Accumulation and Toxicity of CuO and ZnO Nanoparticles Through Waterborne and Dietary Exposure of Goldfish (Carassius auratus)

Ates, Mehmet; Arslan, Zikri; Demir, Veysel; Daniels, James; Farah, Ibrahim O.

Dietary and waterborne exposure to copper oxide (CuO) and zinc oxide (ZnO) nanoparticles (NPs) was conducted using a simplified model of an aquatic food chain consisting of zooplankton (Artemia salina) and goldfish (Carassius auratus) to determine bioaccumulation, toxic effects, and particle transport through trophic levels. Artemia contaminated with NPs were used as food in dietary exposure. Fish were exposed to suspensions of the NPs in waterborne exposure. ICP-MS analysis showed that accumulation primarily occurred in the intestine, followed by the gills and liver. Dietary uptake was lower, but was found to be a potential pathway for transport of NPs to higher organisms. Waterborne exposure resulted in about a 10-fold higher accumulation in the intestine. The heart, brain, and muscle tissue had no significant Cu or Zn. However, concentrations in muscle increased with NP concentration, which was ascribed to bioaccumulation of Cu and Zn released from NPs. Free Cu concentration in the medium was always higher than that of Zn, indicating CuO NPs dissolved more readily. ZnO NPs were relatively benign, even in waterborne exposure (p0.05). In contrast, CuO NPs were toxic. Malondialdehyde levels in the liver and gills increased substantially (p<0.05). Despite lower Cu accumulation, the liver exhibited significant oxidative stress, which could be from chronic exposure to Cu ions. (c) 2014 Wiley Periodicals, Inc. Environ Toxicol 30: 119-128, 2015.

Determination and Health Risk Assessment of Heavy Metals in Drinking Water of Tunceli, Turkey

    Demir, V.; Dere, T.; Ergin, S.; Cakir, Y.; Celik, F.

Tunceli, Turkey is one of the small cities in the Eastern Anatolian Region with substantial natural, cultural and historical heritage that is suitable to be a slow city. Water sources are abundant in the area due to rain and heavy snowfall in the mountains, which seeps into the ground or flows into the rivers. Therefore monitoring and conservation of city's drinking water quality is one of the main issues in the city. The present study was conducted to investigate several heavy metals and cyanide, arsenic, and selenium concentrations of drinking water samples taken from 100 different locations in the city and towns of Tunceli, Turkey during the years 2011 and 2012. In three locations, high levels of copper was found with a range of 2.1-11.5 mg/L, and high levels of arsenic was detected in five locations with a range of 10.5-78.0 mu g/L, exceeding the action levels for copper and arsenic. For health risk assessment, the average daily dose and hazard quotient (HQ) were calculated. The region mean values of HQ were found above 1 for arsenic in the samples collected from Mazgirt, while HQ values were below 1 in the rest of the samples.

Use of acetylcholinesterase, glutathione S-transferase and cytochrome P450 1A1 in Capoeta umbla as biomarkers for monitoring of pollution in Uzuncayir Dam Lake (Tunceli, Turkey)

Yildirim, Nuran Cikcikogiu; Yildirim, Numan; Danabas, Durali; Danabas, Seual

Uzuncayir Dam Lake is one of the most important water bodies in Tunceli. It is polluted by domestic wastewaters together with natural contamination and pesticides, which have had cumulatively negative effects. This study analyzes the effects of pollution by using the fish species, Capoeta umbla as a sentinel species, with a biomarker approach. The approach comprehends a general biomarker of individual fish health, the condition factor and specific biomarkers of contaminant exposure such as glutathione S-transferase (GST) and cytochrome P450 1A1 (CYP1A1) and acetylcholinesterase (AChE) activity. The general water physico-chemical parameters were measured at each sampling sites during each fish-sampling period. Strong seasonal and locational variations were observed among selected markers. In conclusion, the multibiomarker approach used in the present study clearly revealed differences in the fish health among reference and potentially contaminated sites. The study shows that changes of biomarkers such as acetylcholinesterase, glutathione S-transferase and cytochrome P450 1A1 might be useful for the assessment of environmental contamination in the Uzuncayir Dam Lake (Tunceli, Turkey). Ecological risk assessment of heavy metals in surface sediments of northern littoral zone of Lake Cildir, Ardahan, Turkey

