Investigation of antioxidant responses in Gammarus pulex exposed to Bisphenol A

    Tatar, Şule; Türkmenoğlu, Yener

One of the most important environmental problems in the world is micro-pollutants. The aim of this study was to investigate the antioxidant responses of Gammarus pulex to Bisphenol A (BPA), an endocrine-disrupting agent. For this purpose, sublethal concentrations of BPA were applied to G. pulex and biochemical responses were studied. Enzymatic antioxidants superoxide dismutase (SOD) and catalase (CAT) activities and nonenzymatic antioxidants glutathione (GSH) and thiobarbituric acid reagents (TBARS) levels in G. pulex were determined in four different groups during 24 and 96 h. Biochemical biomarkers were measured using commercial kits in a microplate reader. When we compared with control, SOD enzyme activity increased in all groups during both administration periods and CAT enzyme activity decreased in all groups. GSH and TBARS levels were increased after 24 and 96 h of application periods in all groups when compared with control. For changes in SOD and CAT activities and GSH, TBARS levels have been determined to be useful as biomarkers against BPA in G. pulex tissues. It has also been proven that G. pulex is an effective bioindicator that shows BPA pollution in water. The different results of biochemical biomarkers can be evaluated as a marker of possible metabolic processes, and the biochemical response of G. pulex can reveal to some extent the environmental consequences of BPA pollution resulting from industrial waters.

Tatar, Sule; Yildirim, Nuran Cikcikoglu; Serdar, Osman; Erguven, Gokhan Onder

The research study was about revealing the biochemical response of Gammarus pulex related to insecticide methomyl before and after bioremediation by two soil bacteria species, Ochrobactrum thiophenivorans and Sphingomonas melonis. Catalase (CAT), glutathione S-transferase.(GST), cytochrome. P4501A1 (CYP1A1) activities in G. Pulex related to methomyl solution were investigated in 24 h and 96 h. ELISA method was used for test studies. CAT enzyme was decreased in Gammarus pulex that was exposed to methomyl after all exposure period (P < 0.05). CAT activities were returned to control results after bioremediation assays. GST enzyme activity was decreased depending on methomyl exposure during 24 h but increased during 4 days (P < 0.05). After 8 days of bioremediation period, GST activity increased again during 24 h while decreased during 4 days (P < 0.05). CYP1A1 activity increased in Gammarus pulex that was exposed to methomyl after all exposure period (P > 0.05). After bioremediation, statistically significant changes were not revealed in CYP1A1 activities (P > 0.05). According to the results of our study, CYP1A1, CAT, and GST activities in G. pulex sanctioned the capability of Ochrobactrum thiophenivorans and Sphingomonas melonis in methomyl bioremediation. Isolated and enriched Ochrobactrum thiophenivorans and Sphingomonas melonis that were added to 2.5 ppb concentrations of methomyl for 8 days. Each day, chemical oxygen demand (COD) and biochemical oxygen demand (BOD5), pH and dissolved oxygen parameters were monitored. At the final phase of the bioremediation step, it was determined that these bacteria have efficient methomyl bioremediation properties in a mixed corsortia at a rate of 86%. These results show that these bacteria can be used for bioremediate the receiving environments that are polluted by these kinds of insecticides.

Accurate deep neural network model to detect cardiac arrhythmia on more than 10,000 individual subject ECG records

Yildirim, Ozal; Talo, Muhammed; Ciaccio, Edward J.; Tan, Ru San; Acharya, U Rajendra

Background and objective: Cardiac arrhythmia, which is an abnormal heart rhythm, is a common clinical problem in cardiology. Detection of arrhythmia on an extended duration electrocardiogram (ECG) is done based on initial algorithmic software screening, with final visual validation by cardiologists. It is a time consuming and subjective process. Therefore, fully automated computer-assisted detection systems with a high degree of accuracy have an essential role in this task. In this study, we proposed an effective deep neural network (DNN) model to detect different rhythm classes from a new ECG database. Methods: Our DNN model was designed for high performance on all ECG leads. The proposed model, which included both representation learning and sequence learning tasks, showed promising results on all 12-lead inputs. Convolutional layers and sub-sampling layers were used in the representation learning phase. The sequence learning part involved a long short-term memory (LSTM) unit after representation of learning layers. Results: We performed two different class scenarios, including reduced rhythms (seven rhythm types) and merged rhythms (four rhythm types) according to the records from the database. Our trained DNN model achieved 92.24% and 96.13% accuracies for the reduced and merged rhythm classes, respectively. Conclusion: Recently, deep learning algorithms have been found to be useful because of their high performance. The main challenge is the scarcity of appropriate training and testing resources because model performance is dependent on the quality and quantity of case samples. In this study, we used a new public arrhythmia database comprising more than 10,000 records. We constructed an efficient DNN model for automated detection of arrhythmia using these records.

