Pala, Ayşegül; Eroğlu, Mücahit; Mişe Yonar, Serpil; İspir, Ünal; Yonar, Muhammet Enis

This study examined the adverse effects of sublethal concentrations of cadmium (Cd) and the potential of curcumin (CUR) to mitigate Cd-induced toxicity. We administered 1.5 and 3 mg/L of Cd and 100 mg/kg of CUR to fish (Cyprinus carpio) separately and in combination over 14 days. At the end of the experiments, various haematological and immunological parameters, oxidative stress levels and antioxidant parameters of fish were measured, and the differences between the groups were evaluated. Simultaneous CUR treatment with Cd reversed the changes in haematological parameters in fish, increased immune responses, reduced oxidative stress and normalized the impaired antioxidant enzyme activities. In summary, our study showed that dietary supplementation with CUR may be an effective strategy to reduce the toxic effects of Cd in carp.

Curcumin positively affects performance during exercise and subsequent recovery. However, curcumin has limited bioavailability unless consumed in larger doses. In the current study, we examined the impact of a new formulation of curcumin, Next‐Generation Ultrasol Curcumin (NGUC), which is relatively more bioavailable than natural curcumin on exhaustion time, grip strength, muscle damage parameters, and serum and muscle proteins. A total of 28 rats were randomly grouped as control (C, non‐supplemented), exercise (E, non‐supplemented), E+NGUC100 (supplemented with 100 mg/kg BW NGUC), and E+NGUC200 (supplemented with 200 mg/kg NGUC). Grip strength and exhaustion time were increased with NGUC supplementation (p < 0.0001). Creatine kinase (CK), lactate dehydrogenase (LDH), lactic acid (LA), myoglobin, malondialdehyde (MDA) concentrations were reduced in serum, and muscle tissue in NGUC supplemented groups (p < 0.05). In contrast, NGUC supplementation elevated the antioxidant enzyme levels compared to the non‐supplemented exercise group (p < 0.01). Additionally, inflammatory cytokines were inhibited with NGUC administration (p < 0.05). NGUC decreased PGC‐1α, p‐4E‐BP1, p‐mTOR, MAFbx, and MuRF1 proteins in muscle tissue (p < 0.05). These results indicate that NGUC boosts exercise performance while reducing muscle damage by targeting antioxidant, anti‐inflammatory, and muscle mass regulatory pathways.

Palliative effect of curcumin on doxorubicin-induced testicular damage in male rats

Aksu, Emrah Hicazi; Kandemir, Fatih Mehmet; Yıldırım, Serkan; Küçükler, Sefa; Dörtbudak, Muhammed Baheeddin; Çağlayan, Cüneyt; Benzer, Fulya

This study aimed to investigate the effect of curcumin (CUR) on doxorubicin (DOX)-induced testicular damage in male rats. Thirty-five adult male Wistar rats were used. Control group was received saline for 7 days. CUR group received CUR for 7 days. DOX group received single dose DOX on the 5th day. DOX+ CUR-100 group received 100 mg/kg/day CUR for 7 days and DOX injection on the 5th day. DOX + CUR-200 group received 200 mg/kg/day CUR for 7 days and DOX injection on the 5th day. DOX treatment decreased in sperm motility rate, live sperm percentages, cellular antioxidants, and increased malondialdehyde (MDA) levels, necrosis, degenerations, and slimming in seminiferous tubules, and DNA damages in testes by inducing oxidative stress. CUR treatment mitigated significantly these side effects when compared with DOX group in a dose-dependent manner. In conclusion, CUR treatment can be used in the mitigation of DOX-induced testicular toxicity.

