- MakaleAçık Erişim2016Scopus SJRQ1The in vitro effect of cypermethrin on quality and oxidative stress indices of rainbow trout Oncorhynchus mykiss spermatozoa
Kutluyer, Filiz; Benzer, Fulya; Erisir, Mine; Ögretmen, Fatih; Inanan, Burak Evren
There is limited information on the scientific literature about the effect of in vitro exposure of fish sperm to pesticides. In vitro effect of cypermethrin on sperm quality and oxidative stress has not yet been fully investigated. Therefore, the effects of cypermethrin, a type II pyrethroid insecticide, on quality and oxidative stress of spermatozoa were examined in vitro. To explore the potential in vitro toxicity of cypermethrin, fish spermatozoa were incubated with different concentrations of cypermethrin (1.025, 2.05 and 4.1 mu g/l) for 2 h. The motility rate and duration of sperm were determined after exposure to cypermethrin. Reduced glutathione (GSH), glutathione peroxidase (GSH-Px), catalase (CAT) and malondialdehyde (MDA) in spermatozoa were analyzed for determination of oxidant and antioxidant balance. Our results indicated that spermatozoa motility and duration significantly decreased with exposure to cypermethrin. Additionally, activity of GSH-Px (P < 0.05) and MDA and GSH levels increased in a concentration-dependent manner while CAT activity decreased (P < 0.05). Consequently, the oxidant and antioxidant status and sperm quality were affected by quantitative changes and different concentrations of cypermethrin. (C) 2015 Elsevier Inc. All rights reserved.

Altundogan, Hamdi Soner; Topdemir, Aykut; Çakmak, Meltem; Bahar, Nurdan
In this study, an effective cation exchanger was developed from the pine cone by citric acid modification and its hardness removal properties from the waters were investigated. For this purpose, ground pine cone samples were subjected to a citric acid modification following NaOH saponification. Both of the raw (RPC) and modified pine cone (MPC) samples were subjected to standardized hardness removal tests by shaking with hard waters. These tests showed that citric acid modification significantly increases the cation exchange capacity of the pine cone. Most suitable size fraction of pine cone was determined as -16 + 30 mesh (600 mu m < x < 1200 mu m). Also, citric acid modified product obtained from this fraction was used in a continuous system to remove hardness from water. The results of this study showed that the MPC can be used for hardness removal from waters as a cheap, durable and environment -friendly material. Finally, regeneration experiments showed that the MPC can be used for hardness removal from water repeatedly by a simple HCl regeneration. (C) 2015 Taiwan Institute of Chemical Engineers. - MakaleAçık Erişim2016Scopus SJRQ2Toxicity of Engineered Nickel Oxide and Cobalt Oxide Nanoparticles to Artemia salina in Seawater
Ates, Mehmet; Demir, Veysel; Arslan, Zikri; Camas, Mustafa; Celik, Fatih
In this study, the effects of exposure to engineered nickel oxide (NiO 40-60 nm) and cobalt oxide (CoO < 100 nm) nanoparticles (NP) were investigated on Artemia salina. Aggregation and stability of the aqueous NP suspensions were characterized by DLS and TEM. Acute exposure was conducted on nauplii (larvae) in seawater in a concentration range from 0.2 to 50 mg/L NPs for 24 h (short term) and 96 h (long term). The hydrodynamic diameters of NiO and CoO NPs in exposure medium were larger than those estimated by TEM. Accumulation rate of NiO NPs were found to be four times higher than that of CoO NPs under the same experimental conditions. Examinations under phase contrast microscope showed that the nanoparticles accumulated in the intestine of Artemia, which increased with increasing exposure concentration. Differences were observed in the extent of dissolution of the NPs in the seawater. The CoO NPs dissolved significantly while NiO NPs were relatively more stable. Oxidative stress induced by the NP suspensions was measured by malondialdehyde assay. Suspensions of NiO NPs caused higher oxidative stress on nauplii than those of CoO NPs. The results imply that CoO and NiO NPs exhibit toxicity on Artemia (e. g., zooplankton) that is an important source of food in aquatic food chain.

