Then, superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) activities, malondialdehyde (MDA) and glutathione (GSH) level responses of D. polymorpha organism exposed to GCH sublethal concentrations for 24 and 96 hours were determined. In addition to biochemical biomarkers, histopathological responses were evaluated. In conclusion, this study demonstrated the abilities of GCH pesticides to induce oxidative stress. Moreover, MDA, GSH levels, SOD, CAT, GPx activities and histopathological responses could be used as an effective biomarker in D. polymorpha.
- MakaleAçık Erişim2025Determination of Oxidative Stress Responses in Dreissena polymorpha Exposed to Rare Earth Elements (Terbium, Lanthanum, Gadolinium and Praseodymium) with Temperature
Serdar, Osman; Aydın, Ayşe Nur
The continuous development of industry pushes people to search for new resources, and for this reason, the usage areas of Rare Earth Elements (REEs) are increasing day by day. Increasing concentrations of REEs, as a result of increased use, create pollution in the environment and harm living organisms. This pollution interacts with increasing temperature and causes more negative synergistic effects of the pollutant in the environment and in the living body. In this study, sublethal concentration values were determined by literature review and the concentration value was determined as 125 mg/L. In the present study tt was aimed to investigate some oxidative stress and antioxidant responses of Terbium, Lanthanum, Gadolinium and Praseodymium REEs in Dreissena polymorpha at 125 mg/L concentration at 3 different temperatures (16, 18, 20 0C) with biomarkers. For this purpose, 24 and 96 hour experimental trial design was created and 7 D. polymorpha were used in each trial group, and the application experiments were carried out with 3 replications. The samples at the end of the experimental phase were stored at -80 degrees Celsius until they were analyzed. In this study, Superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) enzyme activities and glutathione (GSH) and Thiobarbituric acid (TBARS) level biomarker responses were determined by ELISA test microplate reader. CAYMAN brand SOD (Catalog No 706002), CAT Catalog No 707002) and GPx (Catalog No 703102), GSH (Catalog No 703002) and TBARS (Catalog No 10009055) were used in the study. SPSS 24.0 package program one-way ANOVA (Duncan 0.05) was used for the evaluation of biochemical analyzes. According to the study data, statistically significant decreases were observed in SOD and CAT activities in the oxidative stress responses of REEs on D. Polymorpha with increasing temperature, while there was no significant change in GPx activities. It was determined that there were increases in TBARS levels and decreases in GSH levels. It is thought that the temperature factor, application concentration and application time are effective in the formation of these changes. It can be said that temperature change and pollutants cause oxidative stress in organisms and cause cell damage.

İncele - MakaleAçık Erişim2025Molecular Identification of Anaerobic Gut Fungi Isolated from Mountain Goats
Kar, Bülent; Elinç, Cemal
Anaerobic Gut Fungi (AGF) are found in the Neocallimastigomycota phylum, and it is known that these fungi play an important role, especially in the digestive system of herbivores, as they are the main colonies in the consumption of feed. AGFs provide digestion by breaking down plant tissues with the help of lignocellulosic enzymes they secrete. These enzymes have also been used in the food and feed industry in recent years. In this respect, fungi isolated and identified from different ruminant animals are valuable. In this study, fungi were isolated from mountain goats living in the wild in the Tunceli region and identified morphologically and molecularly. The ITS region of ap-proximately 1500 bp was used in molecular identification. It was determined that 9 isolated fungi belonged to the genera Orpinomyces sp. (AGF-5), Feramyces sp. (AGF-8,10) and Neocallimastix sp. (AGF-1,2,3,4,7,11). Further studies are needed to determine the microbial population in wild ruminant animals.

- MakaleAçık Erişim2025A comparison of cnn and transformer-based models for algal species classification
Ertarğın, Merve; Şimşek, Gökçe Kendirlioğlu
In this study, the classification of Spirulina platensis, Tetraselmis suecica, and Porphyridium cruentum algal species was conducted using RegNetY-008, ConvNeXt-Tiny, and ViT-Tiny models. The experiments were performed on a small and imbalanced dataset, where all models achieved an accuracy of 100%. To assess model performance, training and validation losses were analyzed, confirming the absence of overfitting. Furthermore, Grad-CAM was applied to visualize the decision-making processes of the models. The results indicate that although all models demonstrated high accuracy, the ViT-Tiny model exhibited the most interpretable visualizations by effectively focusing on algal cells. This study highlights the capability of transformer-based models to achieve high performance even on small and imbalanced datasets, offering a promising alternative for biological image analysis.

