Biosynthesis of silver nanoparticles using the aqueous extract of Rheum ribes, characterization and the evaluation of its toxicity on HUVECs and Artemia salina

    Unal, Ilkay; Egri, Sinan

This study describes an eco-friendly procedure to synthesize silver nanoparticles (AgNPs) using fresh and dried stem extract of Rheum ribes. AgNPs formation was confirmed using UV-vis spectrometry. The effects of parameters such as temperature, reaction time, pH, AgNO3 concentration, and plant extract/AgNO3 ratio on NP formation were evaluated. Morphology and structural features of the particles were characterized using SEM, TEM, XRD, and FTIR. The average size, polydispersity index, and zeta potential were determined using DLS. After characterization, among the two AgNP samples, selected one; AgNP sample synthesized by dried Rheum ribes stem extracts (d-AgNPs), which exhibited narrow size distribution and high surface plasmon resonance in the range of 435-445 nm for toxicity assessment. The cytotoxic effect of d-AgNPs on HUVECs cells was evaluated with MTT analysis. Accumulation measurements were made in three periods as 24, 48, and 72 h and elimination values were examined at +24 h using ICP-MS analysis

Unraveling the ecotoxicological effects of micro and nano-plastics on aquatic organisms and human health

Naz, Saima; Chatha, Ahmad Manan Mustafa; Khan, Nisar Ahmed; Ullah, Qudrat; Zaman, Faisal; Qadeer, Abdul; Khan, Ibrar Muhammad; Danabas, Durali; Kiran, Azka; Skalickova, Sylvie

Plastic pollution ranks among the most severe environmental disasters caused by humans, generating millions of tonnes of waste annually. The extensive and unregulated use of plastics has led to ecotoxicity and environmental imbalance. Microplastics (MPs) are prevalent in aquatic environments, and these MPs further degrade into even smaller particles known as nano-plastics (NPs). Both MPs and NPs impact the environment by readily absorbing organic pollutants and pathogens from their surroundings, owing to their bigger surface area to volume ratio. This review focuses on the source of origin, bioaccumulation, and potential impact of MPs and NPs on aquatic organisms and human health. Additionally, the review explores various methods employed for identification and quantification of these particles in aquatic ecosystems. Sufficient information is available on their characteristics, distributions, and effects on marine ecosystems compared with freshwater ecosystems. For plastic particles <10 μm, more toxicological effects were observed compared with larger size particles, in aquatic life. Understanding the mechanism of action and ecotoxicological effects of micro/nano-plastics on the health of aquatic life across various trophic levels, as well as human health, is of utmost importance. We address knowledge gaps and provide insights into future research approaches for a better understanding of the interactive mechanisms between binary pollutants.

The potential ameliorative impacts of rare earth element cerium in dimethoate induced toxicity in freshwater mussel Dreissena polymorpha

Serdar, Osman; Ketenalp, Zozan; Cikcikoglu Yildirim, Nuran

The purpose of this study was to measure the antioxidant parameters to examine the potential ameliorative impacts of the rare earth element (REE) cerium (Ce) in dimethoate (DM) induced toxicity in Dreissena polymorpha. Lethal concentration 50 (LC50) of DM for 96 h was measured. The antioxidant effect of Ce was investigated by exposure of 1/2 of the LC50 value of DM together with 4, 20 and, 100 mg/L of Ce to D. polymorpha. The responses of the biomarkers were determined by measuring the superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione (GSH) and malondialdehyde (MDA) levels. SOD, CAT, GPX and GSH levels were reduced (p < 0.05), while the MDA levels increased significantly (p < 0.05) in mussels exposed to DM. When DM was exposured together with Ce, it was observed that the MDA levels decreased, SOD, CAT, GPX and GSH levels increased during 24 and 96 h. The results of our study showed that Ce exerted a strong antioxidant effect on toxicity induced by DM. It has been shown that Ce may have a compensatory effect with the positive change in the antioxidant response of D. polymorpha to DM. This study is the first report about Ce and insecticide effects in bivalves.

