Yiğit, Uğur; Türkkan, Muharrem; İlhan, Hasan; Şimşek, Tuncay; Güler, Ömer; Derviş, Sibel

This study aimed to evaluate the antifungal activity of copper-boron (Cu-B) nanoalloys against a range of Phytophthora species, including P. capsici, P. citrophthora, P. palmivora, P. cinnamomi, P. nicotianae, P. cactorum, P. plurivora, P. inundata, and P. megasperma. The nanoalloys were synthesized via mechanical alloying under an argon atmosphere, resulting in the formation of nanocrystalline Cu-B nanoalloys with irregular morphology and particle sizes ranging from 50 to 240 nm. At a concentration of 250 µg mL−1, the Cu-B nanoalloys demonstrated complete inhibition of mycelial growth, sporangium production, and zoospore germination in all tested Phytophthora species. The EC50 values for mycelial growth ranged from 28.02 to 120.17 µg mL−1, while for sporangium production and zoospore germination, they were below 10 µg mL−1. Furthermore, the nanoalloys exhibited fungicidal activity against specific Phytophthora species, such as P. capsici, P. citrophthora, P. inundata, and P. megasperma, at concentrations of 100, 250, 250, and 250 µg mL−1, respectively. Notably, the Cu-B nanoalloys displayed significant protective and curative effects on tuber rot severity in P. nicotianae-inoculated potatoes, resulting in reductions of 94.13% and 92.61% compared to the control, respectively, at a concentration of 10 µg mL−1 (P < 0.05). These findings highlight the potential of Cu-B nanoalloys as a promising fungicide for the management of plant diseases caused by Phytophthora spp.

Characteristics of Hozat apricot seed oil as a new source of oilseed

    Aydin, Çağlar Mert

There has been an increase in apricot consumption due to its high bioactive and fiber content. However, the seed, inside part of the fruit, is considered as a food waste. In this study, potential usage of apricot seeds (bitter and sweet), that were cultivated in Hozat region- Tunceli, as an alternative oil source was evaluated. No comprehensive research has been undertaken to assess the seeds as an oil source before. In assessing its suitability for consumption, proximate composition of seed and physicochemical properties of the seeds’ oil had been determined. The proximate composition included the content of protein, ash, moisture, oil and the acidity as pH, in addition; iodine value, refractive index, nutritional quality, total phenolic content, antioxidant content, fatty acid composition and rheological properties of the oil were determined. Also, the effect of variety on them was examined. Overall, oil content was found to be the major component of the seed, thus the seed is good enough to be used as an oil source. Five different fatty acids were identified in the oil, that produced from the seed. These were palmitic, heptadecanoic, oleic, linoleic and linolenic acid. The oil had a refractive index between 1.4617 and 1.4628, iodine value between 100.47 and 102.11 gms, total phenolic content between 0.83 and 1.29 mg GAE g−1 oil, antioxidant activity between 11.21 and 11.60 mg TE g−1 oil for DPPH assay and 7.92–8.20 mg TE g−1 oil for ABTS assay, and apparent viscosity of 0.04 Pa.s. Moreover, viscous flow tests showed that the oil has a Newtonian flow behavior, and the nutritional indexes, that include unsaturated, atherogenicity, thrombogenicity and hypocholesterolemic/ hypercholesterolemic ratio, indicated that the oil is good enough for consumption as an oil source. The variety was found to have a significant effect on total phenolic content, antioxidant activity and refractive index of the oil, that was produced from apricot seed.

