This study aims to evaluate the effects of Momordica charantia on hepatorenal toxicity caused by potassium bromate (KBrO3) in rats. Wistar rats were divided into 4 groups as control, KBrO3, bitter melon (MC), and KBrO3+MC. Examining the antioxidant enzyme grade of the kidney tissues, it was found that the enzyme activities of catalase (CAT) (P<0.05), glutathione peroxidase (GSH–Px) (P<0.01), and superoxide dismutase (SOD) (P<0.01) decreased in the KBrO3 group in comparison to the control. There was a significant decrease (P<0.001) in glutathione (GSH) levels and an increase (P<0.01) in malondialdehyde (MDA) level in the KBrO3 group in comparison to the control. Examining the antioxidant enzyme activities in liver tissue, it was determined that CAT, GSH–Px, and SOD enzymes reduced significantly (P<0.05, P<0.01, and P<0.001, respectively) in the KBrO3 group in comparison to the control, and the enzyme activity of decreased CAT, GSH–Px, and SOD enzymes significantly elevated (P<0.01) in the MC group. There was a reduction in GSH level in the KBrO3 group in comparison to the control (P<0.01), while an increase was recorded in the KBrO3+MC group (P<0.05). MDA level in liver tissue increased in KBrO3 group in comparison to the control (P<0.01) and MC decreased the MDA level. Histopathological analysis results indicate severe degenerative and necrotic lesions in hepatorenal histoarchitecture of KBrO3 rats in comparison to the control. However, application of MC+KBrO3 significantly reduced the induced hepatorenal injury with a concomitant increase in histopathological lesions. From the immunohistochemical aspect, MC revealed apoptosis concomitant with the suppression of necrosis in the KBrO3-treated rats as demonstrated by the caspase-3 activity.

Expression of phoenixin-14 and nesfatin-1 in the hypothalamo–pituitary–gonadal axis in the phases of the estrous cycle

Parlak Ak, Tuba; Yaman, Mine; Bayrakdar, Ali; Bulmus, Ozgur

Phoenixin-14 (PNX-14) and nucleobindin 2 (NUCB2)/nesfatin-1 are regulatory neuropeptides expressed in the hypothalamus. These neuropeptides can be effective in hormonal regulation of the hypothalamo–pituitary–gonadal (HPG) axis and reproductive functions. In the present study, the distribution of PNX-14 and NUCB2/nesfatin-1 in the hypothalamus, pituitary, ovary, and uterus tissues during the phases of the estrous cycle in female rats was investigated. Eighteen Wistar Albino rats determined among animals showing regular estrous cycle by vaginal smear method were divided into three groups: proestrus (Group I), estrus (Group II) and diestrus (Group III). Serum gonadotropin-releasing hormone (GnRH), plasma PNX-14, and NUCB2/nesfatin-1 concentrations were the highest, moderate, and lowest in estrus, diestrus, and proestrus phases, respectively. PNX-14 immunoreactivity in the supraoptic and arcuate nuclei of the hypothalamus and NUCB2/nesfatin-1 immunoreactivity in the paraventricular nuclei were particularly evident in the estrus phase. These neuropeptide immunoreactivities were decreased in different cells of anterior pituitary during proestrus compared with those during estrus and diestrus. PNX-14 immunoreactivity in the ovary, especially during the estrus phase, was diffuse and intense in the granulosa and luteal cells and oocytes, and it was few and weak in theca cells. In addition, NUCB2/nesfatin-1 immunoreactivity was abundant and strong in granulosa and luteal cells, theca and interstitial cells, and oocytes during estrus. In the estrus phase, PNX-14 immunoreactivity was strong in the glandular epithelial cells and stromal cells of the endometrium, also NUCB2/nesfatin-1 immunoreactivity was strong in the epithelial and glandular epithelial cells. As a result, when the estrous cycle was evaluated, it was concluded that the changes in the distribution of PNX-14 and NUCB2/nesfatin-1 at all phases were related to GnRH and that these neuropeptides showed the highest immunoreactivity especially in the HPG axis and uterus tissues of estrus rats.

