Stress-Induced Changes in Tyrosine Hyroxylase Enzyme Activity and Adrenomedullin Levels in Rat Hypothalamus, Adrenal Medulla and Heart Tissues

    Yildirim, Nuran Cikcikoglu; Yurekli, Muhittin

In this study, it was aimed to investigate the effects of chronic cold stress on adrenomedullin levels and tyrosine hydroxylase enzyme activities in some rat tissues. In this study, 12 female Sprague Dawley rats weighing 200 to 250 g were used. Rats were housed under diurnal lighting conditions (12-12 h) with free access to food and water. For this study, rats were divided into two groups, namely control group and cold stress treated group. Adrenomedullin levels were measured by high performance liquid chromatography. Tyrosine hydroxylase enzyme activities was spectrometrically measured. Tyrosine hydroxylase activity was found to be increased significantly (p<0.05) in adrenal medulla, hypothalamus and heart tissues depending on cold stress treatment. Adrenomedullin levels were determined to be decreased in the cold stress treatment group. The differences of between control and cold stress treatment groups were significant (p<0.05). The results suggested that adrenomedullin may play a possible role in adaptation to stress. Exposure to cold increases the synthesis and release of catecholamines. Tyrosine hydroxylase is the rate limiting step in the biosynthesis of catecholamines for this reason cold stress increased tyrosine hyroxylase enzyme activities. Further studies are needed to reveal correlation between adrenomedullin, tyrosine hydroxylase and cold stress.

    Yildirim, Numan; Asma, Dilek

The effect of different cadmium (Cd) concentrations (5 mu M and 500 mu M) on antioxidant enzymes including superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), glutathione reductase (OR, EC 1.8.1.7) and glutathione S-transferase (GST, EC 2.5.1.18), and glutathione levels was studied in white rot fungus Phanerochaete chrysosporium (ME446), grown in liquid medium. Exposure to 500 mu M Cd resulted in significant induction of SOD actvity. Exposure to high (500 mu M) and low concentration (5 mu M) led to a clear induction of CAT activity with respect to control group. Exposure to 5 mu M Cd increased GR activity. GST activity was induced but GSH level reduced by exposure to 5 mu M Cd.

Comparison of Growth Kinetics of Chaetoceros gracilis Isolated from Two Different Areas in the Aegean Sea (The Bay of Izmir and the Homa Lagoon)

Sunlu, F. Sanem; Kutlu, Banu; Buyukisik, H. Baha

In this study, the species Chaetoceros gracilis was isolated from two different ecosystems (sea-lagoon) in the Bay of Izmir with very different physico-chemical environmental parameters. With the batch culture experiments carried out using the small diatom C. gracilis, it is aimed to determine the physiological properties of this species and also to form a basis for model studies by parameterizing its exponential growth rates, its nutrient limiting growth rates and the Michealis-Menten curves that indicate its dependence on nutrients. The fact that it is spread in a wide variety of areas from oligotrophic regions to eutrophic regions shows that this species environmental adaptation is quite interesting.

IN VITRO ANTIMICROBIAL EVALUATION OF COMMERCIAL TEA EXTRACTS AGAINST SOME PATHOGEN FUNGI AND BACTERIA

Yildirim, N.; Bekler, F. Matpan; Yildirim, N. Cikcikoglu; Dikici, A.

The in vitro antimicrobial activity of crude ethanol, methanol, hexane and aqueous extracts of commercial tea types such as fennel (Foeniculum sp.), senna (Cassia sp.), basilrosemary (Ocimum sp.-Rosmarinus sp), daisy (Bellis sp.) and sage (Salvia sp) against pathogen fungi (Colletotrichum coccodes, Epicoccum nigrum, Scopulariopsis brevicaulis) and foodborne pathogen bacteria (Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923, Bacillus cereus, Klebsiella pneumoniae, Enterobacter sp.) were investigated with disk diffusion method. Senna (Cassia sp.) exhibed the most effective antimicrobial activity in both ethanolic and methanolic extracts. The best antimicrobial effect (30.6 mm) against E. nigrium was seen in methanolic senna (Cassia sp.) extract. The results of the study indicate that commercial tea types have showed various levels of antimicrobial activity depend on the type of solvent used in the extraction procedure.