Effects of mutation in myostatin exon 3 on growth and carcass characteristics of Savak Akkaraman sheep

Kar, Bulent; Bahar, O.Z.; Yazdic, Ferit Can; Karaman, Altug

The myostatin gene (MSTN) is one of the main regulatory genes involved in muscle growth and development in many animals. This study aims to determine the variations in the exon 3 region associated with MSTN expression in Savak Akkaraman sheep and analyze their relationship with growth characteristics. Molecular analyses (RLFP, Real-Time PCR and DNA sequencing) were performed using DNA and RNA isolations from 70 different sheep blood and meat samples. As a result of the study, exon 3 region analysis was performed using NgomIV, AluI, DraI, HaeIII and MspI restriction endonucleases to determine the genetic diversity in Savak Akkaraman sheep breed. Two genotypes (MM and Mm) were detected for MSTN/MspI and a monomorphic mm genotype indicating the conserved exon 3 region for MSTN/HaeIII. Additionally, four new Single Nucleotide Polymorphisms (SNPs) were detected at different positions (164th, 204th, 347th, and 441th) in the working exon region. Two of these SNPs (204th T→G: isoleucine→ methionine and 347th C→A-G: proline→glutamine-arginine) caused amino acid changes. In expression studies, it was determined that MSTN gene expression was higher in female sheep than in male sheep (p<0.05). The MM genotype (MSTN/MspI) was significantly associated with increased weight gain and MSTN expression, while TT genotype at SNP 204 and GG genotype at SNP 347 was found to be associated with meat yield and growth traits (p<0.05). As a result, the genetic differences and functional effects of the new genotypes determined in this study may shed light on breeding strategies aiming to increase meat production.

Effect of ecosystems and agricultural practices on Physalis peruviana phytochemicals

    Aydin, Sevinc

As a result of the gradual decrease in natural resources and the increase in chronic diseases caused by unhealthy nutrition, the inclination among consumers toward food products that are both biologically active and beneficial, and environmentally friendly, is increasing. Therefore, environmental factors and agricultural practices that affect the amount of biologically active ingredients in these products have gained importance and become the subject of numerous scientific studies. Goldenberry (Physalis peruviana) is a yearly plant that is grown in mild climates, and it is a plant that is first planted in seed trays for primary development and then in the field for fruit production. Both its leaves, trunk, and roots have medicinal properties and its fruit is rich in components such as vitamins A and C, iron and phosphorus, flavonoid, and carotenoid which have made this plant, and particularly its fruit, more attractive. The amount and biological effect of these components, which are known as secondary metabolites, vary depending on the soil and the climate where this plant is grown and agricultural practices. With this compilation study, our aim is to summarize the studies that examined the positive or negative effects of environmental conditions that affect goldenberry cultivation and agricultural practices applied to increase the yield of the bioactive phytochemical ingredients of this plant, and thus to provide insight for relevant studies to be conducted.

An innovative approach in oat milk production: Ohmic heating

Ak, Ferit; Karakavuk, Emrah; Goksu, Ali; Sabancı, Serdal

This study aimed to produce oat-milk by using the ohmic heating (OH) system and to compare some quality properties of this milk with the milk produced by conventional methods. The solution containing oats was heated from 20 °C to 75 °C using 3 different voltage-gradients, and then oat-milk was obtained by applying a series of processes. It has been determined that as the voltage gradient increased during heating in oat milk production, the processing time shortens, and this time was 482 seconds in the conventional heating process. The effective electrical conductivity values increased as the temperature increased in the OH process during the production of oat-milk and these values vary between 0.13 and 0.41 S/m. Also, some quality characteristics of oat milk such as color and total phenolic content were better preserved in milk obtained by the OH process than conventional heating process. In addition, oat milk obtained by OH process had higher apparent viscosity at all temperatures.

A new hybrid approach for grapevine leaves recognition based on ESRGAN data augmentation and GASVM feature selection

Doğan, Gürkan; Imak, Andaç; Ergen, Burhan; Sengur, Abdulkadir

Grapevine leaf is a commodity that is collected only once a year and has a high return on investment due to its export. However, only certain types of grapevine leaves are consumed. Therefore, it is extremely important to distinguish the types of grapevine leaves. In particular, performing this process automatically on industrial machines will reduce human errors, workload, and thus cost. In this study, a new hybrid approach based on a convolutional neural network is proposed that can automatically distinguish the types of grapevine leaves. In the proposed approach, firstly, the overfitting of network models is prevented by applying data augmentation techniques. Second, new synthetic images were created with the ESRGAN technique to obtain detailed texture information. Third, the top blocks of the MobileNetV2 and VGG19 CNN models were replaced with the newly designed top block, effectively extracting features with the data. Fourthly, the GASVM algorithm was adapted and used to create a subset of the features to eliminate the ineffective and unimportant ones from the obtained features. Finally, SVM classification was performed with the feature subset consisting of 314 features, and approximately 2% higher accuracy and MCC score were obtained compared to the approaches in the literature.