Characterization of viscoelastic properties of minced beef meat thawed by ohmic and conventional methods

    Cevik, Mutlu; Icier, Filiz

Frozen minced meat samples having fat contents of 2%, 10% and 18% were thawed using different methods (refrigeration thawing at ambient temperature of +4 ℃, under running cold water (+4 ℃) thawing, ohmic thawing for 10, 13 and 16 V/cm). Viscoelastic properties were determined by using rheological tests (oscillation and creep/recovery tests). Storage modulus, loss modulus, complex modulus, loss tangent, dynamic viscosity and complex viscosity values of minced meat samples increased as fat content increased. As frequency value increased, the modulus values of meat samples increased but dynamic and complex viscosity values of the samples decreased. The minced meat samples thawed by different methods had recoverable compliance values. The compliance values of meat samples during creep region can be well characterized by Burgers model. Ohmic thawing can be used as an alternative thawing method since it resulted in similar rheological properties of minced meat samples compared to refrigeration thawing at ambient temperature of +4 ℃ and under running cold water (+4℃) thawing.

Cimen, Isil Canan Cicek; Danabas, Durali; Ates, Mehmet

In this study, nanotoxicity tests were made by exposure of Artemia salina to copper (Cu 60–80 nm) and copper oxide (CuO 40 nm) nanoparticles (NPs) at different concentrations (0.2, 1, 5, 10, 25, and 50 mg/l) during some exposure duration. Characterization of Cu and CuO NPs were performed using Transmission Electron Microscope (TEM), Dynamic Light Scattering (DLS), Zeta Potential, X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM), Fourier Transformation Infrared (FT-IR) analyzes. In organisms, the accumulation and elimination rates of NPs was determined by Inductively Coupled Plasma-Mass Spectrometry (ICP-MS) analysis and the oxidative stress effects on A. salina were determined by Glutathione (GSH) and Thiobarbituric acid reactive substances (TBARS) analysis methods. Both NPs were found to differ in accumulation and elimination rates at each application time and in parallel with the increase in concentration. In each group, it was determined that ion release increased with application time. The results showed that the accumulation rates in Cu NPs had a tendency to increase at 48 h and to decrease at 72 h in concentrations of 0, 2 and 1 mg/l, respectively. And in the all other concentrations have been seen an increasing trend within the time. In the CuO NPs (40 nm), accumulation rates were a decrease trend at 48th hour and an increase trend at 72nd hour except 10 mg/l concentration. In the 10 mg/l group was an increase trend with the application period. Cu NP, TBARS value increased with increasing concentrations and the highest increase was observed at 24 h of 5 ppm group. The groups showed a tendency to increase-decrease-increase-decrease in TBARS levels in terms of elapsed time (24th, 48th, 72nd hour and elimination) (5 ppm and 10 ppm groups tended to decrease in TBARS level at 72nd hour). TBARS increased with increasing concentration ratios in CuO NPs (40 nm).

Salacia chinensis exerts its antidiabetic effect by modulating glucose-regulated proteins and transcription factors in high-fat diet fed-streptozotocin-induced type 2 diabetic rats

Erten, Fusun; Orhan, Cemal; Tuzcu, Mehmet; Er, Besir; Defo Deeh, Patrick Brice; Sahin, Nurhan; Özercan, Ibrahim Hanifi; Juturu, Vijaya; Sahin, Kazim

