- MakaleYalnız künye2010Scopus SJRQ3Effect of Potassium Sorbate on Some Microbiological Properties of Cokelek Stored at Different Temperatures
Öksüztepe, Gulsum; Ilhak, O. Irfan; Dikici, Abdullah; Çalicioglu, Mehmet; Patir, Bahri
In this study, the effect of potassium sorbate at 0.01%, 0.05% and 0.1% on the some microbiological (total aerobic mesophilic bacteria-IAMB, coliform bacteria, Lactobacillus-Leuconostoc-Pediococcus, Stapylococcus-Micrococcus. enterococcus and yeast-mold) and chemical (pH, acidity) properties of cokelek stored at 4 +/- 1 degrees C or 22 +/- 1 degrees C was investigated. It was seen that adding potassium sorbate at 0.01%, 0.05% and 0.1% to cokelek did not have an effect on numbers of TAMB, coliform bacteria, lactobacillus- leuconostoc- pediococcus, staphylococcus-micrococcus in cokelek stored at both 4 +/- 1 degrees C and 22 +/- 1 degrees C (P>0.05). However, adding potassium sorbate at 0.05% and 0.1% to cokelek had an effect on numbers of enterococci and yeast-mold in cokelek stored at 4 +/- 1 degrees C (P<0.05). It was also not observed significant difference in pH and acidity levels between control group and treatment groups containing potassium sorbate during storage period (P>0.05).

- MakaleAçık Erişim2010Scopus SJRQ3Effects of cypermethrin on antioxidant status, oxidative stress biomarkers, behavior, and mortality in the freshwater mussel Unio elongatulus eucirrus
Köprücü, Kenan; Yonar, Serpil Mise; Seker, Engin
In this study, the effects of cypermethrin on antioxidant status, oxidative stress biomarkers, behavior, and mortality in the freshwater mussel Unto elongatulus eucirrus were examined. Cypermethrin was applied at concentrations of 5, 10, 20, 40, 80, and 160 mu g/L, for 1, 24, 48, 72, and 96 h. With increasing cypermethrin concentrations, the mussels showed significantly (P < 0.05) increased lipid peroxidation (MDA), which might be associated with decreased levels of reduced glutathione (GSH) and catalase (CAT) activity in the digestive glands and gills of the mussels. Negative correlations were observed between the MDA and the GSH and CAT levels after cypermethrin exposure, indicating a protective role of GSH and CAT against lipid peroxidation, and suggesting the use of these antioxidants as a potential biomarker of toxicity associated with contaminant exposure in freshwater mussels. In addition, at 5 mu g/L, cypermethrin had no effect on the duration of the active and rest periods. All higher concentrations caused inhibition of the filtration activity by reducing the active periods and lengthening the rest periods. The active periods shortened as the cypermethrin concentrations increased, the reduction being 90% of the control at 160 mu g/L. Rest periods were longest (237% longer than the control) at 160 mu g/L. The 48, 72, and 96 h LC50 values of cypermethrin for mussels were determined as 96.50, 77.96, and 59.20 mu g/L, respectively (P < 0.05). The results indicate that the insecticide cypermethrin has a harmful effect not only on nontarget aquatic arthropods and fish, but also on mollusks, although the sensitivity of mussels is less than that of fish. Being a general toxicant for aquatic life, cypermethrin should be used with great caution in agriculture to protect natural waters from contamination.

