The in vitro effect of Lambda-cyhalothrin on quality and antioxidant responses of rainbow trout Oncorhynchus mykiss spermatozoa

Kutluyer, Filiz; Erisir, Mine; Benzer, Fulya; Ögretmen, Fatih; Inanan, Burak Evren

There is little information in the scientific literature about effect of in vitro exposure of fish spermatozoa to pesticides. In vitro effect of Lambda-cyhalothrin (La) on sperm quality and oxidative stress has not been fully explored yet. The effects of LCT, which is a synthetic pyrethroid insecticide, on quality and oxidative stress of spermatozoa were investigated in vitro due to extensively use to control a wide range of insect pests in agriculture, public health, and homes and gardens. To explore the potential in vitro toxicity of La, fish spermatozoa were incubated with different concentrations of LCT (0.6, 1.2 and 2.4 mu g/L) for 2 h. Reduced glutathione (GSH), glutathione peroxidase (GSH-Px), catalase (CAT) and malondialdehyde (MDA) in spermatozoa were analyzed for determination of oxidant and antioxidant balance. Our results indicated that the percentage and duration of sperm motility significantly decreased with exposure to LCT. Activity of GSH-Px and MDA (P < 0.05) and GSH levels (P < 0.05) increased in a concentration-dependent manner while CAT activity decreased (P < 0.05). In conclusion, the oxidant and antioxidant status and sperm quality were affected by increasing concentrations of LCT. (C) 2015 Elsevier B.V. All rights reserved.

Polat, Ridvan; Cakilcioglu, Ugur; Kaltalioglu, Kaan; Ulusan, Musa Denizhan; Turkmen, Zafer

Ethnopharmacological relevance: This paper provides significant ethnobotanical information on medicinal plants in Espiye in the Black Sea Region. Recording such data calls for urgency. This is the first ethnobotanical study in which statistical calculations about plants are carried out by means of FIC method in Northern (Espiye-Giresun) part of Turkey. This study aims to identify the wild plants collected for medicinal purposes by locals of Espiye which is located in the Black Sea Region of Turkey, and to identify the uses and local names of these wild plants. Materials and methods: A field study had been carried out for a period of approximately 2 years (20122014). During this period, information about medicinal use of 55 wild and 15 cultivated plants were collected. Demographic characteristics of participants, names of the local plants, their utilized parts and preparation methods were investigated and recorded. Through face-to-face interviews, we identified and recorded demographic characteristics of the respondents. We interviewed 128 persons who are over the age of 29. The plant taxa were collected within the scope of the study; and herbarium materials were prepared. In addition, the relative significance value of the taxa was determined, and informant consensus factor (FIC) was calculated for the medicinal plants included in the study. Results: We have found out in the literature review of the plants included in our study that 70 plant taxa are already used for medicinal purposes while 3 plants are not available among the records in the literature. The most common families are Asteraceae, Rosaceae, Lamiaceae, Malvaceae, and Ericaceae. We include in our study and report for the first time the medicinal uses of Alchemilla crinita Buser, Stachys sylvatica L., and Thymus leucotrichus Hal. var. leucotrichus Hal. Names of local plants in Turkey vary especially due to vernaculars. The plants that the locals of Espiye use are called with the same or different local names in various parts of Anatolia. Conclusion: We found out that locals living in the research area use for therapeutic purpose 70 plants taxa which belong to 36 families. Turkish citizens with different ethnic backgrounds took the questionnaire. These people use these wild plants in treatment of several diseases. Comparison of the data obtained in this study with the experimental data obtained in the previous laboratory studies on the wild plants which grow in Espiye proved ethnobotanical usages to a great extent. Literature review indicated that the therapeutic plants that grow in Espiye are used in different parts of the world for the treatment of similar diseases. (C) 2015 Elsevier Ireland Ltd. Accumulation and Toxicity of CuO and ZnO Nanoparticles Through Waterborne and Dietary Exposure of Goldfish (Carassius auratus)

Ates, Mehmet; Arslan, Zikri; Demir, Veysel; Daniels, James; Farah, Ibrahim O.