Molecular characterization of apple endophytic bacteria and biological effect on Alternaria rot on apple

Bektas, Idris; Yazdıc, Ferit Can; Kusek, Mustafa

Apple rot disease, caused by Alternaria alternata, is causing significant losses in apple production, especially in post-harvest periods. Alternaria alternata, was isolated from rotten apple fruit of cv Amasya. Identification of the strain isolated from rotten apple fruit was made by ITS sequence and morphological analysis. In the present study, 58 different endophytic bacteria strains were isolated from apple fruit for use in apple rot disease control. Among the isolated strains, 26 endophyte strains were determined to inhibit the radial growth of mycelia of A. alternata in binary culture in PDA media. Based on 16S rRNA sequencing analysis, these antagonistic endophytic bacteria strains were identified as Bacillus ssp. Bacillus methylotrophicus (ib17) showed the highest significant inhibitory effect on the mycelial growth of A. alternata at a rate of 79.5% in PDA medium. The results of in vivo experiments showed that Bacillus licheniformis (ib20) achieved 100% biocontrol when endophyte bacterial strains were inoculated 24 h before the inoculation of A. alternata. When the three bacteria species and A. alternata were inoculated simultaneously, Bacillus myloliquefaciens (ib1), Bacillus licheniformis (ib21) and the Endophytic bacterium (ib16) showed the highest biocontrol efficacy against A. alternata at 81.8%. Furthermore, when the bacterial strains were inoculated at 24 h after inoculation of A. alternata, B. licheniformis (ib21) showed the highest biocontrol efficacy of 83.3% against the fungal pathogen compared to the controls. The endophytic bacterial strains produced fungal cell wall hydrolysing enzymes such as chitinase, cellulase, amylase and protease. According to the results of this study, apple endophytic bacteria are potential candidates to control apple decay caused by A. alternata through cell wall hydrolysing enzymes and reduce disease severity. However, the efficacy of the biological control was dependent on the applied time of epiphytic bacterial strains. Further research is needed on the biochemical mechanisms underlying the activity of these bacteria against A. alternata.

Evaluation of antioxidant, antimicrobial, and bioactive properties and peptide sequence composition of Malatya apricot kernels

Aydın, Çağlar Mert; Çelikbıçak, Ömür; Hayaloğlu, Ali Adnan

BACKGROUND: This study used four different apricot (Prunus armeniaca) kernels cultivated in Malatya during two consecutive years. The varieties were Hacihaliloglu, Hasanbey, Kabaasi, and Zerdali. The physicochemical properties of the kernels were determined, and the bioactive content of the kernels was evaluated using kernel hydrolysates prepared using trypsin. RESULTS: With regard to the physicochemical properties of the kernels, the dry matter ratio and protein content were the highest in the Hacihaliloglu variety; the ash ratio was the highest in the Kabaasi variety, and the free oil ratio was the highest in the Hasanbey variety. The bioactive compound content changed according to kernel variety. Angiotensin-converting enzyme inhibitors activity was found to be the highest in the Hacihaliloglu and Hasanbey varieties, which had the lowest amygdalin content, and Zerdali had the highest amygdalin content. The antioxidant and antimicrobial effects of the kernels varied, with Hasanbey and Kabaasi generally having the highest content in both analyses. Moreover, a concentration of 20 mg mL−1 of the hydrolysate was determined to have a destructive effect for the microorganisms used in this study. The storage protein of the kernels, except Hacihaliloglu, was found to be Prunin 1, with the longest matching protein chain in the kernels being R.QQQGGQLMANGLEETFCSLRLK.E. CONCLUSION: The results suggest that the peptide sequences identified in the kernels could have antihypertensive, antioxidative, and Dipeptidyl peptidase IV (DPP-IV) inhibitory effects. Consequently, apricot kernels show potential for use in the production of functional food products. Of the kernels evaluated in this study, Hacihaliloglu and Hasanbey were deemed the most suitable varieties due to their higher bioactive content and lower amygdalin content.