This study aimed to investigate the properties of Salacia chinensis (Celastraceae, SC) and its molecular mechanism in the type 2 diabetic rats. Forty-two Wistar rats were divided into six groups (n = 7): control, SC (100 mg/kg, per os), high-fat diet (HFD), HFD + SC (100 mg/kg), HFD + streptozotocin (STZ, 40 mg/kg, i.p.), and HFD + STZ+SC. SC decreased serum glucose, insulin, triglycerides, free fatty acid, and malondialdehyde levels, but increased serum total antioxidant capacity (0.33 ± 0.02 versus. 0.79 ± 0.03), compared with the untreated group (p <.001). Additionally, SC elevated the expression of glucose-regulated proteins GLUT2, PPAR-ɣ, p-IRS, and Nrf2, but downregulated NF-κB in the liver and kidney (p <.001). In conclusion, SC could improve insulin resistance by modulation of glucose-regulated proteins and transcription factors in diabetic rats. Practical applications: Present data has contributed to the current ethnomedicinal benefits of SC, through which the SC intake regulated the carbohydrate metabolism and increased the antioxidant capacity. The balance of transcription factors can mediate these efficacies partially and various key proteins involved in energy metabolism, along with oxidative stress and insulin sensitivity.

Temporal and seasonal variations in phytoplankton community structure in Uzuncayir Dam Lake (Tunceli, Turkey)

Kutlu, Banu; Aydın, Rahmi; Danabas, Durali; Serdar, Osman

Seasonal variations in species composition and growth dynamics of phytoplankton in Uzuncayir Dam Lake (Tunceli, Turkey) were studied in relation to ecological factors by collecting horizontal water and phytoplankton samples for over a year. A total of 46 phytoplankton taxa were recorded, where Bacillariophyta was the most abundant in species number representing 24 taxa. Diatoms were the most conspicuous phytoplankton in terms of abundance as well as occurrence frequency. Charophyta, Chlorophyta, Cyanobacteria, Ochrophyta, Miozoa, and Euglenozoa were the least abundant. However, some Diatoms (Gyrosigma attenuatum, Hippodonta hungarica, Meridion circulare, Nitzschia clausii, Nitzschia frustulum, Pantocsekiella ocellata, Rhoicosphenia abbreviata), Charophytes (Cosmarium granatum, Mougeotiopsis calospora), Cyanobacteria (Leiblenia epiphytica, Planktothrix agardhii); Dinoflagellates (Ceratium hirundinella, Parvodinium africanum, Parvodinium inconspicuum, Peridinium cinctum), and Ochrophyta (Dinobryon pediforme) were also noticeable, with their occurrence in all seasons. Canonical correspondence analysis (CCA) and principal component analysis (PCA) were employed to analyze the relationship between occurrence and abundance of phytoplankton species as well as the ecological factors. The results showed that pH and the concentrations of NH4, NO3, and PO4 influenced seasonal occurrence and abundance of phytoplankton species in the lake. Uzuncayir Dam Lake may be classified as an oligotrophic lake with the mean value of nitrate 2.62 mg L−1 and phosphate 0.23 mg L−1 as supported by the occurrence of various phytoplankton groups. Findings related to pollution indices yielded that Uzuncayir Dam Lake is on the edge of the moderate state of organic pollution.

Effects of Zn and ZnO Nanoparticles on Artemia salina and Daphnia magna Organisms: Toxicity, Accumulation and Elimination

Danabas, Durali; Ates, Mehmet; Ertit Tastan, Burcu; Cicek Cimen, Isil Canan; Unal, Ilkay; Aksu, Onder; Kutlu, Banu

In the study, Zn in the size of 40–60 nm and 80–100 nm and ZnO in the size of 10–30 nm were applied to A. salina and D. magna individuals in 7 groups with 3 repetitions. Measurements were made at 24th, 48th and 72nd hours and elimination values were examined at +24 h. LC50 values of NPs were determined and chemical analysis (metal accumulation and elimination), ion quantities which were given to the environment and the survival rates of organisms were determined after the exposure. According to the results of phase contrast microscopy, it was found that both experimental organisms absorbed the NPs in the medium level. In the toxicity results of D. magna, it can be said that Zn NP (40–60 nm) has a highly toxic effect only at 50 ppm concentration for 48 h and lethal dose can be accepted as of 5 ppm at the end of 72 h. In A. salina individuals, it is clearly seen that there is an increase in mortality in organisms parallel to the dose increase. Although all NPs were applied to organisms in low doses corresponding to environmental values, it was observed that toxic effect was in parallel with the increase in time. It is clearly known that there is the inverse proportion between the size of NPs and the toxic effect. The smaller the size of NPs is, the higher the toxic effect becomes When the results of Zn accumulation and elimination of A. salina and D. magna individuals exposed to the Zn and ZnO NPs were examined; it was found that accumulation and elimination occurred in parallel with the increase in concentration at each application hour and elimination. Intensive and possible misuse of nanoscale materials is one of the biggest threats to the environment and all living things worldwide.