Akbulut, Selman; Kalafat, Mustafa
In this paper we give a survey of various results about the topology of oriented Grassmannian bundles related to the exceptional Lie group G(2). Some of these results are new. We give self-contained proofs here. One often encounters these spaces when studying submanifolds of manifolds with calibrated geometries. For the sake of completeness we decided to collect them here in a self-contained way to be easily accessible for future usage in calibrated geometry. As an application we deduce existence of certain special 3 and 4 dimensional submanifolds of G(2) manifolds with special properties, which appear in the first named author's work with S. Salur about G(2) dualities. (C) 2015 Elsevier GmbH. All rights reserved.

- MakaleAçık Erişim2016Scopus SJRQ2Changes in the texture, physicochemical properties and volatile compound profiles of fresh Kashar cheese (<90 days) during ripening
Eroglu, Ali; Toker, Omer Said; Dogan, Mahmut
In this study, four different fresh Kashar cheese samples were ripened for 90 days. The physicochemical properties, texture attributes, fatty acid composition and volatile compound profiles of the samples were measured every 30 days of ripening. The texture properties of the cheese samples were significantly affected by the duration of the ripening period. The results of this study highlighted that texture parameters as a function of ripening period should be considered for both fresh and aged Kashar cheeses to determine the ripening period as they are very important for consumer acceptability and consumption of the end product.

Theileria annulata is an apicomplexan parasite which is responsible for tropical theileriosis in cattle. Due to resistance of T. annulata against commonly used antitheilerial drug, new drug candidates should be identified urgently. Enolase might be a druggable protein candidate which has an important role in glycolysis, and could also be related to several cellular functions as a moonlight protein. In this study; we have described three-dimensional models of open and closed conformations of T. annulata enolase by homology modeling method for the first time with the comprehensive domain, active site and docking analyses. Our results show that the enolase has similar folding patterns within enolase superfamily with conserved catalytic loops and active site residues. We have described specific insertions, possible plasminogen binding sites, electrostatic potential surfaces and positively charged pockets as druggable regions in T. annulate enolase. The purpose of this study was to examine the changes to occur in egg production, egg quality traits, hatchability, and oxidative stress parameters of laying quails fed with high calorie diets and effect of milk thistle (Silybum marianum) seed on these changes. A total of 75 45-day-old quails including 60 females and 15 males were used in the study. The quails were divided into 5 groups with three repetitions. 4 females and 1 male were used in each repetition. The groups of the study were arranged as following; control group (C) consuming corn-soybean based basal diet, oil group (SFO) in which 5% sun flower oil was added into basal diet, oil + milk thistle group (SFO+MT) in which 5% sun flower oil + 1% milk thistle seed was added into basal diet, syrup group (CS) in which 10% corn syrup was added into basal diet, and syrup + milk thistle group (CS+MT) in which 10% corn syrup + 1% milk thistle was added into basal diet. Total egg production was found to be significantly higher in SFO and SFO+MT groups of present study (P<0.05). Feed intake, total egg weight and feed conversion were found similar among the groups (P>0.05). The differences among the groups in albumen rate, yolk rate, dried shell rate and shape index were not statistically significant (P>0.05). The best yolk colour was obtained in SFO+MT and CS groups (P<0.01). There were no statistically differences among the groups in hatchability, hatchability of fertile and embrionic mortality (P>0.05), but the difference in examined fertility were statistically significant (P<0.05). Malonyl dialdehyde (MDA) values of blood, liver and heart tissues were similar among the groups of study (P>0.05). MDA level of kidney significantly increased in CS group, however addition of milk thistle seed into diet was reduced MDA level of kidney in CS+MT group (P<0.01). High calorie diets caused changes in antioxidant system in blood and tissues. Consequently, high-calorie basal diets especially addition of SFO improved only egg production and fertility rate. However feeding quails with corn syrup had significantly higher lipid peroxidation in kidney. Supplementation of milk thistle seed into CS group may protect kidney against free radical damage.