- MakaleAçık Erişim2010Scopus SJRQ2The Nature of the Crustal Structure of the Eastern Anatolian Plateau, Turkey
Yilmaz, Ali; Yilmaz, Huseyin; Kaya, Cemal; Boztug, Durmus
The Eastern Anatolian Plateau (EAP) of Turkey, with an elevation ranging from 1700 to 2000 m, is located between the Eastern Pontide Arc to the north and the Arabian Platform to the south. In this region, pre-Maastrichtian tectonic units representing the crust crop out in only a few localities. As they are covered by Maastrichtian-Quaternary rock units, it is difficult to study the nature and mutual relationships of these pre-Maastrichtian tectonic units. The palaeotectonic units of the EAP comprise two different levels in the present study: (1) The lower level consists of platform-type carbonates and their metamorphic equivalents. These units may represent the Taurus Platform and its metamorphic equivalents. (2) The upper level consists of an ophiolitic-melange prism which is made up mainly of oceanic crust; the prism comprises a complex of ophiolite, ophiolitic melange, and fore-arc deposits. This upper unit represents a subduction-accretion prism and may have originated partly from the North Anatolian Suture to the north, and partly from the South-eastern Anatolian Suture to the south. Continental crustal rocks were thrust over by the ophiolitic melange prism; thus outcrops of them are scarce in the region as they are exposed in tectonic windows through the ophiolitic thrust sheets. The pre-Maastrichtian tectonic units of the EAP are blanketed by Maastrichtian to Quaternary volcanic and sedimentary rock units; these sequences include successive transgressive and regressive intervals and overlie the palaeotectonic units along a pronounced unconformity. Olistostromal units are abundant in the Eocene sedimentary units and were derived from the ophiolites and ophiolitic melange. The Maastrichtian-Quaternary cover is made up of collisional and post-collisional deposits across the whole region. Although the EAP has been experiencing considerable N-S compression, it has not been affected by significant crustal thickening because of the strike-slip tectonic regime that is dominant in the region. - MakaleYalnız künye2010Scopus SJRQ3The Milk Biochemical Parameters Having Economic Importance in Machine Milked Cows
Tekelioglu, O.; Cimen, M.; Bayril, T.
This research was aimed to investigate the effect of machine milking on milk biochemical components having economic importance. In the experiment, 10 Machine Milked (MM) and 10 suckled + Hand Milked (HM) cows were used. The milk fat rates were lower for MM animals in 1st, 2nd and 4th weeks (p<0.05). Protein and total solid rates of milks were similar for both groups during study. There was found positive correlation between milk fat and total solids (r = 0.53; p<0.05), but there were no significant correlations among other milk parameters. Suckling treatment (in BM group) to increase production of milk with maximum levels of milk fat is essential for achieving the economic benefits. Producers, who utilize the MM system during lactation should expect commercial milk to be low in fat content, which may affect cheese processing characteristics.

Yildirim, Numan; Yildirim, Nuran Cikcikoglu; Yildiz, Abdunnasir
In this study, the ability to decolorize two chemically different synthetic dyes (methylene blue and crystal violet) by Pleurotus eryngii strains collected from geographically different regions of Turkey was evaluated on agar plates in static liquid cultures. For all strains, growth and decolorization halos were determined on malt extract agar plates containing 0.4 and 0.6 g/L of crystal violet (CV) or methylene blue (MB). Each P. eryngii strain showed certain decolorization capacities and was able to decolorize both MB and CV under static liquid culture conditions, but not to the same extent. The presence of the dyes in static liquid culture reduced the biomass production of all strains in comparison with control culture growing in the medium without dyes. In solid medium, a positive correlation was found between growth halo and decolorization halo.