Dietary and waterborne exposure to copper oxide (CuO) and zinc oxide (ZnO) nanoparticles (NPs) was conducted using a simplified model of an aquatic food chain consisting of zooplankton (Artemia salina) and goldfish (Carassius auratus) to determine bioaccumulation, toxic effects, and particle transport through trophic levels. Artemia contaminated with NPs were used as food in dietary exposure. Fish were exposed to suspensions of the NPs in waterborne exposure. ICP-MS analysis showed that accumulation primarily occurred in the intestine, followed by the gills and liver. Dietary uptake was lower, but was found to be a potential pathway for transport of NPs to higher organisms. Waterborne exposure resulted in about a 10-fold higher accumulation in the intestine. The heart, brain, and muscle tissue had no significant Cu or Zn. However, concentrations in muscle increased with NP concentration, which was ascribed to bioaccumulation of Cu and Zn released from NPs. Free Cu concentration in the medium was always higher than that of Zn, indicating CuO NPs dissolved more readily. ZnO NPs were relatively benign, even in waterborne exposure (p0.05). In contrast, CuO NPs were toxic. Malondialdehyde levels in the liver and gills increased substantially (p<0.05). Despite lower Cu accumulation, the liver exhibited significant oxidative stress, which could be from chronic exposure to Cu ions. (c) 2014 Wiley Periodicals, Inc. Environ Toxicol 30: 119-128, 2015.

Comparison of effects of mild heat combined with lactic acid on Shiga toxin producing Escherichia coli O157:H7, O103, O111, O145 and O26 inoculated to spinach and soybean sprout

Dikici, Abdullah; Koluman, Ahmet; Calicioglu, Mehmet

The aim of the present study was to investigate the effect of lactic acid against Shiga toxin producing Escherichia coli (O157:1-17 and non-O157 serogroups including O103, O111, O145 and O26) at different conditions. Soybean sprouts and spinach leaves inoculated with each serogroup of E. coli (similar to 7.00 + 1.00 log(10) cfu/g) were treated with the lactic acid solutions at different concentrations (0% (control), 1.5%, 2.0%, or 2.5%) and at different temperatures (20, 40, or 50 degrees C) for 3 min. Results indicated that regardless of the treatment temperature, no significant reduction in the numbers of any serogroup occurred in the control group (0%) (p > 0.05). However, lactic acid at concentration of 1.5%, 2% and 2.5% was found to be effective against all organisms tested. There was no significant difference (p > 0.05) between E. coli O157:H7 and non-O157 STEC serogroups at any treatment group. The highest reductions (ca. 400 log(10) cfu/g) of all serotypes in both produces were observed after immersing into 2.5% lactic acid at 50 degrees C. The results of this study showed that decontamination of fresh produces such as spinach and soybean sprout with lactic acid solutions prepared at mild temperatures (40 degrees C and 50 degrees C) might be an effective safety measure in preventing public health risks associated with these products contaminated with STEC. AimThis study was planned to assess the levels of fatigue and social support in patients with diabetes and to determine the relationship between fatigue and social support in these patients. MethodsA descriptive design was used in this research. The study was conducted between March and June 2013 with 300 sampled patients from 1657 studied participants with type 2 diabetes who presented to the diabetes polyclinics of Malatya State Hospital and Inonu University, Turgut Ozal Medical Center. The Multidimensional Scale of Perceived Social Support and the Fatigue Severity Scale were used in the study. ResultsWhen the correlation analysis carried out between fatigue severity and social support in patients with diabetes was examined, a significant relationship was found in the negative direction between fatigue severity and family support (r=-0.145, P<0.05) as well as overall support (r=-0.132, P<0.05). A statistically significant difference was found between the Fatigue Severity Scale score and sex, education status, occupation, cohabitation, presence of complications, and hemoglobin A1c (P<0.05). Family support was significant only with respect to marital status, occupation, and being employed (P<0.05). ConclusionIt was seen at the end of this research that social support reduced the level of fatigue in a number of patients with type 2 diabetes. Nurses of diabetes patients should carry out fatigue assessments routinely and reflect these in nursing care plans by also associating them with the social support sources of the patient.