Anesthetic Efficiency of Three Medicinal Plant Oils for Aquatic Species: Coriander Coriandrum sativum, Linaloe Tree Bursera delpechiana, and Lavender Lavandula hybrida

Can, Erkan; Kızak, Volkan; Can, Şafak Seyhaneyıldız; Özçiçek, Esin

This study evaluated the potential of three essential oils (EOs) that were obtained from coriander Coriandrum sativum (CEO), linaloe tree Bursera delpechiana (BEO), and lavender Lavandula hybrida (LEO) as anesthetic agents. Convict Cichlids Amatitlania nigrofasciata (Günther 1867) were exposed to eight concentrations of anesthetic (50, 75, 100, 125, 150, 200, 250, and 300 μL/L). After exposure to the anesthetic, the fish were transferred to clean water to recover. All of the EOs produced an anesthetic effect after exposure to the compounds for 30 min at the minimal effective concentration (MEC), which was identified according to deep anesthesia (A5 < 3 min) and full recovery (R3 < 5 min) times. At 50 and 75 μL/L, the total loss of equilibrium was not observed for all tested EOs. The total loss of reflex was induced at a faster rate with higher concentrations of anesthetic in all groups. The recovery time generally increased as the concentration of the anesthetic increased. These findings suggest that CEO, BEO, and LEO are all novel potential anesthetics for aquaculture, and the optimal concentrations were identified as 150 μL/L (A5; 156 ± 1.7 s and R3; 165 ± 2.9 s), 125 μL/L (A5; 176 ± 3.5 s; R3; 125 ± 2.0 s), and 200 μL/L (A5; 20.1 ± 2.4 s and R3; 162 ± 3.4 s), respectively. When considering the active ingredients of EOs, this study also demonstrated that future studies should be focused on the major components such as linalyl acetate, 1.8-cineole, α-pinene, geraniol, and linalool. Their synergistic effects should be examined in herbal anesthetic treatments, since new commercial anesthetics will likely contain them.

Regulation of signaling pathways by β-elemene in cancer progression and metastasis

Qureshi, Muhammad Z.; Attar, Rukset; Romero, Mirna A.; Sabitaliyevich, Uteuliyev Y.; Nurmurzayevich, Sadykov B.; Ozturk, Ozlem; Wakim, Lara H.; Lin, Xiukun; Ozbey, Ulku; Yelekenova, Armida Bakhytbekovna

Entry of β-elemene into various phases of clinical trials advocates its significance as a premium candidate likely to gain access to mainstream medicine. Based on the insights gleaned from decades of research, it seems increasingly transparent that β-elemene has shown significant ability to modulate multiple cell signaling pathways in different cancers. We partition this multicomponent review into how β-elemene strategically modulates various signal transduction cascades. We have individually summarized regulation of tumor necrosis factor related apoptosis-inducing ligand, signal transducers and activators of transcription, transforming growth factor/SMAD, NOTCH, and mammalian target of rapamycin pathways by β-elemene. Last, we will discuss the results of clinical trials of β-elemene and how effectively we can use these findings to stratify patients who can benefit most from β-elemene.