- MakaleYalnız künye2010Scopus SJRQ3THE ROLE OF METHYLATED ADRENOMEDULLIN IN ENZYMATIC ANTIOXIDANT DEFENSE RESPONSE TO COLD STRESS
Yildirim, Nuran Cikcikoglu; Yurekli, Muhittin
The aim of our study was to investigate the effects of methylated adrenomedullin (met-AdM) on some antioxidant enzymes, such as catalase (CAT), glutathione peroxidase (GSH-P-X) and superoxide dismutase (SOD) in rat kidney and heart tissues after cold stress treatment. Male Wistar rats were divided into 4 groups (control, met-AdM, cold stress and cold stress + met-AdM treatments). The rats were kept at room temperature, exposed to a 16/8-hours light/dark cycle. The rats were maintained at ad libitum (free) feeding habit. In met-AdM treated group, animals received intraperitoneal injection of met-AdM (2 mg/g body weight) once a day during a week. For the cold stress exposure, the rats were kept in separate cages at 10 degrees C for a week. Control group rats were kept at laboratory conditions. CAT, GSH-P-X and SOD enzyme activities were investigated in kidney and heart tissues. In kidney tissue compared to control, CAT and GSH-P-X enzyme activity was decreased but SOD was increased in met-AdM-treated group. In met-AdM+cold stress group, only the SOD enzyme activity was significantly increased but CAT and GSH-P-X decreased. SOD and GSH-P-X enzyme activities were increased by cold stress but CAT was decreased in kidney. In heart tissue, CAT and GSH-P-X activities were decreased but SOD was increased in met-AdM-treated group compared to control. All enzyme activities were decreased in met-AdM+cold stress group as well as by cold stress in heart, with regard to control. Cold stress can disrupt the balance in an oxidant/antioxidant system and cause oxidative damage to kidney and heart tissues by altering the enzymatic and non-enzymatic antioxidant status. We can suggest that met-AdM has possible roles in regulation of stress but we need further studies revealing responses of met-AdM in different tissues.

The aim of this study is to determine suitable conditions for egg disinfection of gilthead sea bream (Sparus aurata), red porgy (Pagrus pagrus), and common dentex (Dentex dentex) with glutaraldehyde, hydrogen peroxide and iodine. The eggs were disinfected with 200 ppm glutaraldehyde (T= 2, 4, 8, 16 min), 300 ppm hydrogen peroxide and iodine (T= 5, 10, 15, 20 min) at four different contact times. The hatching rate was determined in triplicates for each treatment. Also, bacteria colonies were counted on tryptic soy agar (TSA) and thiosulphate citrate bile salt sucrose agar (TCBS). At the end of the experiment, bacterial load and hatching rate were assessed together. For glutaraldehyde treatment, optimal hatching rates and bacterial colonies were estimated as CT (Disinfectant concentration in mg.l(-1)* exposure time in minute) 800 for sea bream eggs, CT 1600 for red porgy eggs and CT 800-1600 for common dentex eggs. Also, for hydrogen peroxide treatments, they were determined as CT 3000 for sea bream and red porgy and CT 4500 for common dentex. Finally, in iodine treatments, these parameters were measured as CT 4500-6000 for all species' eggs. However, in our study, it is clear that the glutaraldehyde showed the optimum disinfection effect against the microorganisms because the bacteria could be eliminated completely.

Atar, Hasan Huseyin; Ates, Mehmet
Four hundred shabbout fries (Tor grypus L. 1758), with average initial weight of 0.95+/-0.02 g and total average length of 4.10+/-0.05 cm, were fed for a period of 90 days commercial feed with addition of mannanoligosaccharide (MOS) and vitamin B-12. The shabbouts were divided into four different groups and fed by different feeds (Control, Group A with MOS, Group B with vitamin B-12, Group C with the combination of MOS + vitamin B-12). The effects of feeding through MOS and vitamin B-12 in different proportions on growth in view of live weight and total length, survival rate, feed conversion, condition factor and protein efficacy were studied. The best growth in view of live weight of 4.51+/-0.16 g and length of 7.71+/-0.08 cm was maintained through vitamin B-12 in Group B. As a consequence, the growth parameters of the shabbout fries fed through commercial feeds with the addition of MOS and vitamin B-12 in different doses under empirical conditions showed affirmative effects on the survival rate, feed assessment, condition factor and protein efficacy.