The antiiflammatory effect of Ginkgo biloba in lipopolysaccharide-induced rat sepsis model

Ilhan, Nevin; Susam, Solmaz; Ak, Tuba Parlak; Baykalir, Burcu Gul

Background: Sepsis is characterized dysregulated inflammatory response and is the leading cause of organ failure or injury and death in critical care units. Ginkgo biloba extract 761 (EGb 761) is characterized by antioxidant activity. There is no clear data in the functional role of EGb 761 that inhibiting of proinflammatory cytokines exist in sepsis. The purpose of this study was to investigate the antiinflammatory effect of EGb 761 on the levels of vascular endothelial growth factor (VEGF), matrix metalloproteinase-9 (MMP-9) and inducible nitric oxide synthase (iNOS) in lipopolysaccharide (LPS) induced rat sepsis model. Methods: The animals were randomly divided into the following six groups: the control, LPS, flunixin meglumine (FM), EGb 761, LPS + FM and LPS + EGb 761 groups. Enzyme-linked immunosorbent assay (ELISA) analysis was applied to measure serum levels of VEGF, MMP-9, and iNOS. Results: Serum iNOs (p < 0.05), VEGF (p < 0.01) and MMP-9 (p < 0.05) levels were significantly higher in the sepsis group. Treatment with EGb 761 decreased serum iNOs levels (p < 0.01), VEGF and MMP-9 levels decreased significantly (p < 0.05). Also, treatment with FM decreased iNOs (p < 0.01), VEGF and MMP-9 levels significantly (p < 0.05). Conclusions: It is suggested that sepsis may lead to increase serum iNOs, MMP-9, and VEGF levels and treatment of EGb 761 may have a potential antiinflammatory effect against LPS-induced sepsis.

THE EFFECTS HYDROXYUREA DERIVATIVE SCHIFF BASES ON FATTY ACIDS AND LYPOPHILIC VITAMINS IN LIVER OF RATS

Turkoglu, Semra; Parlak, Akif E.; Yildirim, Isil; Karagozoglu, Yusuf; Alayunt, Naci O.

Schiff bases and their metal complexes have antiviral, antimicrobial, antitumoral, radical scavenging and antioxidant activities. In this study, effects of Schiff base ligand and its metal complexes on the fatty acids and lyphophlilic vitamins of the liver of rats were investigated. The fatty acids of the liver were determined by GC and lyphophlilic vitamins of the liver determined by HPLC. The amounts of stearic acid (18:0), oleic acid (18:1) and monounsaturated fatty acids (MUFA) considerably decreased in the Mn, Ni and Zn complexes groups, whereas the amount of linolenic acid (18:3) and eicosatrienoic acid (20:3) considerably increased in the Ni an Zn complexes groups compared to the control group (P<0.01). However, there was no significant difference (P>0.05) in the amounts of arachidonic acid (20:4), docosahexaenoic acid (22:6), total saturated fatty acids (SFA) and polyunsaturated fatty acids (PUFA), total omega-3 and omega-6 fatty acids between groups. The amounts of vitamin K-1, vitamin D-2 considerably increased (P<0.05) in the Mn and Ni complexes groups as compared to the control. It was observed that the amount of a-tocopherol increased both in the Ni and Co complexes groups when compared to the control group but this increase was parallel to the amount in the same groups (P<0.05). As a result, it can be think that with the application of toxic metals such as manganese, cupper, nickel, cobalt, and zinc unsaturated fatty acids affected the activities of enzymes which are in the liver tissue and in charge in the fatty acid chain elongation.

THE ROLE OF METHYLATED ADRENOMEDULLIN IN ENZYMATIC ANTIOXIDANT DEFENSE RESPONSE TO COLD STRESS

    Yildirim, Nuran Cikcikoglu; Yurekli, Muhittin

The aim of our study was to investigate the effects of methylated adrenomedullin (met-AdM) on some antioxidant enzymes, such as catalase (CAT), glutathione peroxidase (GSH-P-X) and superoxide dismutase (SOD) in rat kidney and heart tissues after cold stress treatment. Male Wistar rats were divided into 4 groups (control, met-AdM, cold stress and cold stress + met-AdM treatments). The rats were kept at room temperature, exposed to a 16/8-hours light/dark cycle. The rats were maintained at ad libitum (free) feeding habit. In met-AdM treated group, animals received intraperitoneal injection of met-AdM (2 mg/g body weight) once a day during a week. For the cold stress exposure, the rats were kept in separate cages at 10 degrees C for a week. Control group rats were kept at laboratory conditions. CAT, GSH-P-X and SOD enzyme activities were investigated in kidney and heart tissues. In kidney tissue compared to control, CAT and GSH-P-X enzyme activity was decreased but SOD was increased in met-AdM-treated group. In met-AdM+cold stress group, only the SOD enzyme activity was significantly increased but CAT and GSH-P-X decreased. SOD and GSH-P-X enzyme activities were increased by cold stress but CAT was decreased in kidney. In heart tissue, CAT and GSH-P-X activities were decreased but SOD was increased in met-AdM-treated group compared to control. All enzyme activities were decreased in met-AdM+cold stress group as well as by cold stress in heart, with regard to control. Cold stress can disrupt the balance in an oxidant/antioxidant system and cause oxidative damage to kidney and heart tissues by altering the enzymatic and non-enzymatic antioxidant status. We can suggest that met-AdM has possible roles in regulation of stress but we need further studies revealing responses of met-AdM in different tissues.