    Pala, Murat; Yuksel, Fahrettin

This study was conducted in Keban dam lake Cemisgezek region during 2000-2001 fishing season. In the study, the effect of mesh size on catching efficiency was analyzed. For this reason, a total length of 800 m gillnet was used, particularly two monofilament gillnets with 50, 60, 70 and 90 mm mesh size, each being 100 m long. During the research, Cyprinus carpio and Squalius cephalus was indicated with own species names, which are caught on 22 hunting attempts. Barbus esocinus, Barbus capito pectoralis and Barbus mystaceus species regarded as Barbus sp., Capoeta trutta and Capoeta umbla species regarded as Capoeta sp. The most frequently caught species in the research were Capoeta sp. on 50 and 70 mm mesh sizes, Barbus sp. on 60 mm mesh size and Cyprinus carpi on 90 mm mesh size. The species that is least frequently fished on all gillnets was Squalius cephalus. When the catch per unit effort was analyzed taking into account all species, it was observed that with 83.1 g m(-1) catch per unit effort, 60 mm gillnet was more efficient than all other gillnets. This gillnet was followed by 79.9, 41.0 and 31.8 g m(-1) catch per unit efforts for 50, 70 and 90 mm mesh sizes, respectively. In conclusion, it has been found out that in the study region, gillnets with 50 and 60 mm mesh sizes are more effective than gillnets with 70 and 90 mm mesh sizes.

The effect of adrenomedullin and cold stress on interleukin-6 levels in some rat tissues

    Yildirim, N. C.; Yurekli, M.

P>Stress known to stimulate sympathetic activity, as well as the hypothalamic-pituitary-adrenal axis (HPA), produces a significant increase in adrenomedullin (AdM) levels, suggesting a regulatory or protective role for AdM in countering HPA activation that follows a variety of stressors. Stressors can modulate the secretion of proinflammatory cytokines. Interleukin (IL)-6 is a potent activator of the HPA and appears to play a pathogenic role in conditions related to stress. In the present study, we investigated the administration of AdM on IL-6 levels in cold exposed rats. Male Wistar rats were divided into four groups as control, adrenomedullin treatment, cold stress and cold stress+adrenomedullin-treated groups. In the adrenomedullin-treated group, animals received intraperitoneal (i.p.) injection of adrenomedullin (2000 ng/kg body weight) once a day for a week. For the cold stress exposure the rats were kept in separate cages at 10 degrees C for a week. Control group rats were kept in laboratory conditions. The concentration of IL-6 was determined using an enzyme-linked immunosorbent assay (ELISA) kit. When compared to control, IL-6 levels increased significantly in the cold stress- and adrenomedullin-treated groups (P < 0 center dot 05). Administration of AdM in addition to cold stress decreased IL-6 levels in lung and liver, but increased in brain and heart when compared to control (P < 0 center dot 05). The results suggest that cold stress may induce increase of rat proinflammatory cytokine IL-6 and adrenomedullin may play a regulatory or protective role for cold stress.

Adrenomedullin administration alters vascular endothelial growth factor levels in rats in cold stress

    Yildirim, Nuran Cikcikoglu; Yurekli, Muhittin

Background: Many endogenous peptides play important regulatory roles in angiogenesis by modulating endothelial cell behavior. Adrenomedullin (AdM) is one of such factors. Angiogenesis and vascular endothelial growth factor (VEGF) are indistinguishable. Exposure to cold environment stimulates capillary angiogenesis. Objectives: Examine the effect of the bioactive peptide AdM on VEGF levels in rat liver, lung, brain, and heart tissues after cold stress treatment. Methods: Male wistar rats were divided into four groups as control, AdM treatment, cold stress and AdM+cold stress treated groups. In AdM-treated group, animals received intraperitoneal injection of AdM (2000 ng/kg body weight) once a day during a week. For the cold stress exposure, the rats were kept in separate cages at 10 degrees C for a week. Results: The administration of AdM increased VEGF levels in all tissues in cold exposed rats. Conclusion: AdM may be a major regulatory factor in angiogenesis by modulating VEGF levels that is closely associated with cold exposure-related metabolic stimulation.

Kayim, Murathan; Bozkurt, Yusuf; Ogretmen, Fatih

The effect of ovulation stimulators (Ovopel and carp pituitary extract) on spawning of scaly carp females were investigated. Following application of Ovopel, eggs were obtained from higher percentage of females than CPE treatment (92.5 and 75.0%, respectively. In the case of Ovopel stimulator, female weights were significantly correlated with living embryo (r = 0.576, p<0.01) and hatching rates (r = 0.684, p<0.01). Also interaction between egg weights and fertilization rates (r = 0.603, p<0.01) and between hatching and living embryo rates (r = 0.679, p<0.01) were determined significant. On the other hand, egg weights were negatively correlated (r = -0.368, p<0.01) with living embryo rates. For the CPE stimulator, egg weights were significantly correlated with fertilization (r = 0.662, p<0.01), living embryo (r = 0.390, p<0.05) and hatching rates (r = 0.443, p<0.01). Also hatching were significatly correlated with fertilization (r = 0.665, p<0.01) and living embryo rates (r = 0.903, p<0.01). In addition, fertilization and living embryo rates significantly correlated (r = 0.615, p<0.01) with each other.