- MakaleYalnız künye2010Scopus SJRQ4Is it Possible to Transform Hatchery-Reared Abnormal Juveniles of Sea Bass (Dicentrarchus labrax L. 1758) into Normal Individuals?
Kayim, M.; Can, E.; Güner, Y.
Lordosis, which is one of the skeletal deformations, was frequently encountered in the culture of sea bass (Dicentrarchus labrax L. 1758) and it affects the biological performance and marketing image of the fish negatively. Thus it causes big economic losses. In larger natural environments, this abnormality in wild-caught individual is rare, whereas the case is the other way round in hatchery-reared sea bass. This problem can be caused by swim bladder malformations, environmental conditions, bacterial factors, water flow rate and swimming activity. However, many researchers reported that all of these factors cause these malformations in different regions of the skeletal system, commonly known as V-shaped curvatures. But in this study, it is not aimed to investigate the possible causes of the observed abnormalities. We focused on whether the abnormal individuals can be transformed into individuals with normal skeletal systems. In this study, a total number of 5636 juvenile individuals (0.09-1.93 g of live weight) were used. Lordosis rates were identified between the days 54 and 110 by randomly taking at least 30 samples from every group on a weekly basis, at triplicate. In this study, 100% of the individuals with this type of deformation were successfully transformed into individuals with the normal skeletal system in a large volume and rectangular ponds with semi-intensive rearing method.

Yildirim, Nuran Cikcikoglu; Yürekli, Muhittin; Yildirim, Numan
Purpose: The aim of this study is to investigate the compensating effects of adrenomedullin on liver tissue after cold stress treatment. Materials and Methods: Male Wistar rats were divided into four groups as control, adrenomedullin, cold stress and cold stress + adrenomedullin treated. Adrenomedullin was given by intraperitoneal injection (2 mg/g body weight) once a day for a week. For cold stress the rats were kept at 10 C for a week. Catalase, glutathione peroxidase and superoxide dismutase activities were measured in liver tissue. Results: Catalase activity was decreased significantly as compared to control in adrenomedullin treated group (p<0.05). There was no significant difference in superoxide dismutase activity (p>0.05). Glutathione peroxidase activity was increased in adrenomedullin treated group but this was statistically insignificant (p>0.05). All enzyme activities were decreased by cold stress (p<0.05). In Adrenomedullin+cold stress group, only the glutathione peroxidase activity was significantly decreased (p<0.05). Conclusion: It is concluded that cold stress induces decrease in catalase, superoxide dismutase and glutathione peroxidase activities in rat liver. Adrenomedullin treatment corrected only glutathione peroxidase activity while decreasing catalase and superoxide dismutase activities further. The discrepancy may be explained by different responses of antioxidant systems to oxidative stress. It can be concluded that oxidative factors may display unexpected effects on the regulation of glutathione peroxidase activities in the adrenonmedullin case.

Yildirim, Nuran Cikcikoglu; Yurekli, Muhittin
In this study, it was aimed to investigate the effects of chronic cold stress on adrenomedullin levels and tyrosine hydroxylase enzyme activities in some rat tissues. In this study, 12 female Sprague Dawley rats weighing 200 to 250 g were used. Rats were housed under diurnal lighting conditions (12-12 h) with free access to food and water. For this study, rats were divided into two groups, namely control group and cold stress treated group. Adrenomedullin levels were measured by high performance liquid chromatography. Tyrosine hydroxylase enzyme activities was spectrometrically measured. Tyrosine hydroxylase activity was found to be increased significantly (p<0.05) in adrenal medulla, hypothalamus and heart tissues depending on cold stress treatment. Adrenomedullin levels were determined to be decreased in the cold stress treatment group. The differences of between control and cold stress treatment groups were significant (p<0.05). The results suggested that adrenomedullin may play a possible role in adaptation to stress. Exposure to cold increases the synthesis and release of catecholamines. Tyrosine hydroxylase is the rate limiting step in the biosynthesis of catecholamines for this reason cold stress increased tyrosine hyroxylase enzyme activities. Further studies are needed to reveal correlation between adrenomedullin, tyrosine hydroxylase and cold stress.