Determination and Health Risk Assessment of Heavy Metals in Drinking Water of Tunceli, Turkey

    Demir, V.; Dere, T.; Ergin, S.; Cakir, Y.; Celik, F.

Tunceli, Turkey is one of the small cities in the Eastern Anatolian Region with substantial natural, cultural and historical heritage that is suitable to be a slow city. Water sources are abundant in the area due to rain and heavy snowfall in the mountains, which seeps into the ground or flows into the rivers. Therefore monitoring and conservation of city's drinking water quality is one of the main issues in the city. The present study was conducted to investigate several heavy metals and cyanide, arsenic, and selenium concentrations of drinking water samples taken from 100 different locations in the city and towns of Tunceli, Turkey during the years 2011 and 2012. In three locations, high levels of copper was found with a range of 2.1-11.5 mg/L, and high levels of arsenic was detected in five locations with a range of 10.5-78.0 mu g/L, exceeding the action levels for copper and arsenic. For health risk assessment, the average daily dose and hazard quotient (HQ) were calculated. The region mean values of HQ were found above 1 for arsenic in the samples collected from Mazgirt, while HQ values were below 1 in the rest of the samples.

Trichlorfon-induced haematological and biochemical changes in Cyprinus carpio: ameliorative effect of propolis

Yonar, M. Enis; Yonar, Serpil Mise; Pala, Aysegul; Silici, Sibel; Saglam, Naim

Trichlorfon is among the most commonly used products to treat fish parasites in aquaculture. We investigated the effectiveness of propolis in alleviating the toxicity of trichlorfon on haematological and oxidant/antioxidant parameters in carp Cyprinus carpio. Fish were exposed to sublethal concentrations (11 and 22 mg l(-1)) of trichlorfon, and propolis (10 mg kg(-1) of fish weight) was simultaneously administered. At the end of 14 d administration, blood and tissue (liver, kidney, gill) samples were collected. Haematological changes (red and white blood cell count, haemoglobin concentration, haematocrit level and erythrocyte indices: mean corpuscular volume, mean corpuscular haemoglobin and mean corpuscular haemoglobin concentration) were determined in the blood samples, while antioxidant parameters (malondialdehyde and reduced glutathione levels and superoxide dismutase, catalase and glutathione peroxidase activities) were evaluated in the liver, kidney and gill samples. Trichlorfon led to negative alterations in the haematological and antioxidant parameters investigated. The administration of propolis alleviated this effect and suggests that fish treated with trichlorfon improve their physiological status when fed a propolis-supplemented diet.

Arikan, Duygu; Saban, Fatma; Bas, Nazan Gurarslan

Objective: This research is done with the aim of determining hospital and health care satisfaction levels of the parents with inpatient children. Methods: Research was carried out descriptively at a university hospital after necessary permissions were taken from institutions. Parents whose children would stay at hospital between April-June 2012 formed the structure of the research and without using any sampling method, 171 volunteer parents were included in research. In collecting data, PedsQL Health Care Satisfaction Scale and descriptive information forms were used. In the statistical evaluation of data percentage distributions, means, standard deviation, t-test, ANOVA, Kruskal Wallis, Mann Whitney U test and alpha coefficient were used. Results: It was determined that there is statistically important difference between communication and technical skill sub-dimensions (p<0,05). It was confirmed that the longer the child hospitalized, general satisfaction of the parents who cannot supply their own necessities (p<0,05), related to moderate technical skill and emotional requirement sub-dimensions decreases (p<0.05). Besides, lower emotional satisfaction subdimension scores for those who had no accommodation (p<0,05), decreased general satisfaction level of the parents who had another child at home (p<0,05). Also family participation and general satisfaction subdimension scores of the parents whose child incurred financial burden was relatively low (p<0,05). Conclusion: Parents were not satisfied with health care services they received. Accordingly, it is offered that the nurses should have a right to make decisions on the procedures related to families' inpatient children and let them be with their children during the procedures and give them information on the procedures that will be applied on the children.