LEAD, CADMIUM AND COPPER CONCENTRATIONS IN LEAVES OF Nerium oleander L. AND Robinia pseudoacacia L. AS BIOMONITORS OF ATMOSPHERIC POLLUTION

Kaya, Gokce; Okumus, Nuh; Yaman, Mehmet

Pb, Cd and Cu concentrations in leaves of Nerium oleander L. and Robinia pseudoacacia L. plants collected from industrial areas in Gaziantep-Turkey were examined for biomonitoring of atmospheric pollution using flame atomic absorption spectrophotometry. The Pb levels in the leaves of Nerium oleander L. and Robinia pseudoacacia L. collected at a distance of 50 m from the battery plant were observed to be 2820 and 1860 mg kg(-1), respectively. The Pb, Cd, and Cu concentrations in the soils studied were in the range of 17-602, 0.142-0.656 and 12-38 mg kg(-1), respectively. Lead concentrations described above for plant leaves were found to be about 2000-fold higher than those in the same plants collected from the control area (1.0 mg Pb kg(-1) for both plant species). Similarly, Cd levels in Robinia pseudoacacia L. collected from polluted area were observed to be about 40 times higher than those from control area. Consequently, it is observed that the leaves of Neriurn oleander L. can be used for biomonitoring of Pb, and Robinia pseudoacacia L. for biomonitoring of both Pb and Cd pollution.

The Effect of Simulated Transportation on Biochemical Plasma Parameters of Japanese Quails

    Tekelioglu, O.; Sezer, M.; Ekici, F.; Atis, O.; Akbas, A.

The effect of the duration of simulated transportation on blood SOD (Superoxide Dismutase), GSH-Px (Glutathione Peroxidase), AST (Aspartate Transaminase), ALT (Alanine Transaminase), GGT (Gamma-Glutamyl Transpeptidase), LDH (Lactic Dehydrogenase) enzyme levels and NO (Nitric Oxide) and MDA (Malondialdehyde) product levels was investigated in Japanese quails. Simulated transportation was carried out with a road simulator which was calibrated for Turkish roads conditions. Animals were loaded into well ventilated carrier boxes and carried at 2 and 5 h on the simulator. Blood samples were collected from 10-weeks-old Japanese Quails just after transportation. A significant (p<0.05) increase in all measured plasma parameters (except GGT) at 5 h transportation was observed. However, all of the plasma parameters have not shown a significant increase at 2 h transportation. This transportation duration might be the evidence of the time duration in still safe zone for Japanese Quails for that condition.

Serezli, Ramazan; Guzel, Senol; Kocabas, Mehmet

In this study, fecundity and egg size of rainbow trout (Oncorhynchus mykiss), Black Sea trout (Salmo labrax) and brook trout (Salvelinus fontinalis) were examined. Two and six years old female fish reared in commercial fish farm were used and the female size, total and relative fecundity and egg size (mm) were determined for the broodstock fish of three tested species. The average weights of broodfish of rainbow trout, Black Sea trout and brook trout 1.357.27+/-406, 532+/-673.7 and 310.40+/-85.0 g, respectively. The total fecundity (number of eggs fish(-1)) was 2.809+/-791, 1.401+/-1374 and 723+/-320, the relative fecundity (number of eggs kg(-1) fish) was 2.180+/-676, 3.558+/-1307 and 2.571+/-1530 and the egg size (mm); 4.95+/-0.2, 4.51+/-0.67 and 4.49+/-0.21 were determined for rainbow trout, Black Sea trout and brook trout, respectively. A significant linear relationship was found total fecundity and the post-stripped fish weight of Black Sea trout (r = 0.96), but this relationship was low for the rainbow trout (r = 0.28) and brook trout (r = 0.17). A negative relationship was observed between the relative fecundity and brood size in rainbow trout (r = 0.57), Black Sea trout (r = 0.58) and brook trout (r = 0.54). A significant linear relationship was observed in rainbow trout (r = 0.63), Black Sea trout (r = 0.86) and brook trout (r = 0.41) (p<0.01).