İncele - MakaleAçık Erişim2024Examining Pre-Service Science Teachers' Personal and Enacted Pedagogical Content Knowledge About Seasons
Kılıç, Aygün
This study examined pre-service science teachers’ pedagogical content knowledge (PCK) about seasons. The framework of this study was based on the Refined Consensus Model (RCM), and pre-service teachers’ topic-specific PCK was also discussed in two dimensions: personal PCK (pPCK) and enacted PCK (ePCK), which are included in this model. The pPCK of the pre-service teachers represents their declarative PCK, and their ePCK represents their dynamic PCK. This qualitative descriptive research included 18 pre-service science teachers. In the study, knowledge of pre-service teachers’ pPCK was collected in vignette-based individual semi-structured interviews, and data on their competencies regarding their ePCK, which they revealed in a real classroom environment, were collected through field observations and teaching video recordings. The study’s qualitative data were evaluated using two separate rubrics developed by the author using the literature. Accordingly, it was determined that the pre-service teachers who participated in the research had limited pPCK and ePCK on the seasons and that their topic-specific pPCK was partially better quality than their topic-specific ePCK. Moreover, pre-service teachers’ ePCK was observed to be different from their pPCK, but their ePCK was based on their pPCK. In this context, it was seen that the results of this research are consistent with RCM.

İncele - MakaleAçık Erişim2024Effect of Dietary Zinc on the Antioxidant Parameters of Juvenile Common Carp (Cyprinus carpio)
Aksu, Önder; Harlıoğ, Muzaffer Mustafa; Mustafa, Shokri; Batool, Zahra
Zinc (Zn) is an essential micro mineral needed for the proper growth and immune function of fish. This investigation was designed to examine the antioxidant role of a fortified diet with different Zn levels in the muscle and liver tissues of carp fry. A four-iso-nitrogen (35% crude protein) practical diet was produced that included graded levels of dietary zinc sulphate as a nutritional zinc resource in the fundamental diet supplemented with increased zinc levels (T1, control, 85 mg Zn kg-1, T2 105 mg Zn kg-1, T3, 125 mg Zn kg-1 and T4, 145 mg Zn kg-1). Even though the SOD and CAT analysis results did not show a linear increase in the increasing Zn ratio in the diets, higher values were obtained compared to the control groups. SOD highest values in T3 for the liver (0.713 ± 0.220 U/ml) and T1 for muscle (0.751 ± 0.144 U/ml), CAT values were highest in T2 for the liver (0.849 ± 0.115 nmol/dk/m) and T2 for muscle (1.059 ± 0.148 nmol/dk/m) was obtained. MDA values were completely higher for the muscle than for the control group, and for the liver, a lower value was obtained in the T2 trial group than in the control group (1.671 ± 0.230 µM). The results of the study showed that Zn contributed significantly to the nutrition of carp fish. It can be concluded that the findings of SOD and CAT analysis endorse the positive contributions of using 105 mg Zn in the diets to promote the antioxidant defense of juvenile carp fish.

The highest trace element accumulated by Pseudevernia furfuracea (21.7±1.0 mg/kg; 75%), Rhizoplaca chrysoleuca (31.9±1.6 mg/kg; 61%), Umbilicaria vellea (16.3±0.8 mg/kg; 59%), Aspicilia calcarea (77.9±3.8 mg/kg; 88%), and Cetraria islandica (22.7±1.1 mg/kg; 75%) was determined as Sr. The highest CFs in lichens investigated were calculated for Sr, Ta, and Li. PLI values for Cetraria islandica, Aspicilia calcarea, and Umbilicaria vellea were greater than 1. As a result, it has been proven that these lichen species can be used as good biomonitors of pollution.