Effect of Tween 20 on antibacterial effects of acidic, neutral and alkaline decontaminants on viability of Salmonella on chicken carcasses and survival in waste decontamination fluids

Dikici, Abdullah; Arslan, Ali; Yalcin, Halil; Ozdemir, Pinar; Aydin, Isil; Calicioglu, Mehmet

The effect of Tween 20 on antimicrobial effects of acidic (lactic acid), neutral (cetyl pyridinium chloride) and alkaline (trisodium phosphate) decontaminants to reduce the numbers of Salmonella inoculated to broiler carcasses were studied. In addition, survival of Salmonella in waste decontamination fluids was investigated. For this purpose, an average of 7 +/- 1 log(10) cfu/ml Salmonella Enteritidis and Salmonella Typhimurium cocktail was surface-inoculated on to a total of 110 broiler carcasses. Contaminated carcasses were immersed in to tap water (control), lactic acid (LA) (1-2%), cetyl pyridinium chloride (CPC) (0.5-1%), or trisodium phosphate (TSP) (8%) and the combinations of these treatments with Tween 20 (TW20) (0.5%) for 15 min. Results indicated that. Salmonella numbers significantly decreased (P < 0.05) in all groups, except control and 1% LA. No significant difference was found between treatments with and without TW20. In waste decontamination fluids, 6.68 and 3.41 log(10) cfu/ml of Salmonella were recovered from control and CPC groups, respectively, while no viable Salmonella cell was detected in LA and TSP groups by direct plating. It was concluded that regardless of TW20, the antimicrobials tested were effective to reduce Salmonella in poultry carcasses. However, the pathogen may survive in neutral waste decontamination fluid such as CPC and can pose a risk for consumer health by contaminating the plant environment. (c) 2012 Elsevier Ltd. Occurrence and Assessment of Chemical Contaminants in Drinking Water in Tunceli, Turkey

    Demir, Veysel; Ergin, Sezai

The objective of this study was to analyze drinking water samples from 21 sites in the city center and seven municipalities of Tunceli, Turkey, in order to determine the presence of nitrate, nitrite, fluoride, bromate, pesticides, polycyclic aromatic hydrocarbons (PAHs), trihalomethanes (THMs), and some other chemicals. In all locations, the concentrations of chemicals investigated were below the permissible limits set by local and international organizations for drinking water. Low levels of nitrate (4.79 +/- 4.20 mg/L), fluoride (0.11 +/- 0.08 mg/L), and THMs (6.63 +/- 5.14 mu g/L) were detected in all locations. A low level of tetra, chloroethane, which is suspected to be a human carcinogen, was also detected in 8 locations in the range of 0.26-0.43 mu g/L. These contaminants may pose adverse health effects or minimum hazard due to long-term exposure. In all locations, bromate, benzene, total PAH, 1-2 dichloroethane, vinyl chloride, acrylamide, and epichloridine levels in drinking water samples were under detection limits.