Kükrer, Serkan; Seker, Sebahat; Abaci, Z. Tugba; Kutlu, Banu

In this paper, the heavy metal levels (Cu, Pb, Zn, Ni, Mn, Fe, As, Cd, Cr, Hg), organic carbon, and chlorophyll degradation products were studied to prove their ecological effects in Lake CA +/- ldA +/- r, where fossil fuels are used as an energy source in the studied area for most of the year, and domestic waste from settlements is discharged directly into the lake. Sediment samples were collected from six sites on the northern shore of CA +/- ldA +/- r Lake, Turkey in November 2012. Enrichment (EF) and contamination factor (CF) values were determined, and Pollution Load (PLI) and Potential Ecological Risk (PER) indices were calculated. Average concentrations of heavy metals in the sediments were, in descending order, Fe > Mn > Zn > Ni > Cr > Cu > Pb > As > Cd > Hg, respectively. According to mean values, the source of these elements may be considered natural due to lack of enrichment in Cu, Pb, Zn, Ni, and Cr in the sediment samples. Regarding enrichment of As, Cd, Mn, and Hg, the highest EF belongs to Hg. PLI and PER values indicate there are moderate ecological risk in the lake.

Temporal Changes in Concentrations of Some Trace Elements in Muscle Tissue of Crayfish, Astacus leptodactylus (Eschscholtz, 1823), from Keban Dam Lake

Aksu, Onder; Adiguzel, Ragip; Demir, Veysel; Yildirim, Numan; Danabas, Durali; Seker, Sebahat; Can, Safak Seyhaneyildiz; Ates, Mustafa

Crayfish (Astacus leptodactylus Eschscholtz, 1823) is the native crayfish species in Turkey. It was exported regularly to Western Europe. In this study, bioaccumulation and temporal trends of some trace elements (arsenic: As, cadmium: Cd, copper: Cu, mercury: Hg, lead: Pb, and zinc: Zn) in edible abdomenmuscle of crayfish from Keban Dam Lake (Elazig, Turkey) were investigated for the 2006-2012 period. Sequence of metal concentration levels was Zn > Cu > Hg > Pb > Cd > As in muscle tissues. The highest concentration of Zn (21.69mg kg(-1)) was detected in 2006, while the lowest (4.35mg kg(-1)) in 2009. In general, it was found that the concentrations of trace elements investigated were lower than the maximum permissible limits of the food regulations of the Ministry of Food, Agriculture, and Livestock (MFAL), the Turkish Food Codex and Commission Regulation (EC). If the crayfish selected for the study are recognized as bioindicators of environmental pollution, then it is possible to conclude that the changes in studied trace elements concentrations in the Keban Dam Lake are being steady.

Occurrence and Assessment of Chemical Contaminants in Drinking Water in Tunceli, Turkey

    Demir, Veysel; Ergin, Sezai

The objective of this study was to analyze drinking water samples from 21 sites in the city center and seven municipalities of Tunceli, Turkey, in order to determine the presence of nitrate, nitrite, fluoride, bromate, pesticides, polycyclic aromatic hydrocarbons (PAHs), trihalomethanes (THMs), and some other chemicals. In all locations, the concentrations of chemicals investigated were below the permissible limits set by local and international organizations for drinking water. Low levels of nitrate (4.79 +/- 4.20 mg/L), fluoride (0.11 +/- 0.08 mg/L), and THMs (6.63 +/- 5.14 mu g/L) were detected in all locations. A low level of tetra, chloroethane, which is suspected to be a human carcinogen, was also detected in 8 locations in the range of 0.26-0.43 mu g/L. These contaminants may pose adverse health effects or minimum hazard due to long-term exposure. In all locations, bromate, benzene, total PAH, 1-2 dichloroethane, vinyl chloride, acrylamide, and epichloridine levels in drinking water samples were under detection limits.