- MakaleAçık Erişim2016Scopus SJRQ2APPLICATION OF INTERVAL TYPE-2 FUZZY SETS DEMATEL METHODS IN MARITIME TRANSPORTATION: THE CASE OF SHIP COLLISION
Celik, E.; Akyuz, E.
Accident analyse in marine industry is one of the critical issues for safety practitioners to prevent loss of life. Although considerable efforts were undertaken to prevent marine accident, numerous researches revealed that marine accidents are still on-going. In order to minimize accidents in the marine transportation, this paper presents a proactive decision-making tool which is integrating Decision-Making Trail and Evaluation Laboratory (DEMATEL) method with interval type-2 fuzzy sets (IT2FSs). As the DEMATEL enables to analyse cause and effect relationship in decision-making, the IT2FSs overcome ambiguity and vagueness of linguistic assessment of decision-makers through the DEMATEL. Thus, significant accident causal factors and their effects can be analysed on the basis of cause-effect diagram. The application of proposed approach is demonstrated with a real ship collision case. Beside its theoretical contribution, the proposed approach provides practical benefits to ship owners and operators to perceive cause and effect relationship and to avoid marine accident.

- MakaleAçık Erişim2016Scopus SJRQ1Geochemistry of the metavolcanic rocks from the Çangaldağ complex in the central pontides: Implications for the middle jurassic arc-back-arc system in the neotethyan intra-pontide ocean
Çimen, Okay; Göncüoğlu, M. Cemal; Sayit, Kaan
The Cangaldağ Complex in northern central Turkey is one of the main tectonic units of the Central Pontide Structural Complex that represents the remains of the poorly known Intra-Pontide branch of the Neotethys. It comprises low-grade metamorphic rocks of intrusive, extrusive, and volcaniclastic origin displaying a wide range of felsic to mafic compositions. Petrographically the complex consists of basalts-andesites-rhyodacites and tuffs with minor amount of gabbros and diabases. On the basis of geochemistry, the Cangaldağ samples are of subalkaline character and represented by both primitive and evolved members. All rock types are variably depleted in Nb compared to LREEs, similar to the lavas from subduction-related tectonic settings. In N-MORB normalized plots, the primitive members are separated into 3 groups on the basis of levels of enrichment. The first group is highly depleted and displays characteristics of boninitic lavas. The second group is relatively enriched compared to the first group but still more depleted than N-MORB. The third group, however, is the most enriched one among the three, whose level of enrichment is around that of N-MORB. The overall geochemical features suggest that the Cangaldağ Complex has been generated with the involvement of a subductionmodified mantle source. The chemistry of the primitive members further indicates that the melts generated for the formation of the Cangaldağ Complex probably occurred in both arc and back-arc regions above an intraoceanic subduction within the Intra-Pontide branch of the Neotethys.