Chemoprotective effects of curcumin on doxorubicin-induced nephrotoxicity in wistar rats: by modulating inflammatory cytokines, apoptosis, oxidative stress and oxidative DNA damage

Benzer, Fulya; Kandemir, Fatih Mehmet; Kucukler, Sefa; Comaklı, Selim; Caglayan, Cuneyt

Doxorubicin (DXR) is one of the most important chemotherapeutic agent. However, nephrotoxicity reduces its clinical utility in humans. The aim of the study was to investigate protective effects of curcumin (CMN) against DXR-induced nephrotoxicity. Rats were subjected to oral treatment of CMN (100 and 200 mg/kg body weight) for 7 days. Nephrotoxicity was induced by single intra peritoneal injection of DXR (40 mg/kg body weight) on the fifth day and then the experiment was terminated on the eighth day. Nephroprotective effects of CMN were associated with decrease in serum toxicity markers and increase in antioxidant enzyme activities. CMN was able to reduced the levels of inflammatory markers such as TNF-α, NF-κB, IL-1β, iNOS and COX-2 in the rats. It also reduced the expressions of apoptotic marker including caspase-3, and oxidative DNA damage marker including 8-OHdG. Collectively, these findings indicated that CMN protect against DXR-induced nephrotoxicity.

Curcumin ameliorates doxorubicin-induced cardiotoxicity by abrogation of inflammation, apoptosis, oxidative DNA damage, and protein oxidation in rats

Benzer, Fulya; Kandemir, Fatih Mehmet; Ozkaraca, Mustafa; Kucukler, Sefa; Caglayan, Cuneyt

Doxorubicin (DXR) is a highly effective drug for chemotherapy. However, cardiotoxicity reduces its clinical utility in humans. The present study aimed to assess the ameliorative effect of curcumin against DXR-induced cardiotoxicity in rats. Rats were subjected to oral treatment of curcumin (100 and 200 mg/kg body weight) for 7 days. Cardiotoxicity was induced by single intraperitoneal injection of DXR (40 mg/kg body weight) on the 5th day and the rats sacrificed on 8th day. Curcumin ameliorated DXR-induced lipid peroxidation, glutathione depletion, decrease in antioxidant (superoxide dismutase, catalase, and glutathione peroxidase) enzyme activities, and cardiac toxicity markers (CK-MB, LDH, and cTn-I). Curcumin also attenuated activities of Caspase-3, cyclooxygenase-2, inducible nitric oxide synthase, and levels of nuclear factor kappa-B, tumor necrosis factor-α, and interleukin-1β, and cardiac tissue damages that were induced by DXR. Moreover, curcumin decreased the expression of 8-OHdG and 3,3′-dityrosine. This study demonstrated that curcumin has a multi-cardioprotective effect due to its antioxidant, anti-inflammatory, and antiapoptotic properties.

Compensatory effects of curcumin on cisplatin-induced toxicity in rabbit testis

    Kandemir, F. M.; Benzer, F.; Yildirim, N. C.; Ozdemir, N.

Curcumin, a widely used spice and colouring agent in food, has been shown to possess potent antioxidant, antitumor promoting and anti-inflammatory properties in vitro and in vivo. The present study was designed to investigate the protective effects of curcumin on changes in the levels of lipid peroxidation and endogenous antioxidants induced by cisplatin (cis-diamminedichloroplatinum II, CDDP) in the testiclular tissue of rabbits. 18 healthy male New Zealand white rabbits were equally divided into three groups of six rabbits each, control, cisplatin, and cisplatin+curcumin. The degree of protection produced by cisplatin was evaluated by determining the level of malondialdehyde (MDA) and glutathione (GSH), the activity of catalase (CAT), glutathione peroxidase (GSH-Px), were estimated from testes homogenates. MDA levels were increased with cisplatin compared to control but in cisplatin+curcumin group, MDA levels were found to be lower than cisplatin group (p<0.05). The activity of CAT and GSH-P-X was decreased in cisplatin and cisplatin+curcumin groups compared to control (p<0.05). In the case of cisplatin+curcumin CAT and GSH-Px activity were increased compared to cisplatin group (p<0.05). GSH levels were decreased with cisplatin but administration cisplatin+curcumin increased the levels of GSH comared to cisplatin group (p<0.05). In the present study, co-administration of curcumin with cisplatin prevented the damage to testes induced by this drug and may be considered as a potentially useful candidate in the combination chemotherapy with cisplatin.