Effect of pretreatment temperature and time on fat bonding and fat reduction in deep-fried surimi

Erol, Nida Demirtas; Cakli, Sukran; Szymczak, Mariusz; Shen, Cangliang; Matak, Kristen E.; Jaczynski, Jacek

Deep frying (DF) is a popular cooking technique resulting in desirable sensory quality. However, DF also results in negative perception related to increased fat uptake and thus poor nutritional quality. In this study, a development of a novel strategy for fat reduction in DF surimi without additives and coatings was attempted. Novel pre-baking and pre-roasting were applied to surimi prior to DF. This novel pre-treatment strategy for DF surimi has not been reported in literature. Total, free, and bonded fat contents and corresponding fat reductions in DF surimi were determined. Cook and moisture losses as well as water holding capacity, expressible moisture, in addition to color and texture were also investigated. Fat uptake during DF of surimi was reduced (P < 0.05) by ≥ 25 % with pre-baking. Pre-baking improved (P < 0.05) or did not (P > 0.05) affect cook and moisture losses, water holding capacity, expressible moisture, color, and texture. However, pre-roasting increased (P < 0.05) fat content and adversely affected most quality parameters. Pre-baking of surimi at lower-temperature (116 °C) for longer-time (12–15 min) followed by DF at lower-temperature (170 °C) for longer-time (4 min) was most optimal for fat reduction, cook and moisture losses, water holding capacity, expressible moisture, color, and texture of surimi.

Changes in quality characteristics and inactivation of Salmonella in cake, including oleogel used as a fat replacer, baked with two different methods

Ozdemir-Orhan, Necla; Eroglu, Zeynep; Omac, Basri

This is the first study to assess the impact of substitution of shortening (50%) with sunflower-beeswax oleogel in cake formulations on the inactivation kinetics of Salmonella spp. and the quality attributes of cakes baked in conventional (CO) and microwave (MWO) ovens. Four distinct cake samples were examined in the study: Cake samples containing oleogel prepared in two different ovens (Oleo-Cake-CO and Oleo-Cake-MWO) and control cake samples baked in two different ovens (Cont-Cake-CO and Cont-Cake-MWO). The control-batter and oleogel-batter demonstrated shear thinning behavior (pseudoplastic, n < 1), with a good fit to the Power Law model, but viscosity and viscoelastic moduli decreased when the oleogel was used in place of shortening in cake recipes. In addition, the Cont-Cake-MWO had the greatest special volume value (2.31 ± 0.04 mL/g), whereas the Oleo-Cake-CO had the lowest (1.65 ± 0.02 mL/g) (p < 0.05). Furthermore, compared to the samples baked with CO, lower water activity and moisture values (p < 0.05) were observed in the samples baked in MWO due to their higher cooking loss values (p < 0.05). In all baking techniques, the addition of oleogel to the cake formula, used as a fat substitute, resulted in higher values for cohesiveness, hardness, springiness, and chewiness (p < 0.05). As a result, the inactivation of Salmonella in cakes slightly reduced with using oleogel as a fat substitute (p > 0.05), whereas it affected some quality properties of cakes baked with both heat treatments.