Geo-environmental site investigation for Tunceli, Turkey municipal solid waste disposal site

Önal, Ayten Oztufekci; Demirbilek, Deniz; Demir, Veysel

The environmental and geological condition of a current solid waste disposal site in the city of Tunceli, Turkey was investigated. Improper geological structure and soil properties of this unsanitary solid waste storage site as well as leachate from this site caused significant pollution in air, soil, and surface water. Natural soil characteristics and geology of the site are presented in this paper. Petrographic properties of vulcanized rubber rocks composed of tuff and volcanic sandstone at the bottom of storage sites and the mineralogical composition of the base composed of these rocks were examined. Permeability of the natural soil and index properties were determined using soil mechanics experiments (moisture content, specific gravity, Atterberg limit tests, sieve analysis, hydrometer analysis, falling-head permeability test, and standard proctor test) conducted on disturbed samples collected from excavated sites and the relationship between mineralogical composition and soil hydraulic behavior was investigated. Natural soil samples of six different parts of the site indicated that, the soil class in four samples is silty sands, sand-silt mixtures, one is clayey sands, sand-clay mixtures and one is clayey silts with slight plasticity. The values obtained using permeability tests for subsoil samples vary from 10(-5) to 10(-8) cm/s. Low permeability (10(-7) and 10(-8) cm/s) at the south part of the site increases to 10(-5) cm/s at the natural soil of the North side. Extensively faulted and fissured volcanic sandstone levels at the site give potential to waste leachate to reach deep levels. The soil composition, hydraulic conductivity, topographic conditions, and other location criteria in the current solid waste disposal site showed that the current disposal site is not suitable for this purpose. Characterization and pollution potential assessment of Tunceli, Turkey municipal solid waste open dumping site leachates

Demirbilek, Deniz; Önal, Ayten Oztufekci; Demir, Veysel; Uslu, Gulsad; Arslanoglu-Isik, Hilal

Environmental monitoring of leachate quality from an open municipal solid waste dumping site in Tunceli, Turkey was studied in this research. The most commonly examined pollution parameters were determined on a seasonal basis. The annual average 5-day biological oxygen demand (BOD5) and chemical oxygen demand (COD) values of station points were measured as 70 and 425 mg/L, respectively, and also the average BOD5/COD ratio (a measure of biodegradability) was calculated as 0.20. The low ratio of biodegradability and slightly alkaline pH values in the leachate samples indicated that the site was characterized by methanogenic conditions. The mean ammonium-nitrogen (NH4 (+)-N) and corresponding phosphate (orthophosphate) values were assayed as 70 and 11 mg/L, respectively. The average solids content in the leachates was measured as 4,681 mg/L (total solids) and 144 mg/L (suspended solids). Very low concentrations of iron, manganese, copper, and zinc in the leachate samples were found and the concentration of cadmium was measured below detection limits. Excessive amount of nutrients and high organic and inorganic pollutant content in the leachates pose serious pollution potential to the environment. Since no drainage system or bio treatment exists in this open dumping site, high permeability of natural soil at the site and in the surrounding area and very fractured and crackled rocks under natural soil are indicators of high groundwater pollution potential in this site.

Bioaccumulation, Subacute Toxicity, and Tissue Distribution of Engineered Titanium Dioxide Nanoparticles in Goldfish (Carassius auratus)

Ates, Mehmet; Demir, Veysel; Adiguzel, Ragip; Arslan, Zikri

The increased use of nanosized materials is likely to result in the release of these particles into the environment. It is, however, unclear if these materials are harmful to aquatic animals. In this study, the sublethal effects of exposure of low and high concentrations of titanium dioxide nanoparticles (TiO2 NPs) on goldfish (Carassius auratus) were investigated. Accumulation of TiO2 NPs increased from 42.71 to 110.68 ppb in the intestine and from 4.10 to 9.86 ppb in the gills of the goldfish with increasing exposure dose from 10 to 100mg/L TiO2 NPs. No significant accumulation in the muscle and brain of the fish was detected. Malondialdehyde as a biomarker of lipid oxidation was detected in the liver of the goldfish. Moreover, TiO2 NPs exposure inhibited growth of the goldfish. Although there was an increase (8.1%) in the body weights of the goldfish for the control group, in the low and high exposure groups 1.8% increase and 19.7% decrease were measured, respectively. The results of this study contribute to the current understanding of the potential ecotoxicological effects of nanoparticles and highlight the importance of characterization of NPs in understanding their behavior, uptake, and effects in aquatic systems and in fish.