Contamination and Ecological Risk Assessment of Heavy Metals in Surface Sediments of the Munzur Stream, Turkey

    Kutlu, Banu

The aim of the present study is to determine the heavy metal concentrations in the surface sediment between 2019-2020 in Munzur Stream,Turkey. The impact of pollution level in the study area was evaluated by geoaccumulation index (Igeo), Contamination Factor (CF), enrichment factor (EF), pollution ecological risk (PER) index, and pollution load index (PLI). The average amount of heavy metals detected in sediment in the study in mg/kg was, in descending order, Fe>Al>Mn>Ni>Cr>Zn >Cu>Pb>Cd>Hg. The data evaluated by several statistical methods indicated that the metals with a positive relationship between them in the Pearson correlation analysis and the elements of the same factor in the factor analysis were mostly the same. The comparison of the results with the sediment quality criteria showed that only the mean value of Mn and Ni exceeded the values of PEL. Heavy metal levels in Munzur Stream sediment exposed to domestic and agricultural wastes, agricultural fertilizers and pesticides, and mines were not high enough to pose a threat to aquatic life.

Cellular responses of spirlin Alburnoides bipunctatus spermatozoa exposed to Bisphenol A: Biochemical response and sperm quality alterations evaluation

Kocabaş, Mehmet; Kutluyer, Filiz; Çakir Sahilli, Yeliz; Aksu, Önder

Plastics are one of the global environmental problems owing to their highest production. Bisphenol A (BPA) is used in the manufacturing of plastics including polycarbonate, polyvinyl chloride and resins. In particular, aquatic ecosystems are adversely affected by the occurrence of micropollutants. In addition, BPA as a typical endocrine-disrupting chemical (EDC) seriously influences androgenic activity and reproduction of aquatic organisms, particularly fish as non-target organism. In toxicological assays, sperm cells can be used as a potential tool for reliable and practical assessment. For these reasons, sperm cells were exposed to lower concentrations (0 µg/L, 0.5 µg/L, 1.25 µg/L, 2.5 µg/L and 5 µg/L) of BPA and the study focused on BPA-induced oxidative stress. The enzymatic [glutathione peroxidase (GSH-Px), catalase (CAT) and superoxide dismutase (SOD)] activities and lipid peroxidation (MDA) were analyzed in spermatozoa. The study findings showed that the activities of SOD and GSH-Px were significantly decreased by BPA but lipid peroxidation (MDA) and CAT acticity increased. Notably, the percentage and duration of motile sperm cells decreased. Collectively, spermatozoa clearly showed the sensitivity even at extremely low concentrations. This is first report to assess the impact of BPA on sperm cells of spirlin Alburnoides bipunctatus.

Comparative Toxicity of Alpha and Gamma Iron Oxide Nanoparticles in Rainbow Trout: Histopathology, Hematology, Accumulation, and Oxidative Stress

Gürkan, Mert; Gürkan, Selin Ertürk; Yılmaz, Sevdan; Ateş, Mehmet

In this study, alpha and gamma iron oxide nanoparticles, characterizations, toxic effects after being exposed to rainbow trout (Oncorhynchus mykiss) at 0, 1, 10 and 25 mg/L concentrations for 10 days, and then 10-day recovery period without any nanoparticle exposure were examined for histopathological (kidney, liver and gill), hematological, iron accumulation and potential for oxidative stress (TBARS and GSH). Histopathological damages significant at the exposure of increasing concentrations of both nanoparticles (increase in melanomacrophage aggregations, epithelial tissue deformations, cytoplasmic vacuolizations, fatty changes, necrosis, pyknosis, hyperplasia, hypertrophy, lamellar fusions, capillary dilatations). Gamma nanoparticles were determined to accumulate more than the alpha nanoparticles. The most Fe accumulation was detected in the liver. The findings of oxidative stress parameters showed that both nanoparticles have the potential to generate oxidative stress. It was concluded that the exposure of alpha and gamma nanoparticles at specified concentrations and durations had a toxic effect on rainbow trout and the toxicity of these nanoparticles was similar.

