Determination of Metal (Cu, Zn, Se, Cr and Cd) Levels in Tissues of the Cyprinid Fish, Capoeta trutta (Heckel, 1843) from Different Regions of Keban Dam Lake (Euphrates-Turkey)

    Danabas, Durali; Ural, Mesut

The aim of this study was to evaluate Copper (Cu), Zinc (Zn), Selenium (Se), Chromium (Cr) and Cadmium (Cd) concentrations in liver, muscle, gills and kidney tissues of Capoeta trutta collected from four sites of Keban Dam Lake, Turkey. The highest heavy metal level in all tissues was for Zn, while Cd was the lowest. The lowest heavy metal levels were generally found at Station 4 (Agin) for all tissues (p < 0.05). There were statistically significant differences among stations for Cu, Zn, Cr and Se in gills; for Cu, Zn, Se and Cr in liver and kidney; and Cu, Zn and Cr in muscle (p < 0.05). Cadmium concentrations did not differ between sites for any of the tissues. Turkish Food Codex sets the maximum limits of Zn, Cu and Cd as, 50, 20 and 0.05 mg kg(-1) dry weight, respectively, in the muscle of fish used for human consumption. In our study, the level of Zn was higher than Turkish permissible limits only at Station 3 (Guluskur). Cadmium levels were much higher than permissible limits at three stations [S1 (Pertek), S2 (Kockale) and S3], whereas Cu levels were within permissible limits in all stations.

Can, Erkan; Kurtoglu, Ilker Zeki; Benzer, Fulya; Erisir, Mine; Kocabas, Mehmet; Kizak, Volkan; Kayim, Murathan; Çelik, Huseyin Tugrul

The objective of the present work is to examine the effects of different doses (0, 10, 20 and 40 g kg(-1) fish diet mass) of kefir on growth performance and oxidant-antioxidant status in the blood and liver tissues of Coruh trout, Salmo coruhensis, in different periods (2 and 3 months). In this study, survival was more than 88.3%, and irrespective of dietary kefir levels at the end of the study. There were no significant differences in SGR (Specific growth rate), FCR (Feed conservation rate), CF (Condition factor) among fish fed diets with 0, 10, 20 and 40 g kg(-1) kefir, however, these growth values were significantly different in terms of durations between 2-month and 3-month treatment (P<0.05). Our results indicated that TAS (Total Antioxidant Status) and TOS (Total Oxidant Status) in blood tissues reduced with kefir treatment at the end of third month (P<0.05). Malondialdehyde (MDA) levels in liver decreased in all groups compared with control group and a considerable extent decrease was observed in 40 g dose of kefir treatment at the end of third month. The data obtained from this experiment indicated that the same dose of kefir was more effective at the end of 3-month treatment than 2-month treatment (P<0.001). Although there was no statistical difference among groups, an increase in the glutathione peroxidase (GSH-Px) enzyme activity was observed in all groups compared to control groups. While catalase (CAT) activity decreased in all groups compared to control group at the end of second month (P<0.01), the decrease was insignificant level at the end of third month. In glutathione (GSH) levels, statistical differences were no observed in all groups compared to the control group with 2-month treatment while there were significant increases with 3-month treatment (P<0.001). It was concluded that kefir could play an antioxidant role and its effectiveness depended on dosage and time of application in Coruh trout, S. coruhensis.

Investigation of Yersinia ruckeri Infection in Rainbow Trout (Oncorhynchus mykiss Walbaum 1792) Farms by Polymerase Chain Reaction (PCR) and Bacteriological Culture

Seker, Engin; Karahan, Murat; Ispir, Unal; Çetinkaya, Burhan; Saglam, Naim; Sarieyyüpoglu, Mustafa

This study has been carried out to investigate the presence of Yersinia ruckeri, the causative agent of Enteric Red Mouth (ERM) disease, in rainbow trout fish breeding farms located in Elazig and Malatya provinces in Eastern Turkey. For this purpose, bacteriological culture and a specific polymerase chain reaction (PCR) have been compare with examine blood and intestine samples collected from adult and young rainbow trout fish as well as water samples collected from all the farms. Y. ruckeri was isolated and identified from eight of the 17 fish farms giving a proportion of 52.9% at fish farm. The disease was noted in five of the rainbow trout farms located in Elazig and three of the 11 rainbow trout farms located in Malatya. The agent was detected in five of the six young fish breeding farms. The isolation percentages of the causative agent at fish level were calculated as 25.7% (131/510) in adult and 31.7% (19/60) in young fish, giving an overall proportion of 26.3% (150/570). All the isolates have been successfully identified as Y ruckeri by species-specific PCR. No isolation could be made from water samples. It is therefore concluded that Y. ruckeri infection poses a significant threat to the fish farms in the study area and necessary precautions should urgently be taken in order to minimize economical losses due to ERM disease. A specific PCR can be utilized successfully in aquaculture for rapid identification of bacterial agents which will help to prevent spread of infectious diseases and will therefore contribute to the productivity of fishery sector.