Column Studies on Effect of Phosphate Immobilization on Removal of Metals from Stormwater Runoff in Bioretention Cell

    Demir, V.; Bin-Shafique, S.

Batch studies indicated that low cost native materials such as loam soil, juniper mulch, hardwood mulch, pine mulch and sawdust performed efficiently as media in bioretention cell for removal of copper and lead from stormwater runoff. Juniper mulch performed best in batch studies was used in column studies. Column studies suggested that the presence of phosphate has a pronounced effect on metal immobilization. This is due to the formation of metal phosphates that have extremely low solubility. The remaining metals were adsorbed on bioretention media of the bioretention cell. Breakthrough adsorption and desorption curves for lead and copper were measured in presence and absence of phosphate immobilization. Copper reached a breakthrough curve in around 20 days and lead in 30 days. Column studies showed that in all samples addition of phosphate also decreased the leaching of lead and copper back to aqueous solution consistent with lead and copper immobilization.

Bioaccumulation of Copper and Zinc in Freshwater Crayfish (Astacus leptodactylus) Fed with Zebra Mussels (Dreissena polymorpha) Collected from Keban Dam Lake, Turkey

Aksu, Onder; Gulec, Azime Kucukgul; Demir, Veysel; Danabas, Durali; Yildirim, Numan; Ozcelik, Mehtap; Can, Erkan; Kocabas, Mehmet

This study is aimed to determine the bioaccumulation of copper and zinc in hemolymph of freshwater crayfish (Astacus leptodactylus) fed with zebra mussel (Dreissena polymorpha) collected from Keban Dam Lake. This is the first study on bioaccumulation of Cu and Zn in hemolymph of males and females of A. leptodactylus. The average concentrations of Cu and Zn in the hemolymph samples were found out as 0.54 and 35.07 ppm respectively for male and 0.42 and 15.59 ppm respectively for female control freshwater crayfish. The levels of Zn in the hemolymph of both male and female ones among control and diet groups were statistically significant (P < 0.05). The Cu levels among control and diet groups in female ones were statistically significant (P < 0.05) but not in the male ones (P > 0.05). The results of current study revealed that feed treatment with boiling was not effective at the levels of metal bioaccumulation in hemolymph. It was observed that this metal bioaccumulation is more related with sex differences.

Some Heavy Metals Accumulation in Tissues in Capoeta umbla (Heckel, 1843) from Uzuncayir Dam Lake (Tunceli, Turkey)

Ural, Mesut; Yildirim, Numan; Danabas, Durali; Kaplan, Olcay; Yildirim, Nuran Cikcikoglu; Ozcelik, Mehtap; Kurekci, Eylem Funda

Concentrations of metals were determined in the gills, liver, kidney, heart and muscle in Capoeta umbla caught from six stations from the Munzur River system. Metal concentrations in the tissues tended to vary significantly among stations (p < 0.05). Liver (Cu, 10.10 +/- A 0.23-23.03 +/- A 9.37 ppm; Zn, 14.67 +/- A 3.01-21.82 +/- A 2.39 ppm; Cd, 18.04 +/- A 4.56-52.69 +/- A 10.65 ppb and Fe, 28.87 +/- A 6.78-115.11 +/- A 34.87 ppm) and kidney (Cu, 1.80 +/- A 0.25-3.70 +/- A 0.62 ppm; Zn, 20.81 +/- A 0.37-29.36 +/- A 0.70 ppm; Cd, 132.06 +/- A 5.29-639.51 +/- A 20.14 ppb and Fe, 24.40 +/- A 1.98-59.39 +/- A 1.97 ppm) tissues showed higher metal concentrations than other tissues. It seems that metal contamination in the river is too high for the health of fish and the people who eat them. The geographical locations of catch, season, nature of diet, and the size of fish used for analyses might lead to different metal concentration in the same fish species.

