effects of Schiff base ligand and its metal complexes on the fatty acids and lyphophlilic vitamins of the liver of rats were investigated. The fatty acids of the liver were determined by GC and lyphophlilic vitamins of the liver determined by HPLC. The amounts of stearic acid (18:0), oleic acid (18:1) and monounsaturated fatty acids (MUFA) considerably decreased in the Mn, Ni and Zn complexes groups, whereas the amount of linolenic acid (18:3) and eicosatrienoic acid (20:3) considerably increased in the Ni an Zn complexes groups compared to the control group (P<0.01). However, there was no significant difference (P>0.05) in the amounts of arachidonic acid (20:4), docosahexaenoic acid (22:6), total saturated fatty acids (SFA) and polyunsaturated fatty acids (PUFA), total omega-3 and omega-6 fatty acids between groups. The amounts of vitamin K-1, vitamin D-2 considerably increased (P<0.05) in the Mn and Ni complexes groups as compared to the control. It was observed that the amount of a-tocopherol increased both in the Ni and Co complexes groups when compared to the control group but this increase was parallel to the amount in the same groups (P<0.05). As a result, it can be think that with the application of toxic metals such as manganese, cupper, nickel, cobalt, and zinc unsaturated fatty acids affected the activities of enzymes which are in the liver tissue and in charge in the fatty acid chain elongation.