Development of new generation cakes fortified with zinc oxide nanoparticles

Kaplan Ince, Olcay; Aydogdu, Burcu; Ince, Muharrem

This study aimed to develop new generation cakes that were fortified with calcined ZnO nanoparticles (CZnO), uncalcined ZnO nanoparticles (UCZnO), beads (B(CZnO)) synthesized by encapsulating the CZnO with sodium alginate (SA), and the beads (B(UCZnO)) synthesized by encapsulating the UCZnO with sodium alginate (SA) and investigated the zinc (Zn) release in fortified cakes in simulated body fluids (SBF). The present study represents a novel method for increasing intestinal absorption and bioavailability of dietary zinc with zinc nanoparticles for use in the preparation of Zn fortified cakes as a dietary supplement to compensate for zinc deficiency in humans. The results revealed that a rapid increase in the release time and rate in the SGF solution was noted in the UCZnO added cakes. It was attributed to increased intestinal absorption and bioavailability as a result of the ultra-small size of ZnO. Also, ZnO release kinetics in SBF was also studied and data were adjusted into different kinetic models involving zero-order, first-order, Higuchi, and Korsmeyer–Peppas models. The present investigation recommends adding UCZnO to the cakes to control and increase the release from the cakes.

Effect of type and concentration of antioxidant on sperm motility, viability, and DNA integrity of climbing perch Anabas testudineus Bloch, 1792 (Pisces: Anabantidae) post-cryopreservation

Maulida, Siti; Eriani, Kartini; Fadli, Nur; Siti-Azizah, Mohd N.; Kocabas, Filiz K.; Kocabas, Mehmet; Wilkes, Martin; Handayani, Luvi S.; Rahayu, Sri R.; Muchlisin, Zainal A.

Sperm quality is preserved through the crucial involvement of antioxidants, which play a vital role in minimizing the occurrence of reactive oxygen species (ROS) during the cryopreservation process. The suitability of the type and concentration of antioxidants are species-dependent, and this study is crucial in order to improve the quality of the climbing perch sperm post-cryopreservation. Therefore, this study aimed to determine the best type and concentration of antioxidants for cryopreservation of climbing perch Anabas testudineus sperm. To achieve this, 6 types of antioxidants, namely, ascorbic acid, beta-carotene, glutathione, butylated hydroxytoluene (BHT), myo-inositol, and alpha-tocopherol, with inclusion of a control were tested in 3 replications at three concentration levels of 0 mg/L (control), 20 mg/L, 40 mg/L, and 60 mg/L. Sperm was diluted in a glucose-base extender at a ratio of 1:60 (sperm: glucose base), then 10 % DMSO and 5 % egg yolk was added before cryopreservation for two weeks. The results showed that the type and concentration of antioxidants had a significant effect on the motility and viability of cryopreserved climbing perch sperm (P < 0.05), where the best results for ascorbic acid, beta-carotene, glutathione, myo-inositol, and alpha-tocopherol were obtained at a concentration of 60 mg/L, while BHT was at a concentration of 20 mg/L. The best results for glutathione, myo-inositol, and alpha-tocopherol were significantly different from other treatments, while the best results for ascorbic acid and beta-carotene (60 mg/L) were not significantly different from the 40 mg/L concentration, while the best results for BHT were not significantly different from the control treatments. Therefore, the best concentration of glutathione, myo-inositol, and alpha-tocopherol was 60 mg/L, while for ascorbic acid and beta-carotene it was 40 mg/L, and BHT was not recommended. DNA integrity analysis indicated the absence of fragmentation in all samples, including fresh, control, and treated sperm. Based on practical and economic considerations, myo-inositol at 60 mg/L was recommended for cryopreservation of climbing perch A. testudineus sperm.

Handayani, Luvi S.; Maulida, Siti; Rahayu, Sri R.; Razi, Nanda M.; Kocabas, Mehmet; Kocabas, Filiz K.; Wilkes, Martin; Siti-Azizah, Mohd N.; Eriani, Kartini; Fadli, Nur

BACKGROUND: Walking catfish, Clarias batrachus is one of the native and most popular freshwater catfish species in Indonesia. However, cultivation faces challenges, particularly due to the scarcity of larvae resulting from underdeveloped breeding technologies. Cryopreservation is a method of storing sperm to maintain viability for a long period and support the breeding technology of the fish. Cryoprotectant, in this context, plays an important role in determining the success of sperm cryopreservation. OBJECTIVE: To determine the best type and concentration of cryoprotectant for cryopreservation of walking catfish sperm. MATERIALS AND METHODS: A total of five different types of cryoprotectants, namely DMSO, glycerol, ethyl glycol, ethanol, and methanol, were tested at four concentration levels namely 0%, 5%, 10%, 15%, and 20%, each with four replications. RESULTS: The type and concentration of cryoprotectant had a significant effect on sperm motility and viability (P<0.05). The best outcomes were obtained with 5% DMSO and ethyl glycol, 10% glycerol and methanol, as well as 15% ethanol. CONCLUSION: The highest motility and viability values were obtained with 5% DMSO, resulting in its recommendation for cryopreservation of walking catfish sperm.