Determination of Metal (Cu, Zn, Se, Cr and Cd) Levels in Tissues of the Cyprinid Fish, Capoeta trutta (Heckel, 1843) from Different Regions of Keban Dam Lake (Euphrates-Turkey)

    Danabas, Durali; Ural, Mesut

The aim of this study was to evaluate Copper (Cu), Zinc (Zn), Selenium (Se), Chromium (Cr) and Cadmium (Cd) concentrations in liver, muscle, gills and kidney tissues of Capoeta trutta collected from four sites of Keban Dam Lake, Turkey. The highest heavy metal level in all tissues was for Zn, while Cd was the lowest. The lowest heavy metal levels were generally found at Station 4 (Agin) for all tissues (p < 0.05). There were statistically significant differences among stations for Cu, Zn, Cr and Se in gills; for Cu, Zn, Se and Cr in liver and kidney; and Cu, Zn and Cr in muscle (p < 0.05). Cadmium concentrations did not differ between sites for any of the tissues. Turkish Food Codex sets the maximum limits of Zn, Cu and Cd as, 50, 20 and 0.05 mg kg(-1) dry weight, respectively, in the muscle of fish used for human consumption. In our study, the level of Zn was higher than Turkish permissible limits only at Station 3 (Guluskur). Cadmium levels were much higher than permissible limits at three stations [S1 (Pertek), S2 (Kockale) and S3], whereas Cu levels were within permissible limits in all stations.

Ozone Disinfection of Eggs from Gilthead Seabream Sparus aurata, Sea Bass Dicentrarchus labrax, Red Porgy, and Common Dentex Dentex dentex

Can, Erkan; Karacalar, Ulviye; Saka, Sahin; Firat, Kursat

The risk of fish pathogen transmission via eggs can be reduced by disinfection in ozonated seawater. The aim of this study was to determine the suitable conditions for ozone disinfection of the eggs of gilthead seabream Sparus aurata, sea bass Dicentrarchus labrax, red porgy Pagrus pagrus, and common dentex Dentex dentex. The eggs were disinfected with a concentration (C) of 0.5 mg of ozone/L of water at four different exposure times (T = 2, 4, 8, and 16 min). The hatching rate was determined in triplicate for each treatment. Bacterial colonies were counted on tryptic soy agar and thiosulfate-citrate-bile salts-sucrose agar. At the end of the experiment, bacterial load and hatching rate were assessed together to determine the optimal ozone treatment values, which were estimated in CT units (i.e., C [= 0.5 mg/L] x T [min]). Optimal values were CT 2-4 (T = 4-8 min; 18 degrees C) for gilthead seabream and red porgy, CT 2 (T = 4 min; 18 degrees C) for common dentex, and CT 4 (T = 8 min; 15 degrees C) for sea bass.

DETERMINATION OF TRACE METALS IN PLANT LEAVES AS BIOMONITOR OF POLLUTION EXTENT BY A SENSITIVE STAT-AAS METHOD

    Kaya, Gokce; Yaman, Mehmet

The aim of this study is to assess the potential of bioindicator/biomonitor plants to determine pollution extent of toxic metals. Lead, cadmium, and copper concentrations in the plants including Platanus sp., Olea europaea L., Vitis vinifera L., and Armeniaca sp. taken from the industrial area of the city of Gaziantep, Turkey, and control location were determined by using STAT-atomic absorption spectrometry. Lead concentration in leaves of Platanus sp. taken from 50m around the battery plant was found to be higher than 3,000 mg kg(-1). The observed Pb concentration in this plant was 40 times higher than that in Armeniaca sp. grown in the same location. The limits of detection for plants were found to be 0.050 mg kg(-1) for Pb and Cu and 0.005 mg kg(-1) for Cd. It was concluded that the Platanus sp. plant can be considered as both biomonitor and hyperaccumulator.