Stress-Induced Changes in Tyrosine Hyroxylase Enzyme Activity and Adrenomedullin Levels in Rat Hypothalamus, Adrenal Medulla and Heart Tissues

    Yildirim, Nuran Cikcikoglu; Yurekli, Muhittin

In this study, it was aimed to investigate the effects of chronic cold stress on adrenomedullin levels and tyrosine hydroxylase enzyme activities in some rat tissues. In this study, 12 female Sprague Dawley rats weighing 200 to 250 g were used. Rats were housed under diurnal lighting conditions (12-12 h) with free access to food and water. For this study, rats were divided into two groups, namely control group and cold stress treated group. Adrenomedullin levels were measured by high performance liquid chromatography. Tyrosine hydroxylase enzyme activities was spectrometrically measured. Tyrosine hydroxylase activity was found to be increased significantly (p<0.05) in adrenal medulla, hypothalamus and heart tissues depending on cold stress treatment. Adrenomedullin levels were determined to be decreased in the cold stress treatment group. The differences of between control and cold stress treatment groups were significant (p<0.05). The results suggested that adrenomedullin may play a possible role in adaptation to stress. Exposure to cold increases the synthesis and release of catecholamines. Tyrosine hydroxylase is the rate limiting step in the biosynthesis of catecholamines for this reason cold stress increased tyrosine hyroxylase enzyme activities. Further studies are needed to reveal correlation between adrenomedullin, tyrosine hydroxylase and cold stress.

The effect of adrenomedullin and cold stress on interleukin-6 levels in some rat tissues

    Yildirim, N. C.; Yurekli, M.

P>Stress known to stimulate sympathetic activity, as well as the hypothalamic-pituitary-adrenal axis (HPA), produces a significant increase in adrenomedullin (AdM) levels, suggesting a regulatory or protective role for AdM in countering HPA activation that follows a variety of stressors. Stressors can modulate the secretion of proinflammatory cytokines. Interleukin (IL)-6 is a potent activator of the HPA and appears to play a pathogenic role in conditions related to stress. In the present study, we investigated the administration of AdM on IL-6 levels in cold exposed rats. Male Wistar rats were divided into four groups as control, adrenomedullin treatment, cold stress and cold stress+adrenomedullin-treated groups. In the adrenomedullin-treated group, animals received intraperitoneal (i.p.) injection of adrenomedullin (2000 ng/kg body weight) once a day for a week. For the cold stress exposure the rats were kept in separate cages at 10 degrees C for a week. Control group rats were kept in laboratory conditions. The concentration of IL-6 was determined using an enzyme-linked immunosorbent assay (ELISA) kit. When compared to control, IL-6 levels increased significantly in the cold stress- and adrenomedullin-treated groups (P < 0 center dot 05). Administration of AdM in addition to cold stress decreased IL-6 levels in lung and liver, but increased in brain and heart when compared to control (P < 0 center dot 05). The results suggest that cold stress may induce increase of rat proinflammatory cytokine IL-6 and adrenomedullin may play a regulatory or protective role for cold stress.

Adrenomedullin administration alters vascular endothelial growth factor levels in rats in cold stress

    Yildirim, Nuran Cikcikoglu; Yurekli, Muhittin

Background: Many endogenous peptides play important regulatory roles in angiogenesis by modulating endothelial cell behavior. Adrenomedullin (AdM) is one of such factors. Angiogenesis and vascular endothelial growth factor (VEGF) are indistinguishable. Exposure to cold environment stimulates capillary angiogenesis. Objectives: Examine the effect of the bioactive peptide AdM on VEGF levels in rat liver, lung, brain, and heart tissues after cold stress treatment. Methods: Male wistar rats were divided into four groups as control, AdM treatment, cold stress and AdM+cold stress treated groups. In AdM-treated group, animals received intraperitoneal injection of AdM (2000 ng/kg body weight) once a day during a week. For the cold stress exposure, the rats were kept in separate cages at 10 degrees C for a week. Results: The administration of AdM increased VEGF levels in all tissues in cold exposed rats. Conclusion: AdM may be a major regulatory factor in angiogenesis by modulating VEGF levels that is closely associated with cold exposure-related metabolic stimulation.