Isoflavonoid production by soy plant callus suspension culture

    Gueven, Alper; Knorr, Dietrich

Production of aglycone type isoflavonoids, Genistein and Daidzein, and p-glycosidic type isoflavonoids, Genistin and Daidzin, by the soy plant callus suspension culture was investigated in a batch system. Genistein and Daidzein concentrations increased during the exponential growth phase of the biomass. Genistin and Daidzin concentrations were not directly related with biomass production. Methyl jasmonate addition to the growth medium (0.1 kg/m(3)) was found to increase isoflavonoid biosynthesis by 5-6 times. Isoflavonoid production in the 7 days old culture increased with pulsed electric field (PEF) application at 1.6 kV, and aglycone forms were influenced to a greater extent. (C) 2010 Elsevier Ltd. Status of freshwater crayfish (Astacus leptodactylus) in Iran

    Karimpour, M.; Harlioglu, M. M.; Aksu, O.

Iranian native crayfish includes one crayfish species, Astacus leptodactylus, with two subspecies, A. l. leptodactylus (lives in freshwater) and A. l. eichwaldi (lives in the Caspian Sea with 12 ppt salinity). Only A. leptodactylus is commonly distributed in Iranian water resources. Nearly 80 years ago local fishermen started to catch Anzali Lagoon crayfish and sold them to foreigners and embassy employees. The consumption of crayfish in Iran is very low, approximately 1 t annually. The first commercially significant crayfish harvest in Iran was 11.3 t from Anzali Lagoon in 1985. It was carried out by a Turkish company. In 1985, the introduction of A. leptodactylus was started into suitable Iranian freshwaters. For example, crayfish from Anzali Lagoon were introduced into Arass water reservoir in 1985. Today, Arass water reservoir is the main resource of crayfish in Iran. There was no further crayfish harvest in Iran until 1993. In that year, a private Iranian company began harvesting the Iranian crayfish and exporting them to European countries. In 1993, 3.2 t of crayfish were exported from Iran. Although there have been fluctuations between years, the export of crayfish in Iran increased remarkably after 1996. The successful results of crayfish introductions gave rise to this increase. In 2003, the export of A. leptodactylus from Iran reached the maximum level (216 t). The commercial value of exported crayfish between 2000 and 2009 varied from 1.5-2.5 million US$ annually and its amount was 106-211.5 t. However, there was a reduction in the export of A. leptodactylus from Iran after 2003, to just above 100 t. In conclusion, in order to increase crayfish production in Iran, introduction of A. leptodactylus into suitable water resources, and management, conservation and monitoring of present crayfish populations should be carried out.

Comparison of the survival and growth performance in rainbow trout (Oncorhynchus mykiss) and brown trout (Salmo trutta fario) fry

Kizak, Volkan; Guner, Yusuf; Turel, Murat; Can, Erkan; Kayim, Murathan

Rainbow trout is an intensively cultured species because of their more cultivable character than brown trout. Culture of brown trout could not be expanded due to low growth performance of brown trout compared to rainbow trout. This experiment was conducted in a commercial trout farm, Aegean area (Turkey). Growth performances and survivals of rainbow and brown trouts from fry to fingerling was observed for 155 days. Growth performances, feed conversion rates (FCR) and survivals were determined. Initial weights of rainbow and brown trouts were 0.1 +/- 0.01 g. Final weights were 26.5 +/- 5.19 and 12.97 +/- 2.74 g, respectively at the end of the experiment. Survival and FCR of rainbow and brown trouts were 83.9, 80 and 0.59, 0.61% respectively. As a result, there is a similarity between these two trout species in point of survivals and FCR's although, growth performance was obviously better than brown trout in early development of rainbow trout.

Antibacterial Effects of Some Fermented Commercial and Homemade Dairy Products and 0.9% Lactic Acid against Selected Foodborne Pathogens

    Yesillik, S.; Yildirim, N.; Dikici, A.; Yildiz, A.