Wavelet-based feature extraction and selection for classification of power system disturbances using support vector machines

Eristi, Hueseyin; Uçar, Ayseguel; Demir, Yakup

This paper presents a new approach for the classification of the power system disturbances using support vector machines (SVMs). The proposed approach is carried out at three serial stages. Firstly, the features to be form the SVM classifier are obtained by using the wavelet transform and a few different feature extraction techniques. Secondly, the features exposing the best classification accuracy of these features are selected by a feature selection technique called as sequential forward selection. Thirdly, the best appropriate input vector for SVM classifier is rummaged. The input vector is started with the first best feature and incrementally added the chosen features. After the addition of each feature, the performance of the SVM is evaluated. The kernel and penalty parameters of the SVM are determined by cross-validation. The parameter set that gives the smallest misclassification error is retained. Finally, both the noisy and noiseless signals are applied to the classifier given above stages. Experimental results indicate that the proposed classifier is robust and has more high classification accuracy with regard to the other approaches in the literature for this problem. A new algorithm for automatic classification of power quality events based on wavelet transform and SVM

    Eristi, Hueseyin; Demir, Yakup

This paper presents a new approach for automatic classification of power quality events, which is based on the wavelet transform and support vector machines. In the proposed approach, an effective single feature vector representing three phase event signals is extracted after signals are applied normalization and segmentation process. The kernel and penalty parameters of the support vector machine (SVM) are determined by cross-validation. The parameter set that gives the smallest misclassification error is retained. ATP/EMTP model for six types of power system events, namely phase-to-ground fault, phase-to-phase fault, three-phase fault, load switching, capacitor switching and transformer energizing, are constructed. Both the noisy and noiseless event signals are applied to the proposed algorithm. Obtained results indicate that the proposed automatic event classification algorithm is robust and has ability to distinguish different power quality event classes easily. (C) 2009 Elsevier Ltd. The Effects of Commercial Diet Supplemented with Mannanoligosaccharide (MOS) and Vitamin B12 on the Growth and Body Composition of the Carp (Cyprinus carpio L. 1758)

    Atar, Hasan Huseyin; Ates, Mehmet

Four hundred carp fries (Cyprinus carpio L. 1758) with average initial weight of 1.1 +/- 0.05 g and total average length of 4.04 +/- 0.04 cm, was fed for a period of 90 days with the addition of commercial feed Mannanoligosaccharide (MOS) and vitamin B-12. The carps were divided into four different groups and fed through different feeds (Control; group A with MOS, group B with vitamin B-12, group C with a the combination of MOS + vitamin B-12). The effects of feeding through MOS and vitamin B-12 in different proportions on the parameters such as growth in view of live weight and total length, survival rate, feed conversion, condition factor and protein efficacy for the carp fries was studied. The best growth in view of live weight was maintained through MOS and vitamin B-12 combination in group C (4.51 +/- 0.13 g), while the best growth in terms of length was obtained through MOS in group A (6.68 +/- 0.08 cm). As a consequence, the growth parameters of the carp fries fed through commercial feeds with the addition of MOS and vitamin B-12 in different doses under empirical conditions shown affirmative effects on the survival rate, feed assessment, condition factor and protein efficacy.

Acrylamide concentrations in grilled foodstuffs of Turkish kitchen by high performance liquid chromatography-mass spectrometry

Kaplan, Olcay; Kaya, Gokce; Ozcan, Cemile; Ince, Muharrem; Yaman, Mehmet

For over ten years, there has been a considerable interest in determination of acrylamide in foodstuffs. It was known that both protein-rich and carbohydrates-rich foods cooked at high-temperatures can cause acrylamide formation. However, carbohydrates-rich foods such as potato chips and biscuit samples have been the common studied foods compared with protein-rich foods such as meat samples. In this study, determination of acrylamide in these two group foods was examined using HPLC-MS. For this purpose, firstly, the parameters that are thought to affect the response in the HPLC-MS analysis were optimized. The optimized conditions were found to be 0.3 ml min(-1) for flow rate of mobile phase, 40 mu l for injection volume, 5 degrees C for column temperature and 70 V for fragmentor potential. The optimized method was applied for the determination of acrylamide levels in Turkish foodstuffs including grilled meat and chicken samples, potato chips, coffee and biscuit. The obtained concentrations for all studied foods were in the range of 20-250 mu g kg(-1). The results showed that acrylamide concentrations highly varied depending on the kind of food samples. 207Pb-206Pb, 40Ar-39Ar and Fission-Track Geothermochronology Quantifying Cooling and Exhumation History of the Kaman-Kirsehir Region Intrusions, Central Anatolia, Turkey

Boztug, Durmus; Güney, Ozlem; Heizler, Matt; Jonckheere, Raymond C.; Tichomirowa, Marion; Otlu, Nazmi