Gammarus pulex as a Model Organism to Assess the Residual Toxicity of Slaughterhouse Wastewater Treated by Electrocoagulation Process

Yildirim, Nuran Cikcikoglu; Tanyol, Mehtap; Serdar, Osman; Yildirim, Numan

It was assessed the efficiency of the electrocoagulation (EC) in slaughterhouse wastewater (SW) treatment by using antioxidant parameters of Gammarus pulex. The SW was treated by EC. Superoxide dismutase (SOD), glutathione peroxidase (GPx) and catalase (CAT) activities and malondialdehyde (MDA) levels in G. pulex exposed to pre- and post-treated of the SW during 24 h and 96 h were analysed. Standard methods were applied during the analysing process of the physicochemical quality parameters for both untreated and treated SW. All measured physicochemical parameters were decreased following the treatment process via EC. After the treatment process, it was observed that while SOD activities and MDA levels were decreased, CAT activities were increased and GPx activities did not exhibit any change. In conclusion, the present study demonstrated the abilities of untreated SW to promote oxidative stress in model organism. The SOD, CAT activities and MDA levels in G. pulex revealed that EC process were efficient in the SW treatment.

Apigenin as an effective anticancer natural product: Spotlight on TRAIL, WNT/β-catenin, JAK-STAT pathways, and microRNAs

Ozbey, Ulku; Attar, Rukset; Romero, Mirna Azalea; Alhewairini, Saleh S; Afshar, Behnaz; Uteuliev, Yerzhan Sabitaliyevich; Hanna-Wakim, Lara; Ozcelik, Beraat; Farooqi, Ammad Ahmad

Wealth of information gleaned from decades of high-impact research work; scientists have disentangled the complicated web of versatile regulators that underlie cancer development and progression. Use of structural biology approaches and functional genomics have helped us to gain new insights into complex nature of cancer, and it is now clear that genetic/epigenetic mutations, overexpression of oncogenes, inactivation of tumor suppressors, loss of apoptosis, and versatility of protein binding partners have contributory roles in carcinogenesis and metastatic spread. It is becoming progressively more understandable that reprogramming of gene expression during and nontranscriptional changes during cancer development and progression are initiated and controlled by deregulated signal transduction cascades, all of which collectively create an incalculable complexity. Data obtained through preclinical and clinical trials revealed that alterations in the targeted oncogenes and other downstream, and parallel pathways played a central role in the development of resistance against different therapeutics. Phytochemicals have regained limelight, and different natural products are currently being tested for efficacy in preclinical studies. Apigenin, a plant-derived flavonoid has considerable pharmacological value and is reportedly involved in the regulation of different signaling cascades. In this review, we have attempted to summarize rapidly evolving understanding of molecular biologists and pharmacologists about the potential of apigenin in the regulation of deregulated signaling pathways in different cancers. We have emphasized on the regulation of WNT/β-catenin and janus kinase/signal transducers and activators of transcription (JAK-STAT) pathways. We also comprehensively discuss how apigenin restored apoptosis in tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)-resistant cancers. The review also gives a snapshot of microRNAs (miRNAs) that regulate wide-ranging biological processes, and it is now clear that each miRNA can control hundreds of gene targets. Apigenin was noted to upregulate miR-520b and miR-101 in different cancers to inhibit tumor growth. Moreover, apigenin-induced apoptotic rate was significantly higher when used in combination with miR-423-5p inhibitors or miR-138 mimics. Better comprehension of linear and integrated signaling pathways will be helpful in effective therapeutic targeting of deregulated signaling pathways to inhibit/prevent cancer.

BACKGROUND: The aromatic amino acid L-tryptophan is a versatile molecule and needed for the biosynthesis of proteins. Due to metabolic functions, it has been widely used in research and clinical trials. OBJECTIVE: The present study aimed to test the usefulness of L-tryptophan as an extender of Tigris scraper (Capoeta umbla) sperm during cryopreservation. MATERIALS AND METHODS: Semen samples were diluted with a ratio of 1:3 (sperm: extender) and base extenders were supplemented with L-tryptophan (0 mM, 0.5 mM, 1 mM, 2 mM and 4 mM). Sperm motility and duration of post-thawed samples were assessed after the cryopreservation process. RESULTS: Our results revealed that motility rate and duration of sperm increased when the cryomedia was supplemented with L-tryptophan. On the other hand, an increase in the concentration of L-tryptophan in the extender caused a significant decrease (p<0.05) in the motility rate of Tigris scraper (C. umbla) sperm. The ideal results were obtained at 1 mM L-tryptophan; however, no motile sperm were shown in any samples when a concentration of 2 mM was used. CONCLUSION: The current study shows the importance of conducting further studies related to long-term storage and reproduction management.