In this study, the antibacterial effects of homemade yoghurt, commercial yoghurt, commercial kefir and probiotic yoghurt on viability of Salmonella typhimurium (ATCC 14028), Escherichia coli (ATCC 25922), Pseudomonas aeruginosa (ATCC 27853) and Staphylococcus aureus (ATCC 25923) was investigated by using disc diffusion method. Antibaterial effects of the samples were tested at 24, 48 h and the 7 day intervals. Homemade yoghurt showed the maximum antibacterial effect against the pathogens. It was determined that the most sensitive pathogenic bacteria to dairy products were Salmonella typhimurium whereas the least sensitive pathogen was Pseudomonas aeruginosa. Comparing to the antimicrobial effect of 0.9% lactic acid which was used as positive control, our results concluded that the bacteria found in fermented dairy products have an antibacterial activity and this activity is not caused by only lactic acid.

Sequential and Selective Extraction of Copper in Different Soil Phases and Plant Parts from Former Industrialized Area

Kaplan, Olcay; Kaya, Gokce; Yaman, Mehmet

Copper (Cu) is an essential trace element for all living organisms; however, excess amounts in soil, plants, and food have negative impacts on the environment and human health. The aim of this study was to determine Cu levels in different chemical phases of soils and the relationship between Cu levels in soil phases and concentrations in plants. Soils and plants grown in these soils from an industrialized area in Turkey were analyzed using a selective and four-stage sequential extraction procedure. Copper levels in exchangeable fractions were found up to 658 mg kg(-1) while total levels were in the range of 133-5609 mg kg(-1). Copper concentrations in plant parts (roots and stem) were in the range of 2.6-240 mg kg(-1). The exchangeable forms of Cu were in the range of 3-22% of total Cu concentrations. The relationships were observed between soil Cu and Rumex plant Cu, and soil Cu and root of Brassicasea plant Cu.

Bioconversion Efficiencies of Lignocellulosic Soy Stalk by Pleurotus eryngii Strains

    Yildirim, Numan; Yildiz, Abdunnasir

The bioconversion of soy stalk (SS) lignocellulose by three different starins of Pleurotus eryngii from various origins by solid state fermentation was studied during 40 days. The effect of rice bran ratio, 5% or 10% (w/w), on lignin degradation and laccase activity were measured througout the fermentations while, total crude protein and C/N ratio of the remaining substrate were determined at the end of the fermentations. The degradation of lignocellulose was found by the determination of acid soluble lignin (Klason lignin) content. All the strains degraded lignin with different extents. The amount of lignin degradation was decreased by the addition of the higher quantities of rice bran (SS+10% RB). The highest lignin degradation was obtained as 24.43% by P eryngii var. ferulae (E) and the highest protein level was found to be %13.32 by P eryngii (H). We determined that the laccase activity was generally decreased by the addition of the higher quantities of rice bran (10%). Maximum laccase activity (204.39 U/L) was obtained by P. eryngii (E) on SS + 5% RB at the end of the fermentation. This study showed that, after fungal treatment soy stalk can be used as a feed for ruminants with its higher feeding value.

Acrylamide concentrations in grilled foodstuffs of Turkish kitchen by high performance liquid chromatography-mass spectrometry

Kaplan, Olcay; Kaya, Gokce; Ozcan, Cemile; Ince, Muharrem; Yaman, Mehmet

For over ten years, there has been a considerable interest in determination of acrylamide in foodstuffs. It was known that both protein-rich and carbohydrates-rich foods cooked at high-temperatures can cause acrylamide formation. However, carbohydrates-rich foods such as potato chips and biscuit samples have been the common studied foods compared with protein-rich foods such as meat samples. In this study, determination of acrylamide in these two group foods was examined using HPLC-MS. For this purpose, firstly, the parameters that are thought to affect the response in the HPLC-MS analysis were optimized. The optimized conditions were found to be 0.3 ml min(-1) for flow rate of mobile phase, 40 mu l for injection volume, 5 degrees C for column temperature and 70 V for fragmentor potential. The optimized method was applied for the determination of acrylamide levels in Turkish foodstuffs including grilled meat and chicken samples, potato chips, coffee and biscuit. The obtained concentrations for all studied foods were in the range of 20-250 mu g kg(-1). The results showed that acrylamide concentrations highly varied depending on the kind of food samples.