The Kaman-Kirsehir region intrusions were generated in a post-collisional extensional setting following Cenomanian-Turonian docking of an oceanic island arc, comprising the supra-subduction zone (SSZ) Central Anatolian ophiolite (CAO), onto the Tauride-Anatolide Platform (TAP). These granitoids have been named, from N to S, the Camsari quartz syenite, Hamit quartz syenite, Bayindir nepheline-cancrinite syenite, Durmuslu nepheline-nosean-melanite syenite porphyry and Baranadag quartz monzonite. They intrude the crustal metasediments of the Central Anatolian Crystalline Complex (CACC) and CAO and are unconformably overlain by Upper Paleocene to Lower-Middle Eocene sediments. The single-zircon Pb-207-Pb-206 evaporation age of the Camsari unit is 95.7 +/- 5.1 Ma; those of the Hamit and Baranadag units are indistinguishable with a weighted mean of 74.3 +/- 4.5 Ma. The amphibole 40Ar-39Ar ages of the Hamit and Baranadag units are almost identical with a weighted mean of 72.7 +/- 0.1 Ma. The apatite fission-track age vs elevation plot for the Camsari, Hamit, Durmuslu and Baranadag samples reveals rapid exhumation (>1 km/Ma) between similar to 57 and similar to 61 Ma, consistent with the results of track-length modelling. This Early-Middle Paleocene rapid exhumation is thought to result from uplift triggered by continent (TAP) - continent (Eurasian plate; EP) collision following the closure of Izmir-Ankara-Erzincan (IAE) ocean that also initiated the formation of peripheral foreland basins in central Anatolia. 207Pb-206Pb, 40Ar-39Ar and Apatite Fission-Track Geothermochronology Revealing the Emplacement, Cooling and Exhumation History of the Karacayir Syenite (N Sivas), East-Central Anatolia, Turkey

Boztug, Durmus; Türksever, Esra; Heizler, Matt; Jonckheere, Raymond C.; Tichomirowa, Marion

The Karacayir syenite, intruding the Palaeozoic crustal metamorphics and unconformably overlain by Upper Paleocene to Eocene Tokus formation in the north of Sivas (east-central Anatolia), has been studied with respect to Pb-207-Pb-206 single zircon evaporation, biotite 40Ar-39Ar, and apatite fission-track geothermochronology. Pb-207-Pb-206 single zircon evaporation dating yields an age of 99.0 +/- 11.0 Ma (Cenomanian-Turonian) which is considered to be the intrusion age. Biotite 40Ar-39Ar age determination gives a cooling age of ca. 65 Ma. Apatite fission-track dating, combined with T-t modeling based on track-length distribution data, determines a fast tectonic exhumation with an uplift rate of > 1 mm/a which occurred 58-61 Ma ago. This Middle Paleocene fast tectonic uplift is considered to have resulted from a compressional regime induced by the collision between the Tauride-Anatolide platform (TAP) and the Eurasian plate (EP) along the Izmir-Ankara-Erzincan (IAE) suture zone following the closure of the north-dipping subducted IAE ocean which belongs to northern Neo-Tethyan realm. This compressional regime has also formed the peripheral foreland basins in central Anatolia.

Heat transfer augmentation by swirl generators inserted into a tube with constant heat flux

Kurtbas, Irfan; Guelcimen, Fevzi; Akbulut, Abdullah; Buran, Dincer

A novel conical injector type swirl generator (CITSG) is devised in this study. Performances of heat transfer and pressure drop in a pipe with the CITSG are experimentally examined for the CITSGs' angle (alpha) of 30 degrees, 45 degrees and 60 degrees in Reynolds number (Re) range of 10,000-35,000. Moreover, circular holes with different numbers (N) and cross-section areas (A(h)) are drilled on the CITSG. In this way, total areas (A(t) = N . A(h)) of the holes on the CITSG are equaled each other. Besides, flow directors having three different angles (beta = 30 degrees, 60 degrees and 90 degrees) to radial direction are attached to every one of the holes. This study is a typical example for decaying flow. All experiments were conduced with air accordingly; Prandtl number was approximately fixed at 0.71. The local Nusselt number (Nu(x)), heat transfer enhancement ratio (Nu(ER)) and heat transfer performance ratio (Nu(PR)) are calculated and discussed in this paper. It is found that the Nu(ER) decreases with increase in Reynolds number, the director angle (beta), the director diameter (d), and with decrease in the CITSG angle (alpha). Likewise, variation of Nu(PR) and Nu(ER) is also essentially similar for the same independent parameters. (C) 2009 Elsevier Ltd. All rights reserved.