The Use of Astacus leptodactylus (Eschscholtz, 1823) as a Test Species for Toxicity Evaluation of Municipal Wastewater Treatment Plant Effluents

    Yildirim, Cikcikoglu N.; Aksu, O.; Tatar, S.; Yildirim, N.

This study evaluated the responses of biochemical biomarkers in Astacus leptodactylus exposed to treated municipal effluents discharged into Keban Dam Lake, Elazig Elazig, Turkey. A. leptodatyclus were exposed to treated municipal effluents and Catalase (CAT), Superoxide dismutase (SOD) activity and lipid peroxidation (TBARS), glutathione (GSH) levels were measured as oxidative stress biomarkers. SOD activity was increased after exposing to treated municipal effluents for 24th and 96th h. CAT activities were decreased from 25.29 to 14.12 nmol/min/ml compared to control in the group exposed to treated municipal effluents for 24 h but it increased after 96 h exposure. GSH levels were decreased from 9.08 to 3.77 mu M compared to control, but MDA levels were increased both at 24 th h and 96 th h after exposure to treated municipal effluents. CAT, SOD activities and MDA and GSH levels in the hepatopancreas of A. leptodactylus are sensitive and suitable biochemical biomarkers for evaluating the toxicity of the treated municipal effluent complex mixtures. Treated municipal effluents exposure was found to cause sub-lethal responses in A. leptodactylus suggesting oxidative stress.

Assessment of impact of α-Fe2O3 and γ-Fe2O3 nanoparticles on phytoplankton species Selenastrum capricornutum and Nannochloropsis oculata

Ates, Mehmet; Cimen, Isil C. C.; Unal, Ilkay; Kutlu, Banu; Ertit Tastan, Burcu; Danabas, Durali; Aksu, Onder; Arslan, Zikri

In this study, the impact of alpha-iron oxide (α-Fe2O3, 20-40 nm) and gamma iron oxide (γ-Fe2O3, 20-40 nm) nanoparticles (NPs) on phytoplankton species Selenastrum capricornutum and Nannochloropsis oculata was investigated Characterizations of the NPs were systematically carried out by TEM, dynamic light scattering, zeta potential, X-ray diffraction, SEM, and Fourier transformation infrared spectroscopy. Acute toxicity was tested between 0.2 and 50 mg/L for each NP for a period of 72 hours exposure. γ-Fe2O3 NP inhibited development of N oculata at the rate of 54% in 0.2 mg/L group with a high mortality rate of up to 82%. α-Fe2O3 NPs were less toxic that induced 97% mortality on N oculata at 10 mg/L suspensions. In contrast, α-Fe2O3 NP inhibited growth of S capricornutum strongly (73%) in 0.2 mg/L group. γ-Fe2O3 NPs showed similar growth inhibition (72%) on S capricornutum in 10 mg/L suspensions. Despite the differential effects, the results indicated acute toxicity of α-Fe2O3 and γ-Fe2O3 NPs on N oculata and S capricornutum.

Effects of Zn and ZnO Nanoparticles on Artemia salina and Daphnia magna Organisms: Toxicity, Accumulation and Elimination

Danabas, Durali; Ates, Mehmet; Ertit Tastan, Burcu; Cicek Cimen, Isil Canan; Unal, Ilkay; Aksu, Onder; Kutlu, Banu

In the study, Zn in the size of 40–60 nm and 80–100 nm and ZnO in the size of 10–30 nm were applied to A. salina and D. magna individuals in 7 groups with 3 repetitions. Measurements were made at 24th, 48th and 72nd hours and elimination values were examined at +24 h. LC50 values of NPs were determined and chemical analysis (metal accumulation and elimination), ion quantities which were given to the environment and the survival rates of organisms were determined after the exposure. According to the results of phase contrast microscopy, it was found that both experimental organisms absorbed the NPs in the medium level. In the toxicity results of D. magna, it can be said that Zn NP (40–60 nm) has a highly toxic effect only at 50 ppm concentration for 48 h and lethal dose can be accepted as of 5 ppm at the end of 72 h. In A. salina individuals, it is clearly seen that there is an increase in mortality in organisms parallel to the dose increase. Although all NPs were applied to organisms in low doses corresponding to environmental values, it was observed that toxic effect was in parallel with the increase in time. It is clearly known that there is the inverse proportion between the size of NPs and the toxic effect. The smaller the size of NPs is, the higher the toxic effect becomes When the results of Zn accumulation and elimination of A. salina and D. magna individuals exposed to the Zn and ZnO NPs were examined; it was found that accumulation and elimination occurred in parallel with the increase in concentration at each application hour and elimination. Intensive and possible misuse of nanoscale materials is one of the biggest threats to the environment and all living things worldwide.