- MakaleAçık Erişim2015Scopus SJRQ1A fuzzy DEMATEL method to evaluate critical operational hazards during gas freeing process in crude oil tankers
Akyuz, Emre; Celik, Erkan
Gas freeing process in crude oil tanker ships is widely recognized one of the most hazardous aspects of shipboard operations. Although the process provides practical benefits to ship by removing the explosive or poisonous gases from the cargo tanks and raising the oxygen level up to 21 percent, the consequences of failure may cause serious damage to human health, marine environment and cargo. Therefore, the crew exercise utmost care and become aware of the potential hazards in gas freeing process. In this context, this paper provides a fuzzy DEMATEL (Decision Making Trial and Evaluation Laboratory) method to evaluate critical operational hazards in gas freeing process. While the DEMATEL method enables to identify and analyse the potential hazards of gas freeing process with respect to causal effect relation diagram, fuzzy sets deal with the uncertainty in decision-making and human judgements through the DEMATEL. Thus, the hybrid approach provides smart solution for safety practitioners to prevent critical hazards in gas freeing process. The results of the research will contribute to maritime safety at sea and prevention of environment pollution as well as loss of life on-board crude oil tankers ships. (C) 2015 Elsevier Ltd. Mutlu, Ekrem; Aydin, Seyit; Kutlu, Banu
The objective of this study was to evaluate the effects of copper sulfate (CuSO4(5 H2O)) on the Nile tilapia (Oreochromis niloticus). Median (LC50) and minimum (LC1O) lethal concentrations of copper sulfate on Nile tilapia were determined via acute tests. LC50 values of copper sulfate for the 48th, 72nd and 96th hours were 13.15, 12.95 and 12.85 mg/L, respectively. LC10 values of 12, 11 and 9.5 mg/L were obtained for similar exposure periods (48th, 72nd and 96th hours), respectively. The effects of 1.5 mg/L dose of copper sulfate on growth rate, gross clinical observations and blood biochemical parameters of Nile tilapia were evaluated after 35, 65 and 95 days of exposure periods. The differences of growth rates between control and copper sulfate-exposed groups were not significant (P>0.05) at the end of 35, 65 and 95 days. Continuous exposure of Nile tilapia to 1.5 mg/L concentration of copper sulfate in water solution for three periods (35, 65 and 95 days) decreased the total protein, albumin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), glucose (except the elevation in the exposed group at the end of 95 days period), magnesium (Mg+2), calcium (Ca+2), iron (Fe+2), potassium (K+), sodium (Na+1), chloride (Cl-) and phosphate values in serum. The cholesterol, low density lipid (LDL), triglyceride, globulin, creatinine, alkaline phosphatase (ALP), total bilirubin, direct bilirubin, uric acid and blood urea nitrogen (BUN) concentrations in the serum increased in copper sulfate-exposed fish.

Tanyol, Mehtap; Uslu, Gulsad; Yönten, Vahap
The aim of our research was to explore the most cost-efficient and optimal medium composition for the production of lipase from Pseudomonas fluorescens (NRLL B-2641) culture grown on sunflower oil cake (SuOC) by applying response surface methodology (RSM). The oil cake was used instead of carbon sources. Peptone, ammonium sulphate and the carbon source (SuOC) were the most important factors as it is obligatory for microbial growth. Subsequently, the optimum values for the carbon source, peptone and ammonium sulphate were found to be 11.10% (w/v), 1.18% (w/v) and 0.83% (w/v), respectively. Experiments carried out under optimum conditions revealed a maximum lipase activity of 10.8 U mL(-1), which was achieved after 48 h of fermentation. The obtained results were finally verified with batch experiments carried out under the optimum conditions evaluated and it was demonstrated that the SuOC from agro-industrial residue as substrates can be used as an inexpensive base (carbon source) for the production of lipase by P. fluorescens (NRLL B-2641).

Gentamicin is commonly used against gram-negative microorganisms. Its therapeutic use is mainly limited by nephrotoxicity. This study was aimed at evaluating the effect of rutin on oxidative stress, inflammation, apoptosis, and autophagy in gentamicin-induced nephrotoxicity in rats. The rats were treated with saline intraperitoneally (group I), 150 mg/kg of rutin orally (group II), 80 mg/kg of gentamicin intraperitoneally for 8 d (group III), or 150 mg/kg of rutin plus 80 mg/kg of gentamicin (group IV). The serum urea, creatinine, kidney malondialdehyde (MDA), and reduced glutathione (GSH) levels and superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) activity and protein concentration were measured, and renal histopathology analysis and immunohistochemical staining were performed. Rutin pretreatment attenuated nephrotoxicity induced by gentamicin by reducing the urea, creatinine, and MDA levels and increasing the SOD, CAT, and GPx activity, and the GSH levels. The rutin also inhibited inducible nitric oxide synthase (iNOS), cleaved caspase-3 and light chain 3B (LC3B), as evidenced by immunohistochemical staining. The present study demonstrates that rutin exhibits antioxidant, anti-inflammatory, anti-apoptotic, and anti-autophagic effects and that it attenuates gentamicin-induced nephrotoxicity in rats.