Performance of artificial neural networks and traditional methods in determining selected growth parameters of Alburnus sellal Heckel, 1843

    Ozcan, Ebru Ifakat

In this study, predictions were made on the growth performance of Alburnus sellal Heckel, 1843 from the Munzur River using back propagation artificial neural networks and ANN algorithms. Statistical growth models used in fish biology and results obtained from artificial neural networks were compared. A total of 239 samples were used in this comparison. It was determined that the population is distributed across age groups 0–VII. The relationship between length and weight was calculated as W = 0.0046L3.198 for all individuals. The von Bertalanffy growth parameters were calculated for all individuals: Lt = 21.93 [1 – e–0.158 (t + 2.11)]; Wt = 102.29 [1 – e–0.158 (t + 2.11)]3.198. The growth performance index (Ф’) value was 1.880 for all individuals. The condition factor varied between 0.479 and 1.115 for females and between 0.533 and 1.076 for males. The Mean Absolute Percent Error (MAPE) statistic was used, which is a widely used method to measure the accuracy of the predictions made. It was determined that ANNs MAPE (%) values were better than MAPE values calculated for the length–weight relationship and von Bertalanffy growth function models for A. sellal. This study shows that ANNs can be used as an alternative useful method for predicting population parameters. ANN models are therefore an effective tool to describe fish growth parameters. They have been found to be a useful predictive tool. The developed models can be used to predict future sustainable fish management.

Impact of the arginine silicate inositol complex on bone metabolism in broiler chickens with tibial dyschondroplasia caused by manganese deficiency

    Sahin, E.; Ipcak, H.H.; Orhan, C.; Denli, M.; Erten, F.; Ozercan, I.H.; Balci, T.A.; Sahin, K.

    1. Tibial dyschondroplasia (TD) is a skeletal disorder in broilers that has financial implications, necessitating dietary modifications to reduce the prevalence of this disease. This study explored how arginine silicate inositol complex (ASI) supplementation affected tibial growth plate (TGP) and overall bone health in broilers with manganese (Mn) deficiency-induced TD. 2. A total of 240 broiler chicks were divided into four groups, each consisting of 60 birds (15 replicates of four broilers each) as follows: i) Control, with 60 mg Mn per kg of diet; ii) ASI, with 60 mg Mn and 1 g ASI per kg of diet; iii) TD, with 22 mg Mn per kg of diet, and iv) TD+ASI, with 22 mg Mn and 1 g ASI per kg of diet. 3. It was found that ASI supplementation increased tibial bone length in Mn-deficient TD broilers (p = 0.007). There was no Mn x ASI interaction for other bone morphometry variables (p > 0.05). However, both tibial bone mineral content and density were affected by Mn and ASI (p < 0.05). With ASI supplementation, serum bone-specific alkaline phosphatase and osteocalcin levels were elevated in the TD+ASI group compared to the TD group (p < 0.001). In the TD group, osteoprotegerin (OPG) levels in the TGP decreased compared to the control groups (p < 0.001). 4. In contrast, ASI supplementation in the TD broilers counteracted the decrease in OPG compared to TD broilers without ASI supplementation (p < 0.001). The Mn level and ASI supplementation significantly influenced the OPG/receptor activator of the nuclear factor-κB ligand ratio (p < 0.001). 5. In conclusion, the results demonstrated that inclusion of ASI in broiler diets could enhance bone formation variables by controlling OPG levels in the TGP, potentially serving as an effective method to decrease the occurrence of TD.