Accumulation and Toxicity of CuO and ZnO Nanoparticles Through Waterborne and Dietary Exposure of Goldfish (Carassius auratus)

Ates, Mehmet; Arslan, Zikri; Demir, Veysel; Daniels, James; Farah, Ibrahim O.

Dietary and waterborne exposure to copper oxide (CuO) and zinc oxide (ZnO) nanoparticles (NPs) was conducted using a simplified model of an aquatic food chain consisting of zooplankton (Artemia salina) and goldfish (Carassius auratus) to determine bioaccumulation, toxic effects, and particle transport through trophic levels. Artemia contaminated with NPs were used as food in dietary exposure. Fish were exposed to suspensions of the NPs in waterborne exposure. ICP-MS analysis showed that accumulation primarily occurred in the intestine, followed by the gills and liver. Dietary uptake was lower, but was found to be a potential pathway for transport of NPs to higher organisms. Waterborne exposure resulted in about a 10-fold higher accumulation in the intestine. The heart, brain, and muscle tissue had no significant Cu or Zn. However, concentrations in muscle increased with NP concentration, which was ascribed to bioaccumulation of Cu and Zn released from NPs. Free Cu concentration in the medium was always higher than that of Zn, indicating CuO NPs dissolved more readily. ZnO NPs were relatively benign, even in waterborne exposure (p0.05). In contrast, CuO NPs were toxic. Malondialdehyde levels in the liver and gills increased substantially (p<0.05). Despite lower Cu accumulation, the liver exhibited significant oxidative stress, which could be from chronic exposure to Cu ions. (c) 2014 Wiley Periodicals, Inc. Environ Toxicol 30: 119-128, 2015.

Evaluation of Alpha and Gamma Aluminum Oxide Nanoparticle Accumulation, Toxicity, and Depuration in Artemia salina Larvae

Ates, Mehmet; Demir, Veysel; Arslan, Zikri; Daniels, James; Farah, Ibrahim O.; Bogatu, Corneliu

In this study, Artemia salina (crustacean filter feeders) larvae were used as a test model to investigate the toxicity of aluminum oxide nanoparticles (Al2O3 NPs) on marine microorganisms. The uptake, toxicity, and elimination of -Al2O3 (50 nm and 3.5 m) and -Al2O3 (5 nm and 0.4 m) NPs were studied. Twenty-four and ninety-six hour exposures of different concentrations of Al2O3 NPs to Artemia larvae were conducted in a seawater medium. When suspended in water, Al2O3 NPs aggregated substantially with the sizes ranging from 6.3 nm to >0.3 mu m for spherical NPs and from 250 to 756 nm for rod-shaped NPs. The phase contrast microscope images showed that NPs deposited inside the guts as aggregates. Inductively coupled plasma mass spectrometry analysis showed that large particles (3.5 m -Al2O3) were not taken up by Artemia, whereas fine NPs (0.4 m -Al2O3) and ultra-fine NPs (5 nm -Al2O3 and 50 nm -Al2O3) accumulated substantially. Differences in toxicity were detected as changing with NP size and morphology. The malondialdehyde levels indicated that smaller -Al2O3 (5 nm) NPs were more toxic than larger -Al2O3 (0.4 mu m) particulates in 96 h. The highest mortality was measured as 34% in 96 h for -Al2O3 NPs (5 nm) at 100 mg/L (LC50 > 100 mg/L). -Al2O3 NPs were more toxic than -Al2O3 NPs at all conditions. (c) 2013 Wiley Periodicals, Inc.