Antimicrobial resistance of emerging foodborne pathogens: Status quo and global trends

    Koluman, Ahmet; Dikici, Abdullah

Emerging foodborne pathogens are challenging subjects of food microbiology with their antibiotic resistance and their impact on public health. Campylobacter jejuni, Salmonella spp. and Verotoxigenic Escherichia coli (VTEC) are significant emerging food pathogens, globally. The decrease in supply and increase in demand lead developed countries to produce animal products with a higher efficiency. The massive production has caused the increase of the significant foodborne diseases. The strict control of food starting from farm to fork has been held by different regulations. Official measures have been applied to combat these pathogens. In 2005 EU declared that, an EU-wide ban on the use of antibiotics as growth promoters in animal feed would be applied on 1 January 2006. The ban is the final step in the phasing out of antibiotics used for non-medical purposes. It is a part of the Commission's strategy to tackle the emergence of bacteria and other microbes resistant to antibiotics, due to their overexploitation or misuse. As the awareness raises more countries banned application of antibiotics as growth promoter, but the resistance of the emerging foodborne pathogens do not represent decrease. Currently, the main concern of food safety is counter measures against resistant bugs.

Effect of mixed use of thyme and fennel oils on biochemical properties and electrolytes in rainbow trout as a response to Yersinia ruckeri infection

Gulec, Azime Kucukgul; Danabas, Durali; Ural, Mesut; Seker, Engin; Arslan, Ali; Serdar, Osman

The aim of study was to compare the effects of supplementation of food with herbal oils (Thymus vulgaris and Foeniculum vulgare) on biochemical properties and electrolytes of rainbow trout infected with Yersinia ruckeri. In total, 120 healthy fish (mean weight 84 +/- 1.02 g) were equally divided into four experimental groups. The experimental study was carried out for one week. The first group was control without supplementation and infection, the second group was infected and without oil supplementation, the third group was supplemented with oils (10 ml.100 g(-1) rates) for one week and infected with Y ruckeri and the last group was oil supplemented without infection. Results indicated that fish fed with dietary supplements showed enhanced bactericidal activity, total protein, albumin, cholesterol, triglyceride and bilirubin compared to the control (P < 0.05). As the value of herbal oils was increased in diets, the plasma glucose level decreased. The levels of K, Na, Ca, and Mg increased whereas Cl values decreased, compared to the control. It can be concluded that diet supplementation with herb oils used in this study can increase disease resistance by increasing levels of some biochemical parameters and electrolytes in rainbow trout to Y ruckeri infection.

The objective of the present study was to obtain data about cooking time and temperature of kiymali pide in the restaurants and to investigate thermal inactivation of E. coli O157:H7 during experimental kiymali pide making. A field study was conducted in randomly selected 23 of 87 pide restaurants. Processing parameters including oven temperature, cooking period and post-cooking temperature were determined. Kiymali pide samples were prepared using ground beef filling experimentally inoculated with E. coli O157:H7 (7.6 log(10) CFU/g). Pide samples were cooked at a conventional oven at 180 degrees C for 180, 240, 270, 300 and 330 s. Results of the current study suggest that cooking kiymali pide at 180 degrees C for at least 330 s (5.5 min) may provide sufficient food safety assurance (>= 6 log(10) CFU/g) for E. coli O157:H7. Effects of essential oil compounds on survival of Listeria monocytogenes and Escherichia coli O157:H7 in cig kofte

Dikici, Abdullah; Ilhak, Osman Irfan; Çalicioglu, Mehmet

This study was undertaken to evaluate the effects of some essential oil compounds for the inactivation of Listeria monocytogenes and Escherichia coli O157:H7 in cig kofte. In experiment 1, a cig kofte batch was inoculated with high levels of E. coli O157: H7 or L. monocytogenes (7.00 log(10) cfu/g) and the batches were divided into 6 groups. Each group was treated with 18 mL/kg of normal saline, cineole, limonene, carvone, linalool, or eugenol. Treatment with carvone, linalool, and eugenol resulted in a significant (P < 0.05) decrease in the numbers of both pathogens. In experiment 2, the cig kofte was inoculated with the same pathogens and each group was treated with 0 (control), 20, 22.5, and 25 mL/kg of eugenol. Increases in concentration did not cause a difference in pathogen reduction. The results showed that eugenol, carvone, and linalool can be used for the inactivation of E. coli O157: H7 and L. monocytogenes in cig kofte or similar foods.
